Untersuchungen zur Adaptation pilzlicher Pathogene an Azolwirkstoffe | Analyses of adaptation of fungal pathogens to azole fungicides

German. Der Einsatz von Einzelpräparaten auf Basis von Azolen findet im Agrarbereich Anwendung gegen ein breites Spektrum pilzlicher Pathogene. Zu diesen zählt auch Fusarium culmorum. Die Bekämpfung erfolgt gezielt oder bei gleichzeitiger Behandlung anderer Erreger durch Azolwirkstoffe wie Prochloraz, Prothioconazol, Metconazol, Tebuconazol, Flusilazol, Propiconazol und weiteren. Dabei besteht die Gefahr der Ausbildung von Resistenzen. In unseren Untersuchungen wurde nachgewiesen, dass Fusarium culmorum nach kurzer Zeit an hohe Dosen des Fungizides Tebuconazol adaptiert werden kann und danach eine quantitative Kreuzresistenz gegen alle untersuchten Azolfungizide entwickelt. Dies lässt sich an Hand von Assays auf Agarplatten mit unterschiedlichen Azolkonzentrationen signifikant nachweisen und konnte durch mikroskopische Aufnahmen bestätigt werden. In diesen wurde beobachtet, dass es unter Einfluss von Tebuconazol aber auch von Prochloraz zur Veränderung des Hyphenwachs-tums kommt, die sich durch Anschwellen der Hyphenspitzen ausprägt. In Vitalfärbungen mit Fluores-zeindiazetat konnte die geringere Empfindlichkeit qualitativ nachgewiesen werden. Da F. culmorum humanpathogen ist, besteht durch quantitative Kreuzresistenz die Gefahr, dass im medizinischen Bereich Antimykotika auf Azolbasis ebenfalls eingeschränkt wirken.

English. Azole fungicides are used against a broad spectrum of fungal pathogens in agriculture. Among others, these treatments comprise direct or indirect application of Prochloraz, Prothioconazole, Metconazole, Tebuconazole, Flusilazole, Propiconazole and other azole agents on Fusarium culmorum. These treatments are related to the occurrence of resistance against azole fungicides. In our investigations we observed adaptation of Fusarium culmorum to high doses of Tebuconazole and detected a quantitative cross resistance to all determined azole compounds. In agar plate assays colony diameters were measured microscopically and significant differences between the tebuconazole adapted and non adapted F. culmorum strains were observed. Pictures after fluorescence staining with calcofluor white showed alterations in hyphal growth and swelled hyphal tips. Vital stains by fluoresceine diacetate showed semi quantitatively a lower sensitivity. F. culmorum is a human as well as plant pathogen that is treated in agriculture and in human medicine by azole single compound fungicides/ antimycotica. Hence, quantitative cross resistance leads to restricted effectiveness of azole fungicides against Fusarium culmorum.
