Effect of alternate high carbohydrate and high fat diets on glucose utilization in vivo and in vitro

Experimental diets of high carbohydrate content or alternating high carbohydrate and high fat content were fed to rats for 2-day periods over 20 days in an attempt to explain why the latter diet pattern seems to prevent impaired glucose tolerance in response to fat ingestion. At the end of the diet period, rats were force fed corn oil 4 times at 6 hour intervals and fasted overnight. Analyses were then done on liver glycogen accumulation, the in vivo incorporation of radioactivity of oral U-14 C-glucose and the in vitro utilization of U-14 C-glucose by liver and adipose tissue. The alternating diet produced a greater accumulation of liver glycogen and absorption of radioactivity, as well as higher utilization of U-14 C-glucose by liver and adipose tissue than the high carbohydrate diet, which may explain why the alternating diets prevent impaired glucose tolerance.
