Problems still persist in determining the magnitude and understanding the nature of adult protein requirements. The various methods used to determine nutrient requirements have shortcomings, as is reviewed in the article. It is contended that new methodological approaches must involve initial development of a biologically appropriate model for the nature of the requirements within which experimental results can be evaluated. Examining nitrogen balance in the adult, an important factor in protein requirement assessments, is addressed from the standpoint of different model approaches. Cyclic changes in body protein over periods of days on a fixed intake, and cyclic autocorrelative changes of body nitrogen over longer time periods are two models discussed. It is concluded that identification and investigation of the behavioral characteristics of the body protein and amino acid pool must be a primary objective of research on protein requirements.(jd-b)
