Restriction fragment length polymorphism in a subclass of the 'Mandarin' soybean cytoplasm

Mitochondrial DNA (mtDNA) from a number of soybean, Glycine max (L.) Merr., cultivars was examined by restriction endonuclease digestion to search for possible differences among cytoplasmic backgrounds. In these analyses it was observed that mtDNAs from certain members of the 'Mandarin' cytoplasm, represented by 19 cultivars maternally derived from 'Lincoln', were different from 38 cultivars derived from other cytoplasms. A 10.5-kb (Kilobase) HindIII fragment that was absent from the Lincoln class of mtDNA was present in the mtDNA of the other cytoplasms tested. Use of the 10.5-kb fragment as a hybridization probe in Southern analyses revealed the presence of a smaller 2.3-kb HindIII fragment that was unique to the Lincoln-derived mtDNAs. The 2.3-kb HindIII fragment was cloned and used in Southern hybridizations of mtDNA to screen for differences among a large number of soybean cultivars. A portion of the 2.3-kb HindIII fragment was found at a second location common to the mitochondrial genome of all cultivars tested.
