Reactivity of anti-ionized calcium-binding adapter molecule 1 (Iba1) antibodies against chicken macrophages by immunohistochemistry

We tested two commercially available anti-ionized calcium-binding adapter molecule 1 (Iba1) antibodies for detecting chicken macrophages in formalin-fixed, paraffin-embedded tissue sections by immunohistochemistry (IHC). We first evaluated the ability of mouse- or rabbit-derived commercial antibodies to recognize chicken Iba1 in IHC, and then determined optimal staining conditions. Chicken macrophages in paraffin sections of pulmonary mycosis were successfully detected with a rabbit anti-Iba1 antibody in combination with preheated Tris-EDTA buffer (pH9) before IHC. In a second experiment, we evaluated the distribution of Iba1-antigen positive cell types by IHC. The rabbit anti-Iba1 antibody reacted with Kupffer cells in the liver, microglial cells in the brain, and macrophage-like cells in the lymphoid organs. Vascular endothelial cells were also positively stained in IHC. Our results suggest that the rabbit anti-Iba1 antibody can be used to detect macrophages in chickens, but it can also react with non-macrophage cell types. Detection of chicken macrophages by IHC using the rabbit anti-Iba1 antibody should be confirmed with morphological assessment of IHC positive cells.
