Evaluation of variations in amylose-iodine absorbance spectra

Use of variation in the spectra of amylose-iodine complexes to identify and characterize starches and amyloses is reported. Variations in absorption, wavelengths of max. absorbance and slopes of the absorbance peaks were determined for pure amyloses, corn starches (high amylose, normal and waxy corn starches) and starches from barley, oats, native potato, lintnerized potato, rye, sorghum and wheat. Variations in these variables, as a result of differences in amylose mol. wt., were small but quantitatively reproducible. This allowed differentiation of: amyloses of different mol. wt.; high amylose and normal corn starches; starches from different botanical sources; and raw and modified starches. It is concluded that measurement of the complete visible spectrum of the amylose-iodine complex allows detection of major differences between starches and positive identification of starches with unusual composition.
