Distribution of Sugars in the Soybean Embryo

In order to investigate the possibility of using single seed selection to change sugar content in soybean (Glycine max (L.) Merr.) seed, the distribution of sugars in the soybean embryo was studied. Seeds of soybean cultivars ‘Jogun’ and ‘Hokkaido’ were sliced into three equal parts with the slices being made parallel to the root: shoot axis. The slices were then analyzed for sugar content by gas-liquid chromatography of trimethylsilyl derivatives. Sucrose content was highest in Position I, represented by the slice containing the root:shoot axis, and lowest in Position III. Stachyose was lowest in Position II and approximately equal in Positions I and III. Raffinose content was invariant for all positions. A significant cultivar ✕ position interaction for sucrose content indicates that selection for differences in this character based on analyses of only parts of a cotyledon might be unreliable.
