Optimization of pectinase production by Aspergillus Niger Using Citrus Rinds

During present investigation pectin was isolated from orange and sweet lemon peels Potato dextrose agar (PDA) was enriched with citrus pectin and Aspergillus niger was grown on it. Effect of various physicochemical parameters, such as temperature (°C), pH, metal ions (%), incubation period (days) and nitrogen source (%) on the production of enzyme pectinase was studied. The enzyme production gradually increased till 3rd day of incubation, showing maximum activity on 4th day, after which it gradually decreased. The most suitable nitrogen source for maximum enzyme production was ammonium sulphate, while supplementation the medium with MgS04 also enhanced pectinase production. Maximum pectinase activity was observed at pH 5, whereas 35°C was the optimum temperature for maximum pectinase activity.
