German. Das VP1 des IBDV-Stammes P2 wurde als Fusionsprotein mit dem Maltose-bindenden Protein in E. coli in ausreichender Quantität bei einer Inkubationstemperatur von 20 °C exprimiert. Die nachfolgende Analyse des mittels Affinitätschromatographie gereinigten Fusionsproteins ergab, dass neben dem vollständigen Fusionsprotein auch C-terminal verkürzte Fusionsproteine vorhanden waren. Mit diesem gereinigten VP1-enthaltenden Fusionsprotein wurde keine RNA-abhängige RNA-Polymeraseaktivität nachgewiesen. Anschließend wurde das VP1 des IBDV-Stammes P2 unter Nutzung eines rekombinanten Baculovirus in eukaryoten Sf9-Zellen exprimiert. Dabei wurde ebenfalls eine Expression C-terminal verkürzter VP1-Proteine neben dem vollständigen VP1 beobachtet. Das Baculovirus-exprimierte VP1 zeigte in vitro RNA-abhängige RNA-Polymeraseaktivität. Damit wurde erstmalig eine RNA-abhängige RNA-Polymeraseaktivität für das VP1 des IBDV nachgewiesen. Das Reaktionsprodukt der VP1-abhängigen Polymeraseaktivität war eine doppelsträngige RNA, welche an einer IBDV-spezifischen Matrize synthetisiert wurde. Die optimale Reaktionstemperatur für die Polymeraseaktivität war unter den gewählten in vitro-Bedingungen 37 °C. Die Anwesenheit von zweiwertigen Metallkationen war notwendig, wobei nach Zusatz von Mg2+-Ionen die höchste Effizienz der Polymerasereaktion zu beobachten war. Nach Zusatz anderer zweiwertiger Metallkationen (Co2+, Mn2+) wurde eine reduzierte Polymeraseaktivität des VP1 beobachtet. Da nach Zusatz von weiteren zweiwertigen Kationen keine Polymeraseaktivität zu beobachten war, besitzt das VP1 wahrscheinlich eine Selektivität bezüglich notwendiger zweiwertiger Ionen. Nach Mutation der Aminosäure Asparagin (D) an Position 416 des VP1 zu Alanin (A) zeigte ein im Baculovirus-System exprimiertes VP1-D416A in vitro keine Polymeraseaktivität. Damit besitzt diese Aminosäure eine Bedeutung für die RNA-abhängige RNA-Polymeraseaktivität des VP1 und stellt somit somit in Übereinstimmung mit der Computeranalyse sehr wahrscheinlich einen wesentlichen Bestandteil des Motivs A der katalytischen „Handflächen“-Domäne des VP1 dar. Dieses Ergebnis fand in vivo mittels des reversen genetischen Systems für IBDV eine Bestätigung, da nach Substitution der Aminosäure 416 kein infektiöses Virus generiert wurde.

English. VP1 of IBDV-strain P2 was expressed as fusion protein with maltose-binding protein in E. coli. Sufficient quantities were obtained at an incubation temperature of 20 °C. The subsequent analysis of the fusion protein, which was purified by affinity chromatography, showed the existence of both full-length and C-terminally truncated fusion proteins. By using this purified VP1-containing fusion protein no RNA-dependent RNA-polymerase activity was observed. VP1 of IBDV strain P2 was then expressed in eucaryotic SF9 cells using a recombinant baculovirus. An expression of C-terminally truncated VP1 together with full-length VP1 was observed as before. VP1 expressed with recombinant baculovirus possessed in vitro RNA-dependent RNA-polymerase (RdRp) activity was shown here for the first time. The reaction product of the VP1-dependent polymerase activity was a double-stranded RNA, which was synthesised at an IBDV-specific template. The temperature optimum for polymerase activity was 37 °C under the chosen in vitro conditions. The presence of divalent metal cations was necessary for enzyme activity. The highest efficiency of the polymerase reaction was observed after addition of Mg2+-ions. Furthermore, addition of other divalent metal cations (Co2+, Mn2+) resulted in a reduced polymerase activity. Since no polymerase activity was observed after addition of further divalent cations, VP1 probably possesses a selectivity regarding the necessary divalent cations. A baculovirus expressed VP1 mutant (VP1-D416A) which contained a mutation of amino acid asparagine (D) at position 416 of VP1 to alanine (A) did not show in vitro polymerase activity. Therefore this amino acid is important for the RNA-dependent RNA-polymerase activity of VP1 and thus probably represents a vital part of the motif A of the catalytic “palm” domain of RdRp in agreement with the computer analysis. This result was supported in vivo by using the reverse genetics system for IBDV, since generation of infectious virus was not observed after substitution of amino acid 416.
