Melhoramento genético da proteína da soja: eliminação de lipoxigenases e das subunidades RAPD-PCR na seleção de genótipos e na identificação de marcadores ligados ao gene Cgy1 | Soybean protein quality improvement: genetic elimination -PCR of lipoxygenases and the to select genotypes and to identify marker linked to the Cgy1 gene

Este trabalho foi realizado com genótipos de soja com ausência de lipoxigenases (LOX) e das subunidades α' e G4 componentes das proteínas de reserva da soja, para verificar o efeito dessas ausências nas características físico-químicas das sementes. Também foi utilizada a técnica de RAPD-PCR para selecionar indivíduos geneticamente mais próximos do progenitor recorrente e na identificação de marcador RAPD ligado ao gene que codifica a subunidade α'. Os genótipos utilizados foram obtidos por meio de cruzamentos de progenitores contrastantes para os caracteres selecionados, seguidos de seleção por técnicas bioquímicas não destrutivas. Observou-se drástica redução na evolução de hexanal em genótipos sem lipoxigenases, indicando melhoria de sabor para alimentação humana. Não foi observada diminuição nos teores de proteína total da semente com a eliminação de lipoxigenases viiiou das subunidades α' e G4. A eliminação da subunidade G4 promoveu aumento do teor de metionina, enquanto com a eliminação de α' ocorreu diminuição no teor desse aminoácido. Foram utilizadas características fenotípicas e a técnica de RAPD-PCR para estimar as distâncias genéticas entre indivíduos das populações F 2 e F 3 (selecionados para ausência de LOX e das subunidades protéicas α' e G4) em relação à linhagem PB 2,3 (linhagem sem LOX 2 e 3) a ser usada como progenitor recorrente. Essas populações foram derivadas dos seguintes cruzamentos: 'PBTN' (sem LOX 1, 2 e 3)]. Tanto os caracteres fenotípicos quanto os marcadores RAPD permitiram a discriminação de indivíduos geneticamente mais próximos da linhagem PB 2,3, sendo sua em fase de repulsão ao gene que codifica a subunidade α'.

This work was carried out on soybean genotypes lacking lipoxygenases and the storage protein effect of this absence on the physical-chemical characteristics of seeds. The RAPD-PCR technique was also used to select individuals genetically more similar to the recurrent parent and to identify the RAPD marker linked to the gene which codifies the α' subunit. The genotypes were obtained from crosses among contrasting progenitors for the selected characters, followed by nondestructive biochemical analysis. A drastic reduction in the evolution of hexanal in genotypes without lipoxygenases was observed, indicating food flavor enhancement. No decrease in total seed protein content was observed with the elimination of lipoxygenases or α' and G4 subunits. The elimination of the G4 subunit promoted an increase in the methionine content where as elimination of the α' subunit caused a decrease in the amount of this amino acid. Phenotypic characteristics and the RAPD-PCR technique were used to estimate the genetic distances among individuals of the F 2 and F 3 populations (selected for the absence of LOX and of the α' and G4 protein subunits) in relation to PB 2,3 (a line without LOX 2 and 3) which will be used as recurrent parent. These populations were derived from the following crosses 'PBTN' (without LOX 1, 2 and 3)]. Both phenotypic characters and the RAPD markers allowed for discriminating individuals genetically more similar to the line PB 2,3, with their combined use being more efficient. The use of the Bulked Segregant Analysis technique made possible the identification of a RAPD marker linked in repulsion to the α' gene.
