Banana cv. Kayu is an unique cultivar of banana Bali. It is becoming increasingly rare and is rarely seen for sale in the market. Conservation is needed to maintain Bali's local genetic resources. Ex-situ conservation efforts can be done through plant propagation through in vitro culture, which will produce a large number of plantlets that are genetically similar to their mother plant. The study used planting material in the form of meristemoids previously induced from banana stumps with the hormone cytokinin. The existence of various in vitro media for banana led this study to test the best media for banana cv. Kayu. The aim of the study was to test and analyze the effect of various types of vitro media for the growth of banana cv. Kayu meristemoids. The study used a Factorial Completely Randomized design with 2 factors and 5 replications. The first factor was the type of media, namely MS, BMM and AGS. The second factor was growth regulators, namely: ZO=without ZPT, Z1= BAP 5 ppm, Z2= TDZ 2 ppm, Z3= BAP 5 ppm + TDZ 2 ppm. The variables observed were shoot growth, number of shoots, number of leaves, number of roots and root length. The results showed that MS media treatment gave the highest results in the variable of shoot emergence, number of shoots, number of leaves, number of roots and root length compared to BMM and AGS media. The treatment growth regulator 2 pmm TDZ (Z2) gave the highest results on the number of leaves, number of roots and root length. The highest shoot emergence was obtained by 5 ppm, 2 ppm TDZ (Z1 and Z2) which was 1.86 cm.
