E-mail: [email protected] Fasciolosis is a zoonotic disease caused by a parasite Fasciola hepatica that can infect different species of animals, especially ruminants. The aim of this study to evaluate the profile of the immune response in bovine livers naturally and chronically infected by F. hepatica, using immunohistochemical technic by lymphocytes T and B, as well IgG plasma cells. Samples were obtained from bovine livers macroscopically positive for flukes. We collected fragments of 4 cm2 of each lobe, right and left, of the 100 livers condemned for flukes. The samples were placed in 10% formalin, identified and targeted for general histopathological evaluation of morphological and intensity of inflammatory infiltration. We selected at random, 18 samples (six samples for each intensity of inflammation), which were submitted to immunohistochemical evaluation to T lymphocytes (CD3+), B lymphocytes (CD79α+) and IgG plasma cell (IgG+). The labeled cells by immunohistochemistry were counted in ten port space at random from each lobe. We calculated the mean number of cells per lobe. No significant difference was observed between the expression of T lymphocytes, B lymphocytes and IgG plasma cells in the right lobe (p=0.5817) and left lobe (p=0.9367). Mean T lymphocytes (CD3+) of the left lobe has been greater than the right lobe (p=0.019), as well as B lymphocytes (CD79α+) (p=0.00018) and IgG plasma cells (p=0. 0002). The CD3+ T lymphocytes in the left hepatic lobe significantly different (p=0.024) relative to the intensity of infiltration, showing the difference between mild to severe (p<0.05). CD79α+ B lymphocytes in the right hepatic lobe showed significant differences (p=0.008) in the different intensities of infiltration being observed between mild and severe (p<0.01) T and B lymphocytes and IgG plasma cells are present in immune response in livers of cattle with chronic fasciolosis, being more evident in the left hepatic lobe. The immunohistochemical assessment was effective in cell immunophenotype characterization of this inflammatory response caused by natural infection becoming important in the study of bovine fasciolosis.

Portuguese. A fasciolose é uma zoonose causada por Fasciola hepatica, um trematodeo digenético, que pode infectar diferentes espécies animais, especialmente ruminantes. Objetivou-se com este estudo avaliar o perfil da resposta imune frente à infecção por F. hepatica em fígados de bovinos cronicamente e naturalmente acometidos, por meio da imunofenotipagem dos linfócitos T e B e dos plasmócitos produtores de IgG. As amostras foram obtidas de fígados de bovinos macroscopicamente positivos para fasciolose. Foram coletados fragmentos de aproximadamente 4 cm2 de cada lobo, direito e esquerdo, de 100 fígados condenados por fasciolose. As amostras foram acondicionadas em formalina 10%, devidamente identificadas e direcionadas para avaliação histopatológica geral das alterações morfológicas e intensidades do infiltrado inflamatório, sendo selecionadas, aleatoriamente, 18 amostras (seis amostras para cada intensidade de inflamação), as quais foram submetidas a avaliação imunoistoquímica para linfócito T (CD3+ ), linfócito B (CD79α+ ) e plasmócito produtor de IgG (IgG+ ). As células marcadas pela imunoistoquímica foram contadas em dez espaços porta, aleatoriamente, de cada lobo hepático. Foram calculadas as médias do número de células por lobo hepático. Não foi observada diferença significativa entre a expressão de linfócitos T, linfócitos B e plasmócitos produtores de IgG no lobo direito (p=0,5817) e também lobo esquerdo (p=0,9367). A média de linfócitos T (CD3+ ) do lobo esquerdo foi maior que do lobo direito (p=0,019), assim como para linfócitos B (CD79α+ ) (p=0,00018) e para os plasmócitos produtores de IgG (p=0,0002). Os linfócitos T CD3+ no lobo hepático esquerdo diferiram significativamente entre si (p=0,024) em relação à intensidade de infiltração, mostrando diferença entre as intensidades leve e severa (p<0,05). Os linfócitos B CD79α+ no lobo hepático direito apresentaram diferença significativa (p=0,008) nas diversas intensidades de infiltração, sendo verificada entre leve e severa (p<0,01). Linfócitos T e B, e células plasmáticas produtoras de IgG estão presentes na resposta imune de fígados bovinos com fasciolose cronica, sendo mais evidentes no lobo hepático esquerdo. A avaliação imunoistoquímica mostrou-se eficaz na caracterização do imunofenótipo celular presente na resposta inflamatória causada pela infecção natural tornando-se importante no estudo da fasciolose bovina.
