Spilanthes acmella: A Natural Weed Reservoir of Sugarcane Leaf Yellows Phytoplasma Strain (16SrI-B) in Eastern Uttar Pradesh, India

During survey of sugarcane fields at Gorakhpur, Kushinagar and Faizabad districts of eastern Uttar Pradesh, phytoplasma suspected little leaf and leaf yellowing symptoms were recorded on Spilanthes acmella weed in 2019–2020. Since this weed species is not listed yet as phytoplasma host, the present study was investigated with characterization of phytoplasma strain associated with this weed. The expected amplicons of ~ 1.25 kb and ~ 480 kb were achieved in symptomatic Spilanthes leaf samples from all the three locations with the phytoplasma specific universal 16S rRNA and secA gene specific primers, respectively. Sequence comparison analysis of 16S rRNA and secA genes confirmed the association of ‘Ca. P. asteris’-related strain with symptomatic Spilanthes leaf samples. However, comparing the restriction profiles of 16S rRNA gene in virtual RFLP analysis revealed that the Spilanthes strain associated was related to phytoplasma subgroup, 16SrI-B. This is the first report of 16SrI-B group phytoplasmas association in S. acmella worldwide, which may be a natural potential host of sugarcane leaf yellows phytoplasma strain in Eastern Uttar Pradesh, India.
