Etablierung eines direkten Kokultur-Modells zur Analyse der Interaktion zwischen mikrovaskulären Endothelzellen und epidermalen Keratinozyten | Establishment of a direct co-culture model for analysis of interactions between microvascular endothelial cells and epidermal keratinocytes

In der vorliegenden Arbeit wurden drei direkte in vitro-Kokultur-Modelle aus Endothelzellen und Keratinozyten, die das Studium der Zell-Zell-Interaktion ermöglichen, entwickelt und etabliert. Im Fokus der Untersuchungen standen des Weiteren die Identifizierung der Zelltypen und eine morphologische Charakterisierung. Die zelluläre Interaktion zwischen den Keratinozyten und den Endothelzellen ist vor allem für die Gewebehomöostase im bovinen Zehenendorgan von entscheidender Bedeutung und nimmt somit in der Pathogenese von Klauenerkrankungen eine zentrale Stellung ein. Hierfür wurden die direkten Kokulturen unter denselben standardisierten Kultivierungsbedingungen in drei unterschiedlichen Ansätzen zusammengeführt, die im Rahmen dieser Arbeit als Kokultur-System 1 (K+E), Kokultur-System 2 (E+K) und Kokultur- System 3 (Suspension) benannt wurden. Im Kokultur-System 1 (K+E) wurde zunächst ein subkonfluenter Monolayer aus Keratinozyten auf den mit Kollagen Typ I beschichteten Kulturgefäßoberflächen kultiviert, der im weiteren Sinne die Aufgabe eines „feeder layers“ erfüllen sollte. Zu untersuchen war, ob bereits adhärente im subkonfluenten Monolayer liegende Keratinozyten einen Einfluss auf die Adhärenz und folgende Differenzierung der Endothelzellen ausübten. Im umgekehrten Fall war im Kokultur-System 2 (E+K) zu untersuchen, inwieweit Keratinozyten im Zuge ihrer Adhärenz und Migration auf die Differenzierung bereits in subkonfluenter Monokultur befindende Endothelzellen Einfluss nehmen. Das Ziel der Untersuchungen im Kokultur-System 3 (Suspension) war zu beobachten, wie Endothelzellen und Keratinozyten, die zum selben Zeitpunkt in Kultur gebracht wurden, sich in der Adhärenz und Differenzierung gegenseitig beeinflussen. Die Einsaat der Zellen im Kokultur-System 3 erfolgte in Suspension, somit wurde eine zufällige Verteilung der Zelltypen auf dem Boden der Kulturgefäßoberfläche erreicht. Die Kokulturen wurden im Langzeitmodell jeweils auf beschichteten Glasplättchen beziehungsweise Millicell®-PCF- Filtereinsätzen phasenkontrastmikroskopisch beziehungsweise transmissionselektronenmikroskopisch angezüchtet. Die Kultivierung aller Kokultur-Systeme erfolgte in dem Erhaltungsmedium DMEM+. Gemäß des Versuchaufbaus der Kokultur-Systeme 1 (K+E) und 2 (E+K) wurden die Keratinozyten und die Endothelzellen in Monokultur jeweils über 24 Stunden in dem entsprechenden Selektivmedium inkubiert (s. 3.2). In allen direkten Kokultur-Systemen konnten Endothelzellen und Keratinozyten erfolgreich kokultiviert werden. Die Endothelzellen zeigten in allen Kokultur-Systemen Angiogenese-artige Strukturen, die den charakteristischen Merkmalen in Form der Aussprossung, linearer Aneinanderreihung und der Ausbildung Kapillar ähnlicher Strukturen in vitro ähneln (Stadien- Einteilung gemäß Bahramsoltani, 2003; Käßmeyer, 2006). Die unvollständig differenzierten Keratinozyten zeigten eine stabile Adhärenz auf den mit Kollagen Typ I, einer Komponente der interstitiellen Matrix, beschichteten Glasplättchen und Millicell®-PCF- Filtereinsätzen. Charakteristisch für die Keratinozyten der Kokultur-Systeme 1 bis 3 (K+E, E+K, Suspension) war die Bildung einer mehrlagigen Zellschicht. Nach der vierwöchigen Langzeitkultivierung entstand in allen Kokultur-Systemen eine mehrlagige Kokultur aus Endothelzellen und Keratinozyten. Durch die immunzytochemische Markierung mit einem Antikörper gegen Zytokeratin 14 (CK14) wurden die epidermalen Keratinozyten im Kokultur-System 2 (E+K) identifiziert. Ein Antikörper gegen Zytokeratin 19 (CK19) diente als Marker für die mikrovaskulären Endothelzellen in den auf Glasplättchen kultivierten Kokultur- Systemen 1 (E+K) und 3 (Suspension). In den transmissionselektronenmikroskopischen Untersuchungen erfolgte die Identifizierung der Endothelzellen und Keratinozyten in der frühen Phase der Kultivierung anhand der Lokalisation der Zellen gemäß des Versuchaufbaus des entsprechenden Kokultur-Systems, während in der späten Phase der Kultivierung spezifische morphologische Merkmale eine Identifizierung und Charakterisierung der Zellen ermöglichten. Dabei wurden innerhalb endothelialer Zellkolonien in den Kokultur-Systemen 1 (E+K) und 3 (Suspension) Lumen umschließende Kapillar- ähnliche Strukturen aus Endothelzellen beobachtet. Die durch überlappende Zellenden benachbarter Endothelzellen formierten Hohlräume waren teils mit Zellfragmenten oder elektronenoptisch leer. Im Kokultur-System 2 (E+K) formierten die Endothelzellen eine mehrlagige Zellansammlung, die gekennzeichnet war durch ein Netzwerk aus reich verzweigten Zellen. Des Weiteren zeigten sich Anzeichen für intussuszeptionelle Remodellierung der entstandenen Hohlräume. Die Keratinozyten bildeten in allen Kokultur-Systemen (E+K, K+E, Suspension) eine mehrlagige Zellschicht aus, auf denen die Endothelzellkolonien lokalisiert waren. Charakteristisch für die Keratinozyten war ihre stabile Adhärenz auf den beschichteten Filtermembranen. Transmissionselektronenmikroskopisch konnte eine eindeutige Kompartimentierung durch Ausbildung eines erweiterten mit fibrillärem Material gefüllten extrazellulären Spaltraums zwischen den Endothelzellen und den Keratinozyten beobachtet werden. Resultierend aus den eigenen Beobachtungen scheinen Keratinozyten einen zentralen regulatorischen Einfluss auf die endotheliale Morphogenese auszuüben. Daher sind diese Kokultur-Systeme besonders für weitere Untersuchungen endothelial-epidermaler Regulationsmechanismen geeignet. In künftigen Untersuchungen zur Pathogenese von Klauen- und Hufrehe verspricht der Einsatz der hier entwickelten Kokultur-Systeme neue Erkenntnisse zu den Mechanismen im Initialstadium der Erkrankung.

In the work presented in this thesis three direct in vitro co-culture models of endothelial cells and keratinocytes were developed and established. These models enable studies of cellular interactions between these two cell types. The work focused on identification of the cell types present in culture and a detailed morphological characterization. The cellular interaction between keratinocytes and endothelial cells is of particular relevance for the tissue homeostasis in the bovine claw. Cellular interactions play a key role during the pathogenesis of claw diseases. The direct co-cultures were assembled using three different technical approaches but under identical standardized culture conditions. The three different approaches are identified as coculture system 1 (E+K), co-culture system 2 (K+E) and co-culture system 3 (suspension). In the co-culture system 1 (E+K) initially a sub-confluent monolayer of keratinocytes was established on culture dishes coated with collagen type I. This monolayer served as a kind of “feeder-layer” for the endothelial cells seeded subsequently on top of the keratinocytes. The hypothesis tested with this model was that keratinocytes which adhere to the dish as monolayer already have an influence on adhesion and subsequent differentiation of endothelial cells seeded on top. On the other hand the experiments carried out with the co-culture system 2 (K+E) examined if keratinocytes during their migration and adhesion have an influence on endothelial cells that have established a sub-confluent layer already. The aim of the experiments done with the coculture system 3 (suspension) was to study the mutual influence of endothelial cells and keratinocytes regarding their adhesion and differentiation, when cultured simultaneously. Cell seeding in the co-culture system 3 was done in suspension. Therefore a random distribution of the different cell types on the bottom of the culture dish was accomplished. The co-cultures were grown for extended periods of time (long term model) on coated glass plates or Millicell®-PCF-inserts respectively. Cells were examined using phase contrast microscopy and transmission electron microscopy. A maintenance medium DMEM+ was used for cultivating all co-culture systems. According to the experimental setup of the co-culture systems 1 (K+E) and 2 (E+K), keratinocytes and endothelial cells were incubated in monoculture in the corresponding selective medium for a duration of 24 hours (s. 3.2). Endothelial cells and keratinocytes were successfully cultured in all three direct co-culture systems. In all systems the endothelial cells displayed angiogenic structures with characteristic features of angiogenesis such as sprouting, linear arrangement, and formation of capillary like structures in vitro (stages according to Bahramsoltani, 2003; Kässmeyer, 2006). The incomplete differentiated keratinocytes showed a stable adhesion to the glass plates and Millicell®-PCFinserts, which were coated with collagen type I, a component of the interstitial matrix. Characteristic for the keratinocytes in co-culture system 1 to 3 (K+E, E+K, suspension) was the creation of a stratified layer. After 4 week long term cultivation a multilayered co-culture of endothelial cells and keratinocytes developed in all three systems. The immunocytochemical reaction with an antibody to cytokeratin 14 (CK14) identified epidermal keratinocytes in the co-culture system 2 (E+K). An antibody to cytokeratin 19 (CK19) was used as marker for microvascular endothelial cells in the co-culture systems 1 (K+E) and 3 (suspension). Endothelial cells and keratinocytes were identified by electron microscopic examination in the early stage of cultivation using their localization corresponding to the sequence of seeding in the respective co-culture system as a criterion. During the late stages of cultivation specific morphological criteria enabled the identification and differentiation of these two cell types. Within the endothelial cell colonies in the co-culture systems 1 (E+K) and 3 (suspension) capillary like structures consisting of endothelial cells which enclosed a lumen were detected. The overlapping cell processes of neighbouring endothelial cells surrounded cavities which were either filled with cell fragments resp. fibrous material or were electron lucent. In co- culture system 2 (E+K) the endothelial cells form a multilayered cell cluster, which is characterized by a network of abundant branching cells. Furthermore signs of an intussusceptional remodeling of the cavities were revealed. The keratinocytes build up a stratified layer of cells covered by endothelial cells in all three systems (E+K, K+E, suspension). A stable adhesion on the coated membranes was characteristic for the keratinocytes. Transmission electron microscopic examination clearly showed formation of compartment between endothelial cells and keratinocytes by a dilated intercellular space filled, with fibrillar material. The results presented here support a central regulatory influence of keratinocytes on the endothelial morphogenesis. Consequently these co-culture systems are suitable for further investigation into the endothelial–epidermal interactions. The use of the developed co- culture systems in future studies of the pathogenesis and pathomechanisms of bovine and equine laminitis has the potential to shed light on the cellular interactions occurring during its initial stages.
