The Relationships between Nucleic Acids, Proteins and Flower Bud Formation in Loquat

The variations of nucleic acids and proteins in the leaves or buds during flower bud formation in potted and field-grown ‘Zaozhong’ trees were studied. There was a close relationship between nucleic acids and flower bud differentiation, which requires the participation of abundant nucleic acid. The total nucleic acid, DNA and RNA contents ascended gradually in the physiological differentiation period. RNA content showed two peaks during inflorescence primordium differentiation and stamen and pistil primordium differentiation, the second peak being higher than the first one. DNA content displayed a single peak, which occurred between the two peaks of RNA content. The peaks of RNA/DNA ratio appeared at the same time as the RNA content. In floral buds, the total nucleic acid, RNA, DNA contentsd, and RNA/DNA ratio were all higher than in the vegetative buds. During flower induction, soluble protein content in the leaves on floral shoots rose, while during flower differentiation, it fell sharply. Flower induction and differentiation were two distinct phases related with each other. The two-dimensional electrophoresis diagrams of proteins extracted from floral buds and vegetative buds were on the whole similar to each other. However, in the diagram from floral buds there were two proteins with an iso-electric point (pI) 5.350±0.008 and 4.730±0.032, and a molecular weight (MW) of 14.110±111 and 66.446±261, respectively, which were not present in vegetative buds. The two acidic soluble new proteins might be closely related with flowering in loquat.
