Wie Hunde und andere Tierarten der Ordnung Carnivora nehmen Katzen eine Sonderstellung in Bezug auf den Vitamin-A-Stoffwechsel ein: Sie besitzen physiologisch hohe Vitamin-A-Konzentrationen im Blut. Ursache dafür ist das Vorhandensein eines erhöhten Anteils an Retinylestern, die unspezifisch im Blut durch Lipoproteine transportiert werden. Während beim Mensch und bei Ratten dieser unspezifische Transportweg nur in Zusammenhang mit einer Vitamin-A-Hypervitaminose zu beobachten ist, tritt dieser bei Katzen und anderen Karnivoren physiologisch neben dem streng homöostatisch regulierten Transport von Retinol durch RBP auf. Die für Hunde und andere Kaniden belegte physiologische Ausscheidung von Vitamin A mit dem Harn konnte für Katzen bisher nicht nachgewiesen werden. Diese Beobachtung wird als eine Ursache der erhöhten Anfälligkeit von Katzen angenommen, auf langfristige Aufnahmen großer Mengen Vitamin A mit den typischen Symptomen einer Vitamin-A-Intoxikation zu reagieren. Ziel dieser Arbeit war die Analyse der Konzentration von Retinol, Retinylestern und des RBP sowie der Carotinoide im Blutplasma, Harn, Leber und Nieren von zufällig ausgewählten Hauskatzen. Eingebunden in die Untersuchung war die Analyse des Vitamin-A-Stoffwechsels im Organismus nierenkranker Katzen. Daher wurden von 35 klinisch gesunden Katzen (unterschiedlich in Rasse, Geschlecht und Alter) Blut- und Harnproben genommen. Zusätzlich wurden von 12 Tieren, die aus unterschiedlichsten Gründen euthanasiert wurden, Blut-, Harn-, Gallenflüssigkeits-, Lebergewebs- und Nierengewebsproben entnommen. Da die Proben konsekutiv in einer Tierarztpraxis gesammelt wurden, waren kaum Informationen zum Ernährungsstatus erhältlich. Die Vitamin-A- (Retinol und Retinylester) sowie die Carotinoidbestimmung erfolgten mittels Umkehrphasen- Hochdruck-Flüssigkeits-Chromatographie (RP-HPLC). Nach SDSPolyacrylamidgel- Elektrophorese der Blut- und Harnproben wurde im Western-Blot das RBP nachgewiesen. Gleichzeitig erfolgte die immunhistologische RBP-Analyse an Leber und Nierenschnitten. Vitamin A wurde in allen untersuchten Proben (Blut, Harn, Gallenflüssigkeit, Leber und Niere) in Form von Retinol und Retinylestern (Retinololeat, -palmitat und -stearat) analysiert. RBP konnte in allen Blutproben sowie in den Hepatozyten der Leber und den Epithelzellen des proximal gewundenen Nierentubulussystems, nicht jedoch im Harn gesunder Katzen nachgewiesen werden. Die Gesamtkonzentration an Vitamin A im Blutplasma betrug im Durchschnitt 976 ± 458 ng/ml. Retinol hatte einen Anteil von 25 % an der Vitamin-A-Gesamtkonzentration. Der überwiegende Teil des Vitamin A liegt im Katzenblut jedoch als Retinylester vor (75 %). Auch bei den Retinylestern ist ein deutliches Verteilungsmuster zu erkennen. Retinylstearat dominiert mit 60 % des Gesamtestergehaltes, während der Gehalt an Retinylpalmitat 37 % und Retinyloleat lediglich 4 % des Gesamtestergehaltes betrug. Im Vergleich zu anderen Karnivoren haben Katzen sehr hohe Vitamin-A-Konzentrationen in der Leber. Da jedoch innerhalb der Ordnung Carnivora große Spezies spezifische Unterschiede der Leberwerte auftreten, kann eine Beurteilung des Vitamin-A-Status anhand der Vitamin-A-Speicherkapazität der Leber nur bei Kenntnis entsprechender Grunddaten erfolgen, die jedoch definierte Fütterungsversuche erfordern. Wie auch bei anderen Karnivoren ist die Leber der Hauptspeicher für Vitamin A. Dabei unterscheidet sich jedoch die Vitamin-A-Konzentration der Leber von Katzen (4314 ± 2043 µg/g) deutlich von der anderer Carnivoren. Dagegen können in der Niere von Katzen nur sehr geringe Vitamin-A-Konzentrationen (11,4 ± 8,2 µg/g) detektiert werden. Die Nieren von Katzen scheinen daher eine untergeordnete Rolle bei der Vitamin-A-Speicherung zu spielen. Die Vitamin-A-Konzentration in der Nierenrinde war höher als im Nierenmark. Im Gegensatz zu früheren Untersuchungen konnte bei der Hälfte der untersuchten Katzen Vitamin A im Harn in Form von Retinol (24 ± 80 ng/ml) und Retinylestern (20 ± 62 ng /ml) analysiert werden. Somit konnte erstmals nachgewiesen werden, dass Katzen auch in der Lage sind, Vitamin A - wenn auch in geringen Mengen - mit dem Harn auszuscheiden. Da diese Verbindungen lipophilen Charakter haben, müssen sie im wässrigen Milieu des Harns an ein bisher unbekanntes Protein gebunden vorliegen. Da RBP in keiner Harnprobe gesunder Katzen analysiert wurde, scheidet es als mögliches Retinol-Trägerprotein im Harn aus. Das eventuelle Vorhandensein eines dem Kaniden THP ähnlichen Proteins bei Katzen bedarf weiterer Untersuchungen. Das Unvermögen der Katzen, größere Mengen an Vitamin A mit dem Harn auszuscheiden, könnte als eine mögliche Ursache für deren erhöhte Anfälligkeit gegenüber chronischer Vitamin-A-Hypervitaminose betrachtet werden. Im Gegensatz zu den gesunden Katzen wurden bei nierenkranken Katzen, die auch veränderte Lipidstoffwechselwerte aufwiesen, deutliche Unterschiede im Vitamin-A-Stoffwechsel festgestellt. Bedingt durch tubuläre Schäden des Nierengewebes kommt es zu Veränderungen im RBP- Metabolismus. Eine massive Ansammlung von RBP im Bereich der Epithelzellen des proximalen Tubulussystems konnte in den histologischen Schnitten beobachtet werden. Gleichzeitig kommt es bei vier Tieren zur Ausscheidung von RBP- gebundenem Retinol mit dem Harn. Möglicherweise können die stark erhöhten Retinylesterwerte im Blutplasma nierenkranker Katzen auf eine verminderte renale Ausscheidung der Retinylester zurückgeführt werden. Eine Korrelation zwischen den Schädigungen im Nierengewebe und der RBP-Ausscheidung konnte jedoch nicht nachgewiesen werden. Der Gesamtesterwert der Harnproben der kranken Katzen (durchschnittlich 2,77 ng/ml) ist gegenüber den gesunden Tieren (durchschnittlich 20,5 ng/ml) deutlich reduziert. Neben der Ausscheidung von Vitamin A über den Harn konnte auch die Ausscheidung von Retinol (0,67 µg/ml) mit der Gallenflüssigkeit nachgewiesen werden. Im Blutplasma aller untersuchten Katzen konnte β-Carotin in einer durchschnittlichen Konzentration von 14 ng/ml analysiert werden. Daneben wurden ebenfalls Lycopin und Lutein in ähnlicher Konzentration sowie Spuren von α-Carotin, β-Cryptoxanthin und Zeaxanthin nachgewiesen. Diese Ergebnisse lassen vermuten, dass β-Carotin und Carotinoide nicht nur in pharmakologischen Mengen aufgenommen, sondern physiologisch aus dem Futter intestinal resorbiert werden. Weder β-Carotin, noch andere Carotinoide konnten in der Leber, Gallenflüssigkeit, Niere oder im Harn nachgewiesen werden. Erkrankte Tiere weisen als Ausdruck einer möglichen Unterversorgung bzw. eines erhöhten Bedarfs deutlich geringere Konzentrationen an Carotin und anderen Carotenoiden im Blutplasma auf. Dieser Ansatzpunkt sollte bei der Versorgung kranker Katzen Berücksichtigung finden. Katzen sind zwar nicht in der Lage, Vitamin A aus β-Carotin zu synthetisieren, jedoch stellen sie ein potentielles Modell zur Untersuchung von Carotinoiden als Antioxidanzien und Immunmodulatoren dar, da es keine Beeinflussung durch die Pro-Vitamin-A-Wirkung gibt. Katzen nehmen also auch innerhalb der Ordnung Carnivora eine Sonderstellung hinsichtlich der Ausscheidung von Vitamin A mit dem Harn und des Carotinoidstoffwechsels ein und bieten noch einige interessante Ansatzpunkte für weitere Untersuchungen zur Erforschung des Vitamin-A-Stoffwechsels von Carnivoren.

Cats and other species of the order Carnivora possess a particular position with respect to their vitamin A metabolism, because they do transport vitamin A in the blood physiologically as retinyl esters bound to all fractions of plasma lipoproteins. In humans and rodents, however, elevated concentrations of plasma retinyl ester are only observed as a postprandial consequence of vitamin A absorption in the gut or under conditions of an acute or chronic vitamin A intoxication. In cats and carnivores this way of retinyl ester transport in plasma is physiologically accompanied with the transport of retinol bound to retinol binding protein (RBP). In cats, the physiological excretion of vitamin A with the urine, which has been verified in dogs and other canines has not been investigated hitherto. In cats this observation is supposed to be a cause for the increased susceptibility to longterm intake of high amounts of vitamin A leading to typical symptoms of vitamin A intoxication. Therefore, the objective of this study was to determine concentrations of retinol, retinyl esters and RBP as well as carotenoids in plasma, urine, liver and kidneys of randomly selected domestic cats. The analysis of the vitamin A metabolism in cats suffering from several diseases was also part of the investigation. Blood and urine were obtained from 35 clinically healthy cats of different pedigree, sex and age. Additionally, 12 cats were necropsied and samples of liver, bile and kidney were collected for the determination of vitamin A, caroteniods and RBP. Since the samples were randomly obtained from an animal hospital, no information concerning their nutritional status was available. Vitamin A (retinol and retinyl esters) and carotenoids were determined using a modified gradient reversed phase HPLC- system (Waters, Eschborn, Germany). The presence of RBP in plasma and urine was assessed using SDS-polyacrylamidgel electrophoresis and subsequent Western blot analysis. The distribution of RBP in liver and kidney samples was carried out using immunohistochemistry. In general, vitamin A was detected as retinol and retinyl esters (retinyl oleate, palmitate and stearate) in all specimens investigated (plasma, urine, bile, liver and kidney. RBP could be verified in all blood samples as well as in the hepatocytes of the liver and the proximal convoluted tubules of the kidney epithelial cells. RBP was not detectable in urine of healthy cats. The mean total vitamin A concentration in the plasma was 976 ng/ml. In the plasma, vitamin A was predominantly present as retinyl esters (75 %), whilst retinol shared 25 % of total vitamin A concentration. With regard to the retinyl ester pattern, retinyl stearate dominated with 60 % of total ester concentration, whilst retinyl palmitate was representing 37 % and retinyl oleate 4 % of total retinyl esters. In comparison with other carnivores, investigated cats had high vitamin A concentrations in the liver (4314 ± 2043 µg/g). Because there are species specific differences in the hepatic vitamin A concentrations among carnivores, a judgement of the vitamin A status by means of the vitamin A storage capacity of the liver can only be made if basic data are available which, however, requires defined feeding experiments. By contrast, only small vitamin A concentrations (11.4 ± 8.2 µg/g) can be detected in kidneys of cats. The kidneys of cats seem to play only a minor role in the storage of vitamin A. The vitamin A concentration in the renal cortex was higher than those in the renal medulla. Contrary to earlier investigations, in half of the investigated urine samples, vitamin A could be detected in form of retinol (24 ± 80 ng/ml) and retinyl esters (20 ± 62 ng /ml). Thus, it could be verified for the first time that cats are also able to excrete vitamin A physiologically in the urine. Since the lipophilic nature of vitamin A, it must be bound to protein in the aqueous environment of the urine. Because RBP could not be found in any of the urine samples of healthy cats, RBP can be excluded as possible carrier protein for vitamin A in urine. The existence of a similar protein in cats as THP in dogs and other canines needs further investigations. The inability of cats to excrete higher amounts of vitamin A with the urine could be considered as a possible cause for their increased susceptibility to chronic vitamin A intoxication. In contrast to healthy cats, distinct differences were found in the vitamin A metabolism in cats suffering from kidney disease, which showed also changed in lipid metabolism values. Because of the pathohistological alterations in the kidney tubules, modifications in the RBP metabolism can also be noted. Immunohistological sections show an accumulation of RBP in the epithelium cells lining the proximal convoluted tubules of the kidneys. Simultaneously, RBP-bound retinol was detected in urine samples of diseased cats. It is likely that the high concentration of retinyl esters in the plasma of cats suffering from the kidneys disease can be due to a decreased renal excretion of retinyl esters. The total concentration of retinyl esters in urine of diseased cats (mean: 2.77 ng/ml) is clearly reduced in comparison to healthy cats (mean value 20.5 ng/ml). In addition to the renal excretion of vitamin A, an excretion of retinol (0.67 ng/ml) with the bile could also be verified. β-carotene in a mean concentration of 14 ng/ml could be found in plasma of all examined cats. Moreover, lycopene and lutein in a similar concentration as well as traces of α-carotene, β-cryptoxanthin and zeaxanthin were detected. These results suggest that β-carotene and other carotenoids are physiologically absorbed in the intestine from the feed. Neither β-carotene nor any other carotenoids could be verified in liver, bile and kidney or in the urine. Diseased cats show less concentrations of β-carotene and other carotenoids in plasma, which is likelydue to a lower dietary supply and/or raised carotenoid requirement. This fact should be considered during the supply of diseased cats, because cats seem to be unable to synthesize vitamin A from β-carotene. That is why they represent a potential model for the investigation of carotene and carotenoids as antioxidants and immune modulators since there is no influence by the pro-vitamin-A-effect. In conclusion, the results of the study show that domestic cats possess a peculiar role among carnivores with regard to the storage of vitamin A in the liver, the excretion of vitamin A in the urine and metabolism of the β-carotene and carotenoids. These animals still offer a quite interesting model for further investigations.
