Study and evaluation of newly created interspecific somatic hybrids between cultivars (Solanum tuberosum L., 2n=4x=48) and wild potato species (S. bulbocastanum, S. tarnii, S. pinnatisectum, 2n=2x=24) via protoplast fusion for selection of resistance to late blight

Protoplast fusion was conducted between late blight resistant wild-type potato (S. bulbocastanum, S. tarnii, S. pinnatisectum) and potato cultivars (Solanum tuberosum L.) to obtain interspecific somatic hybrids. The protocols for protoplast isolation, fusion, and culture were adapted from Morller and Wenzel (1992) and modified by Thieme (1997) used. 1612 calluses from seven different fusion hybrids were generated, of which 188 progeny plants were generated. Flow cytometry (Dinu and Thieme, 2001) and single sequence repeat methods (S. Feingold, 2005; Yu-Su Song et al. 2005) were used to examine all the progeny plants and identify 69 interspecific somatic hybrids which were hexaploid (2n=6x=72). By using detached leaflet assay, the interspecific somatic hybrid plants were able to show the late blight resistant trait; especially the combination between Solanum tuberosum L. and S. bulbocastanum which suggests the protoplast fusion could be used for establishing late blight resistant potato. The progeny plants were characterized and some potential lines (highly resistance to late blight) were selected for further breeding program. This study is essential and contributes to the development of a methodology for selecting and breeding of late blight resistant potato in Vietnam.
