Efficacy of A fish scale bacterial isolate FSD-B3 encapsulated in nontoxic biodegradable gel matrix for alkaline protease activity augmentation

The present investigation was carried out to check the efficacy of a bacterial strain, FSD-B3 for alkaline protease activity after the immobilization process. Owing to the nontoxic biodegaradable property of the Sodium alginate, it was used for entrapment purpose. Interestingly, FSD-B3 strain (isolated from fish scale) was identified as Bacillus sp., displaying hydrolysis index of 440% in skimmed milk agar plate. Following immobilization, the enzyme activity augmentation was observed in skimmed milk agar plate as well as in fermentation broth (supplemented with fish scale as a substrate). In agar plate nearly 40% rise in hydrolysis index was observed in immobilized cells as compared to free cells. The enzyme assay performed in fermentation media also complemented the results of plate assay by showing an increase of 26% alkaline protease activity after the entrapment process. Thus, sodium alginate served as a good matrix for immobilization and enhanced the enzyme activity as expected. Further studies on plant growth promoting activities of Bacillus FSD-B3 are in offing.
