Genetic diversity studies in some mango genotypes using DNA markers

The mango (Mangifera indica L.) is one of the most important tropical fruits of the world. There is a large varietal wealth created because of high heterozygosity and propagation by seeds in this crop. The present day cultivars are mostly based on the selection of clones/chance seedlings. Due to this large varietal wealth among Mangifera indica cultivars, nomenclature ambiguity is one of the main problems hindering improvement. This has also created a problem in the choice of parents for breeding programmes, as genetically distant parents would ensure better recombinants. In the present investigation, genetic variability among 11 mango genotypes was studied using inter-simple sequence repeat primers. Out of the primers tested, di-nucleotide and tri-nucleotide repeats gave clear and reproducible band profiles. (GA)n, (CT)n, (CA)n, (TC)n, (AC)n, (TG)n, rich primers gave good amplification profiles. A sum of 77 amplicons was generated that resulted in 93.5% polymorphism in the samples used. Clustering of the genotypes based on UPGMA method generated three groups. Highest similarity was observed among ‘Dwarf rumani’ and ‘Creeping’. ‘Moreh’ and ‘Sabre’ both being polyembryony types grouped together in a cluster 2. The high polymorphism and reliable amplification confirmed the utility of DNA marker for genetic diversity studies in mango.
