Untersuchung der Plasmakinetik und des Metabolismus von L-Theanin bei gesunden Probanden nach Bolusgabe von isoliertem L-Theanin und Grüntee

<strong>Hintergrund</strong>: L-Theanin (5-N-Ethyl-L-Glutamin) ist eine nicht-proteinogene Aminosäure, die v.a. durch den Verzehr von Tee (<em>Camellia sinensis</em>) aufgenommen wird. Da L-Theanin die kognitive Leistungsfähigkeit und die Gemütslage verbessern soll, wird L-Theanin in isolierter Form als Nahrungsergänzungsmittel Suntheanine® hergestellt und in funktionellen Getränken zugesetzt. Als Metabolite von L-Theanin werden Ethylamin und Glutaminsäure betrachtet, basierend auf tierexperimentellen Studien. Daten zum Metabolismus von L-Theanin im menschlichen Organismus fehlen. Daher sollte die vorliegende Studie klären, ob die Bolusgabe von 100 mg L-Theanin in isolierter Form und in Form von Grüntee zu einer Konzentrationsänderung von L-Theanin, Ethylamin, Glutaminsäure u.a. proteinogener Aminosäuren in Plasma, Erythrozyten und Urin von gesunden Probanden führt.<br /><strong>Methode</strong>: In einer randomisierten Crossover-Studie erhielten 12 gesunde Probanden morgens nüchtern 100 mg L-Theanin in isolierter Form als Kapsel (Suntheanine®) und nach einer zweiwöchigen Washout-Phase dieselbe Dosis in Form von 250 ml Grüntee. An zwei weiteren Studientagen erhielten drei Probanden zusätzlich jeweils 50 mg und 200 mg L-Theanin als Kapsel. Am jeweiligen Untersuchungstag wurden 8 Blutentnahmen durchgeführt (vor sowie 0,5 h, 0,75 h, 1 h, 1,5 h, 2 h, 3 h und 24 h nach Bolusgabe) und die Probanden sammelten Nüchtern- und 24-Stunden-Urin. Die Konzentration von L-Theanin, der proteinogenen Aminosäuren und Ethylamin in Plasma, Erythrozyten und Urin wurde nach Vorsäulenderivatisierung mittels reversed-phase HPLC analysiert. Die Konzentrations-Zeit-Kurven von L-Theanin im Plasma wurden mit Hilfe eines Ein-Kompartiment- Modells simuliert. Die statistische Auswertung erfolgte mittels SPSS 19.0.<br /><strong>Ergebnisse</strong>: Die Konzentration von L-Theanin im Plasma stieg nach Bolusgabe von 100 mg L-Theanin an und erreichte nach 24 h wieder die Konzentration zur Baseline. Dabei war die Plasmakinetik von L-Theanin nach Aufnahme beider Studienprodukte vergleichbar und die maximale Plasmakonzentration von L-Theanin wurde 0,8 h nach Aufnahme von Kapsel (24,3 ± 5,7 μmol/l) und Grüntee (26,5 ± 5,2 μmol/l) erreicht. Das berechnete Verteilungsvolumen von 16 bis 17 Litern lässt eine Verteilung von L-Theanin in die peripheren Gewebe und gut durchbluteten Organe vermuten. Der Verzehr beider Studienprodukte führte außerdem zu einem Konzentrationsanstieg von L-Theanin in den Erythrozyten – bis 24 h nach Bolusgabe – und zu einer renalen Ausscheidung von L-Theanin. Die Konzentration von Ethylamin und Glutaminsäure im Plasma stieg nach Intervention ebenfalls an und beide Substanzen wurden mit dem 24-Stunden-Urin ausgeschieden. Die AUC von L-Theanin und Ethylamin im Plasma sowie L-Theanin in den Erythrozyten änderte sich dosisabhängig nach Aufnahme von 50 mg, 100 mg und 200 mg isoliertem L-Theanin. Die Konzentration anderer proteinogener Aminosäuren in Plasma, Erythrozyten und Urin blieb unverändert. Auf Grundlage der ausgeschiedenen Stoffmenge von L-Theanin und Ethylamin im Urin sowie der kinetischen Parameter von L-Theanin im Plasma scheint die Bioverfügbarkeit von L-Theanin zwischen 52% und 74% zu liegen.<br /><strong>Schlussfolgerung</strong>: L-Theanin wird nach Aufnahme von Kapsel und Grüntee rasch resorbiert und scheint größtenteils zu Ethylamin und Glutaminsäure hydrolysiert zu werden, welche mit dem Urin ausgeschieden werden. Ein geringer Anteil von L-Theanin wird intrazellulär retiniert. Plasmakinetik und Metabolismus von L-Theanin sind nach Aufnahme beider Studienprodukte vergleichbar, so dass nach Grünteeverzehr – in Hinblick auf die funktionellen Wirkungen von L-Theanin – vergleichbare Effekte eintreten können wie nach Konsum von isoliertem L-Theanin. Die diskutierten funktionellen Wirkungen von L-Theanin sind demnach möglich, könnten jedoch auch durch potenzielle Endprodukte des Metabolismus von L-Theanin erklärt werden. Da Glutamin im menschlichen Organismus aus Glutaminsäure synthetisiert wird, könnte L-Theanin zudem eine alternative Glutaminquelle für kritisch Kranke darstellen.

<strong>Kinetics of L-Theanine uptake and metabolism in healthy participants after ingestion of L-Theanine via capsules and green tea</strong> <br /><strong>Background</strong>: L-Theanine (5-N-ethyl-L-glutamine) is a non-proteinogenic amino acid primary ingested by tea (<em>Camellia sinensis</em>). Since L-Theanine is suggested to improve cognition and mood, L-Theanine is available as nutritional supplement and is now used as an ingredient in functional drinks. In rats, ethylamine and glutamic acid increased in plasma and urine after L-Theanine intake. Since data on the metabolic fate of L-Theanine from human studies are lacking, the present study investigated the kinetics of L-Theanine uptake and its metabolites ethylamine and glutamic acid in plasma and erythrocytes as well as their urinary excretion after bolus intake of capsules and green tea in healthy participants.<br /><strong>Methods</strong>: Within a randomized crossover trial, 12 participants ingested a bolus of 100 mg L-Theanine after an overnight fast, either in isolated form via capsules (Suntheanine®) or via 250 mL green tea. On further occasions, three participants received an additional bolus of 50 mg and 200 mg L-Theanine via capsules, respectively. All treatments were separated by a washout period of 2 weeks. On each study day, blood and urine were collected before and up to 24 h post consumption (blood: 0,5 h, 0,75 h, 1 h, 1,5 h, 2 h, 3 h and 24 h) to determine the concentration of L-Theanine, proteinogenic amino acids, and of ethylamine in plasma, erythrocytes, and in urine by reversed-phase HPLC following pre-column derivatization. Each plasma concentration-time curve of L-Theanine was fitted using a 1-compartment model. Statistical analysis was performed using SPSS 19.0.<br /><strong>Results</strong>: In plasma, L-Theanine concentration increased time-dependently and returned to baseline 24 h after intake of 100 mg L-Theanine, with similar plasma kinetics after both treatments. Maximum plasma concentration of L-Theanine occurred 0.8 h after intake of 100 mg L-Theanine via capsules (24.3 ± 5.7 μmol/L) and tea (26.5 ± 5.2 μmol/L), respectively. It can be assumed that L-Theanine is not only distributed in plasma, but also in further tissues with regard to the hypothetical distribution volume of 16 - 17 liter. Furthermore, L-Theanine increased in erythrocytes and was excreted by urine with comparable results after both treatments. Ethylamine and glutamic acid increased in plasma and were excreted by urine after intake of capsules and tea. The AUC of L-Theanine and ethylamine in plasma as well as L-Theanine in erythrocytes increased dose-dependently after intake of 50 mg, 100 mg, and 200 mg L-Theanine via capsules. Other proteinogenic amino acids in plasma, erythrocytes and urine were not affected. Considering the renal excretion of L-Theanine and ethylamine within 24 h and the maximum plasma concentration of L-Theanine in plasma with regard to the distribution volume, 52% - 74% of ingested L-Theanine should reach the organism, respectively. <br /><strong>Conclusion</strong>: L-Theanine is rapidly absorbed in healthy participants after intake of capsules and green tea. The major part of L-Theanine seems to be hydrolyzed to ethylamine and glutamic acid, which are excreted by urine. A minor part of L-Theanine is retained in erythrocytes. Kinetics and urinary excretion of L-Theanine, ethylamine, and glutamic acid are comparable after intake of capsules and green tea. Thus, functional effects of L-Theanine intake are plausible and may result from L-Theanine, ethylamine, glutamic acid, or other by-products of the metabolism of L-Theanine. As glutamic acid is converted to glutamine, future studies should investigate if L-Theanine may be an alternative source of glutamine which is essential for critically ill patients.
