Toxoplasma gondii in pork production: a stable to table approach | Toxoplasma gondii dans la production porcine : une approche de l'étable à la table

English. Toxoplasma gondii is an intracellular coccidian parasite and one of the most successful parasites worldwide. Humans, as intermediate hosts, become infected with T. gondii through ingestion of oocysts (e.g. when handling soil or cat litter, or on unwashed vegetables) or tissue cysts in raw or undercooked meat. Meat appears to be a major source of T. gondii infections in Europe, as in a European multi‐center case control study 30 to 63% of infections in pregnant women were attributed to meat, whereas 6 to 17% were most likely soil borne (Cook et al., 2000). With 40.4 kg per household in 2014, pork is the first meat consumed in France, and three quarters is eaten in the form of sausage and salami products (FranceAgriMer, 2015). The contribution of pork meat consumption to human infections with T. gondii is not known, nor is the risk of transmission of the parasite via the consumption of cured meat products. As early as 1999, the Codex Alimentarius Commission recommended that microbiological risk assessment should be based on a quantitative estimate. The high seroprevalence of T. gondii infection in France in humans (32%) and the fact that the main mode of contamination is food-borne, fully justifies the conduct of a quantitative microbiological risk assessment (QMRA). In France, we have the T.gondii seroprevalence in various meat-producing species (ovine, bovine, pig, horses) enabling such an analysis. It lacks however the quantitative estimates concerning the distribution of parasites in the various parts (muscles) of a carcass and the reduction of the parasite load according to the cooking, preparation or preservation, as identified by an expert group of ANSES in 2005 (AFSSA, 2005). The present project aims at answering this lack of knowledge for pork products, providing the basis for a robust QMRA analysis in France, a European country with a high prevalence of Toxoplasma infection among humans. Following the scientific question, the present research project aims to respond, based on three main areas of study consisting of (i) a thorough investigation of T. gondii predilection sites in pigs experimentally infected with two different stages (tissue-cyst versus oocyst); (ii) evaluate the impact of the manufacturing process (including different concentrations of nitrites and NaCl) and the conservation of dry sausage on the viability of T. gondii; (iii) a quantitative microbiological risk assessment to predict the relative contribution of the various raw pork products (dry sausage, dry ham, etc) to human T. gondii infections in France.

French. La toxoplasmose est une zoonose parasitaire causée par un protozoaire intracellulaire, Toxoplasma gondii. Chez les immunodéprimés, on décrit des formes graves telles qu'encéphalite et choriorétinite. De plus, chez la femme enceinte, une primo-infection à T. gondii peut provoquer des malformations du fœtus, voire un avortement. L'infection humaine se fait principalement par voie orale, soit par ingestion d'oocystes rejetés par les chats (hôtes définitifs); soit par consommation de viande crue ou insuffisamment cuite contenant des kystes tissulaires (hôtes intermédiaires). La viande de porc est la viande la plus consommée en France (40,4 kg par ménage et par an) et un véhicule potentiel, puisque, les ¾ sont dégustés sous la forme de produits de charcuterie-salaison. Pourtant la possibilité d'infection par T. gondii via la consommation de produits crus de salaison est moins précise. Ceci s'explique d'une part par une méconnaissance du tropisme musculaire du parasite chez le porc et d'autre part par une absence de données scientifiques robustes permettant d'évaluer l'effet inhibiteur des étapes de fabrication et de conservation de ces produits. Ce projet de recherche se propose de répondre sur la base de trois axes d'étude consistant à (i) décrypter de manière approfondie l'organotropisme de T. gondii au sein de carcasses de porcs expérimentalement infestées avec deux modalités d'infestation différentes (kyste versus oocyste) ; (ii) évaluer l'impact du procédé de fabrication (incluant différents taux d'incorporation en nitrites et en NaCl) et de la conservation du saucisson sec et jambon sec sur la viabilité de T. gondii; (iii) une analyse quantitative du risque lié à la consommation humaine de produits de charcuterie
