Maintenance of somatic embryos and cell lines from floral parts of Punica granatum L. 'Ganesh'

An efficient protocol for the maintenance of embryogenic callus and cell lines on ¾ strength Murashige and Skoog liquid media with and without filter paper boat was standardized. Floral explants such as filaments and segments of petals were isolated from unopened flower buds of Punica granatum ‘Ganesh’. The individual and combined effects of growth regulators such as Thidiazuron (TDZ), indol-3-acetic acid (IAA) and Picloram at 0.5, 1.0 and 2.0 mg/L concentrations were studied. Light brownish embryogenic calli suspension was established within 60 days in MS media supplemented with Picloram. However, precocious germination of somatic embryos was more prominent in presence of indol-3-acetic acid (IAA). On the contrary, media supplemented with TDZ favored the growth of greenish, highly nodulated mass of embryogenic calli. Greenish globular to bipolar stage embryos were more prominent even after 540 days of growth period on media supplemented with TDZ supported by filter paper boat, whereas light yellowish mass of friable callus was induced on media containing IAA. The present study is more efficient for the maintenance of bipolar somatic embryos and embryonic cell lines in absence of rotary shaker without any browning of the media and frequent subculture in pomegranate.
