Supplementation with resveratrol during culture improves the quality of in vitro produced bovine embryos

Oxidative stress is one of the major factors affecting mammalian in vitro embryo development. The aim of this study was to evaluate whether the supplementation of culture media with the antioxidant Resveratrol improves in vitro bovine blastocyst development and quality, assessed by the blastocyst rate and total cell number respectively. Abattoir-derived oocytes were matured and fertilized in vitro according to a standard procedure. Twenty one hours after in vitro fertilization, presumptive zygotes were cultured in media supplemented with 0, 0.2, 0.5, 1.0, 5.0 or 10 µM Resveratrol. On day 7, blastocyst rates were recorded and embryos were stained with Hoechst (5 µg/ml) for total cell number evaluation. Results revealed that the use of 0.2 µM Resveratrol during in vitro culture significantly increased (P<0.05) blastocyst rate in comparison to the control (54.84 and 40.69 % respectively), but was similar to 0.5, 1.0 and 5.0 µM Resveratrol (48.68, 49.93 and 46.78% respectively), while high concentrations of Resveratrol (10 µM) significantly reduced (P<0.05) embryo development respect to all the evaluated concentrations (27.91%). Total cell number was significantly increased (P<0.05) by 0.5 µM Resveratrol respect to 0, 5.0 and 10 µM treatments, (125.1, 107.4, 106.4 and 102.9 respectively) but was similar to 0.2 and 1.0 µM Resveratrol (115.8 and 113.7 respectively). In conclusion, the supplementation of bovine embryo culture media with low Resveratrol concentrations (0.2 to 1 µM) increase blastocyst rate and improve embryo quality in terms of total cell number.
