Avaliação da promoção de crescimento em arroz e trigo por rizóbios nativos simbiontes de Eythrina falcata | Growth promotion in rice and wheat by symbiontic native rizhobia of Erythrina falcata

A utilização de rizóbios como promotores de crescimento em gramíneas são estudadas em muitos trabalhos, pois estes micro-organismos apresentam capacidade de fixação biológica de nitrogênio em plantas leguminosas e possuem outros mecanismos que beneficiam plantas não-leguminosas produzindo fitormônios e aumentando a absorção de nutrientes. O Rio Grande do Sul é um estado reconhecido como produtor de arroz (Oryza sativa) e trigo (Triticum aestivum) produzidas frequentemente onde também ocorrem leguminosas nativas que fazem simbiose com rizóbios, como a corticeira-da- serra (Erythrina falcata). Assim, uma vez que esses rizóbios fazem simbiose com E. falcata podem atuar como promotores de crescimento de plantas de arroz e trigo. Este trabalho teve como objetivo isolar e selecionar rizóbios nativos de E. falcata que possam ser inseridos em lavouras de culturas anuais. Os isolados de rizóbios foram obtidos de nódulos de E. falcata cultivadas em casa de vegetação inoculadas com suspensão de solo. Para realizar a autenticação e avaliar a eficiência simbiótica dos isolados, estes foram re-inoculados em E. falcata e observou-se a nodulação e quantificada a produção de massa seca das plantas. Os rizóbios foram caracterizados quanto à sua morfologia, capacidade de solubilização de fosfato tricálcico (Ca3(PO4)2) e produção de ácido indol-acético (AIA), com e sem adição de triptofano. Ainda, os rizóbios foram avaliados quanto ao efeito na germinação em sementes de arroz e trigo, e à promoção de crescimento em casa de vegetação. Identificaram-se os isolados pela amplificação e sequenciamento do DNA utilizando um fragmento do 16S ribossomal. Apenas os rizóbios Ef4, Ef12 e Ef13 solubilizaram fosfato tricálcico, e todos produziram AIA com adição de triptofano. Os isolados não foram eficientes na fixação de nitrogênio em E. falcata. Por outro lado, todos os rizóbios aceleraram a germinação de sementes de arroz, porém em trigo não houve efeito das inoculações. Os tratamentos com inoculação dos rizóbios Ef3, Ef11 e Ef13 em arroz apresentaram valores semelhantes ao tratamento com 100% de N. O rizóbio Ef13 e Ef4 estimularam o aumento no número de perfilhos em arroz. As plantas de arroz inoculadas com Ef13 foram capazes de absorver mais nitrogênio que os demais tratamentos inoculados. Com base na análise de DNA, todos os isolados foram identificados como pertencentes ao gênero Bradyrhizobium.

The use of rhizobia to growth promoting in grasses has been well studied in several works, because besides their role in promoting fixation of atmospheric nitrogen in leguminous plants, rhizobia may also provide several other benefits in non-leguminous plants by producing phytohormones and facilitating plant nutrients acquisition. Rio Grande do Sul state is recognized as a great rice (Oryza sativa) and wheat (Triticum aestivum) producer. These cultures are often cropped under natural grassland or forest slopes, where there was co-occcurence of native species of legumes fixing atmospheric nitrogen such as Erythrina falcata (Corticeira-da-serra). Then, since these native rhizobia associated with Erythrina falcata can act as growth promoting in rice and wheat plants, our work aimed to isolate and select native rhizobia from corticeira-da-serra, in order to selected rhizobia that are capable to promote growing in those crops. Isolates of rhizobia were obtained from nodules of Corticeira-da-serra cultivated in greenhouse conditions that were previously inoculated with soil suspension. After, in order to perform the authentication and evaluate the symbiotic efficiency of each isolate, we re-inoculated the on Corticeira-da-serra and observed if there was nodulation and determined plant production dry matter. Rhizobia were characterized regarding their morphology, ability to solubilise tricalcium phosphate (Ca3(PO4)2), and indole acetic acid (IAA) production with and without tryptophan. Yet, rhizobia were evaluated regarding their effects on germination of rice and wheat seeds and plant production in greenhouse conditions. We identify the isolates through amplification and DNA sequencing using 16S ribosome fragment. Only rhizobias identified as UFRGS Ef4, Ef12 and Ef13 was able to solubilize tricalcicum phosphate, while all of them could produce IAA with tryptophan. On the other hand, all the isolates increased seeds germination rate of rice but in wheat don’t have effect on inoculation. Rice inoculated with Ef3, Ef11 and Ef13 showed similar values when compared to rice fertilized with 100% of recommended dose of nitrogen. However, our finding showed that Ef13 can stimulate the number of branch. The rice plants inoculated with UFRGS Ef13 absorbed more nitrogen when compared to the others treatments. On wheat, no differences were observed in seed germination and number of branch. Finally, based on dna analysis all the isolates in this work were identified as Bradyrhizobium ssp.
