Saturated, n-6, or n-3 fatty acids and cholesterol supplementation: differential effects on liver and heart lipid composition

We have investigated the effect of a high saturated-fat diet (butter fat), a high n-6 polyunsaturated-fat diet (corn oil), and a high n-3 polyunsaturated-fat diet (menhaden fish oil) with or without additional cholesterol on the membrane lipid composition of rat heart and liver. Liver plasma membranes and heart crude membranes from animals fed the corn oil diet demonstrated increased total n-6 fatty acids, while tissues from animals fed the menhaden fish oil diet had increased total n-3 fatty acids. This effect was independent of cholesterol addition to the diet. However, in the liver plasma membrane, dietary cholesterol significantly increased C18:In-9 in corn oil and butter fat-fed animals. Liver plasma membranes of cholesterol-supplemented, corn oil- or butter fat-fed animals also demonstrated a decreased ratio of C20:4n-6/C18:2n-6. Heart membranes demonstrated no changes in C18:In-9, C18:2n-6, or C20:4n-6 due to cholesterol supplementation.
