Strontium-, Barium-, Cadmium-, Kupfer-, Zink-, Mangan-, Chrom- und Antimonkonzentrationen in Leber, Nierenrinde und Nierenmark der Spezies Katze, Hund und Pferd in Abhängigkeit von Alter und Geschlecht | The strontium, barium, cadmium, copper, zinc, manganese, chromium and antimony concentrations in liver, kidney cortex and kidney medulla of the species cat, dog and horse, stratified by age and sex

Die vorliegende Arbeit hatte das Ziel, die Gehalte von Strontium (Sr), Barium (Ba), Cadmium (Cd), Kupfer (Cu), Zink (Zn), Mangan (Mn), Chrom (Cr) und Antimon (Sb) in unterschiedlichen Geweben (Leber, Nierenrinde und Nierenmark) von drei Spezies (Hund, Katze und Pferd) zu bestimmen. Die Konzentrationsmessungen der Elemente erfolgten nach Mikrowellenaufschluss mittels induktiv gekoppelter Plasma-Massenspektrometrie (ICP-MS). Die Gewebeproben von 50 Hunden, 47 Katzen und 10 Pferden wurden im Institut für Tierpathologie der Freien Universität Berlin von eingeschläferten Tieren entnommen. Elf Pferdeproben wurden von einem Schlachtbetrieb zur Verfügung gestellt. Die drei Spezies wiesen in den Lebern sehr ähnliche Sr- Konzentrationen auf. Katzen und Pferde zeigten im Nierenmark, Hunde in der Nierenrinde die höchsten Sr-Konzentrationen (MWT; p < 0,05). Eine Geschlechtsabhängigkeit konnte in Leber und Nierenmark der Hunde statistisch nachgewiesen werden. Hündinnen akkumulierten mehr Strontium in diesen beiden Geweben (MWT; p < 0,05). Die 5 bis 8jährigen Stuten speicherten im Vergleich zu den Wallachen/Hengsten tendenziell mehr Strontium im Nierenmark. Bezogen auf die anderen Altersgruppen akkumulierten die jüngsten Katzen (0 bis 1 Jahr) die höchsten Sr-Konzentrationen in den untersuchten Geweben. In allen Geweben der Hunde und Katzen wurden ähnliche Ba-Konzentrationen gemessen. Die männlichen Tiere der drei Spezies akkumulierten tendenziell mehr Barium als die weiblichen Artgenossen. Generell zeichneten sich die Pferde im Vergleich zu den Katzen und Hunden durch die höchsten Cd-Konzentrationen in den Geweben aus. Innerhalb der jeweiligen Spezies wiesen die Nierenrinden die größten Konzentrationen auf. Stuten und Hündinnen speicherten mehr Cadmium in den Lebern, weibliche Katzen in den Nierenrinden als ihre männlichen Artgenossen (p < 0,05). Hunde und Katzen zeichneten sich durch die höchsten Zn- Konzentrationen in den Lebern aus, während Pferde fast ähnliche Zn- Konzentrationen in Leber und Nierenrinde aufwiesen. Hündinnen akkumulierten mehr Zink in Leber und Nierenrinde als die Rüden (p < 0,05). Auffällig war, dass die Zn-Konzentrationen in den Lebern der Katzen über denen der Pferde und Hunde lagen. Bei der Betrachtung der Cu- und Mn-Konzentrationen fiel eine hohe Konzentration in den Lebern der jeweiligen Spezies auf. Beim Vergleich der drei Spezies miteinander wiesen die Hunde in allen Geweben die höchsten Cu- Konzentrationen auf. Hengste und Wallache speicherten tendenziell größere Mengen an Kupfer in Leber und Nierenrinde als Stuten. Bei Stuten wurde in der Nierenrinde etwa die Hälfte der Mn-Konzentration der Hengste/Wallache ermittelt (p < 0,05). Katzen zeigten generell die höchsten Cr-Konzentrationen in allen Geweben. Eine Geschlechtsabhängigkeit (p < 0,05) bestand bei den Hunden, wobei die Hündinnen mehr Chrom akkumulierten als die Rüden. Bei Hunden und Katzen konnten in allen Geweben ähnliche Sb-Konzentrationen nachgewiesen werden, wobei diese über denen der ermittelten Konzentrationen in Leber, Nierenrinde und Nierenmark der Pferde lagen. Bei Rüden wurde in der Leber eine Sb-Konzentration, die etwa ein Zehntel unter den Sb-Konzentrationen der Hündinnen lagen, ermittelt. In Abhängigkeit von Alter und Geschlecht akkumulierten die Hündinnen mehr Antimon in allen Geweben als die Rüden. Im Gegensatz zu den Hunden und Katzen wurden bei den Stuten in den untersuchten Geweben keine Sb-Konzentrationen gemessen.

The present work aimed at determining the concentrations of the elements strontium (Sr), barium (Ba), cadmium (Cd), copper (Cu), zinc (Zn), manganese (Mn), chromium (Cr) and antimony (Sb) in different tissues of three animal species. To quantify the elements, inductive coupled plasma mass spectrometry (ICP-MS) was used, after the samples had been ashed by microwave treatment. In total, tissue samples from liver, kidney cortex and kidney medulla of 50 dogs, 47 cats and 21 horses were investigated. Almost all samples used in this study (dogs, 50; cats, 47; horses, 10) were from animals kindly provided by the Institute of Veterinary Pathology of Freie Universität Berlin. Additional horse samples (n=11) were obtained from a slaughterhouse. The analysis of strontium showed that liver samples of all three species contained similar concentrations of the element. The highest strontium concentrations were found in the medulla of kidney of cats and horses, and in the kidney cortex of dogs (Mann-Whitney Test, p < 0.05). Furthermore, a sex-dependent strontium accumulation was observed in the liver and the medulla of kidney of dogs (p < 0.05). The female dogs accumulated more strontium in the both tissues. Other studies stratifying for sex and age indicated a trend that female horses (age group: 5 - 8 years) accumulated increased amounts of strontium in the medulla of kidney, when compared to males. However, all the different tissue samples of the youngest female cats (age group: 0 – 1 year), by trend, seemed to have accumulated the highest strontium concentrations, compared to the other age groups of the cats. For barium, similar median values were obtained in the dog and cat samples. In tendency, all the male animals within the three species, accumulated more barium than the respective female animals. Concerning the cadmium analysis, the horse samples showed the highest cadmium accumulation compared to cats and dogs. Intra-species-studies demonstrated that the kidney cortices displayed the highest cadmium concentrations. Compared to males, female horses and dogs, respectively, stored more cadmium in their livers, whereas female cats stored cadmium much more in the cortices of kidney (p < 0.05). The studies investigating zinc showed that dogs and cats displayed characteristically the highest zinc concentrations in their livers, whereas horses contained very similar zinc concentrations in both, liver and kidney cortex. Sex-specific investigations showed that female dogs accumulated more zinc in livers and cortices of kidney than males (p < 0.05). Notably, the overall zinc concentrations were much higher in liver samples of cats compared to horses and dogs. Analysis of copper and manganese revealed that both elements accumulated highest in the liver within each species. Interspecies comparisons showed that dog samples displayed the highest copper amounts compared to both other species. Furthermore, by trend, male horses stored higher amounts of copper in livers and cortices of kidney than females. With respect to manganese, female horses were found to accumulate about half the amount of the element in their kidney cortices, when compared to males (p < 0.05). Interspecies studies on chromium accumulation revealed that, in general, cats showed the highest chromium accumulation in all the tissue samples studied here. In dogs, a sex specific dependency (p < 0.05) could be observed, whereby females accumulated more chromium than male dogs. All the different tissue samples of dogs and cats investigated showed similar antimony concentrations, which all were higher than in liver, in kidney cortex and in the medulla of kidney from horse samples. Notably, the liver samples from male dogs contained tenfold lower antimony concentrations than female samples. In general, when stratified by sex and age, female dogs did accumulate more antimony in all respective tissues than males. In contrast to dogs and cats, no antimony accumulation was detectable in any of the different tissue samples from female horses.
