Effects of the growth regulators for the induction of somatic embryos from different explants of Echinocactus parryi Engelm., an endemic and endangered species

Abstract Introduction: The list of threatened species is enhancing and needs to integrate plant tissue culture tools with conventional techniques that support the appropriate management of these species. Objective: To assess the effects of the growth regulators for the induction of somatic embryos from explants of Echinocactus parryi Engelm. Materials and methods: A completely randomized design was utilized to evaluate three types of explants (mature seeds, shots and green callus) cultured on basal Murashige & Skoog media (MS) with different growth regulators concentrations (2, 4-D [dichlorophenoxy acetic acid], BAP [6-benzylaminopurine] and kinetin). Histological analysis of the embryogenic structures was performed. Results and discussion: The auxin 2, 4-D induced both embryogenic and organogenic callus from seeds and shoot explants. The globular stage did not evolve to their maturity, presumably because of 2, 4-D accumulation. The compact callus explants were the more efficient to induce 19.2 somatic embryos per explant when they were cultured in the medium with 0.5 mg∙L-1 kinetin. However, the latest phases did not germinate, probably due to abnormalities generated by genetic and epigenetic changes in the DNA that can cause abnormal somatic embryos. The histology image demonstrated that the globular and torpedo structures were visible under a microscope showing stained nucleus and numerous starch grains. Conclusions: E. parryi is a species that can produce a high number of embryogenic structures, which represents a great potential to grow massive plants.

Resumen Introducción: La lista de especies amenazadas está incrementando y necesita la integración de herramientas de cultivo de tejidos vegetales con técnicas convencionales que apoyen el manejo adecuado de dichas especies. Objetivo: Evaluar los efectos de reguladores de crecimiento en la inducción de embriones somáticos a partir de explantes de Echinocactus parryi Engelm. Materiales y métodos: Se utilizó un diseño completamente aleatorio para evaluar tres tipos de explantes (semillas maduras, brotes y callo verde) cultivados en medio basal Murashige & Skoog con diferentes concentraciones de reguladores de crecimiento (2, 4-D [ácido diclorofenoxiacético], BAP [6-bencilaminopurina] y kinetina). Se realizó un análisis histológico de las estructuras embriogénicas. Resultados y discusión: La auxina 2, 4-D indujo callo embriogénico y organogénico a partir de explantes de semillas y de brotes. La fase globular no evolucionó hasta su madurez, debido presumiblemente a la acumulación de 2, 4-D. Los explantes de callo compacto fueron los más eficientes para inducir 19.2 embriones somáticos por explante cuando se cultivaron en medio con 0.5 mg∙L-1 de kinetina; sin embargo, las últimas fases no germinaron, probablemente a causa de anomalías generadas por cambios genéticos y epigenéticos en el ADN que pueden causar embriones somáticos anormales. La imagen histológica demostró que las estructuras globulares y de torpedo fueron visibles al microscopio mostrando el núcleo teñido y numerosos granos de almidón. Conclusiones: E. parryi es una especie que puede producir un número elevado de estructuras embriogénicas, lo que representa un gran potencial para el cultivo masivo de plantas.
