Der Membrananker des pestiviralen Glykoproteins Erns - Untersuchungen zur Lipidbindungsspezifität und Struktur

German. Das Glykoprotein Erns ist ein 42-48 kDa großes, essentielles Strukturprotein und zudem ein bedeutender Virulenzfaktor von Pestiviren. Für die letztgenannte Funktion ist die intrinsische RNase-Aktivität von Erns verantwortlich, die auch für die Ausbildung einer Persistenz im Fötus benötigt wird, sowie das für die Homodimerisierung erforderliche Cystein 171. Es wird davon ausgegangen, dass die enzymatische Aktivität von Erns ein wesentlicher Faktor für die Repression der angeborenen Immunantwort nach einer Pestivirusinfektion darstellt. Den bisherigen Hypothesen zufolge ist dazu die seit längerem bekannte partielle Sekretion des Erns von Bedeutung, da größere Mengen an sekretiertem Erns im Blut infizierter Tiere detektiert werden können. Die partielle Sekretion des Erns wird mit der besonderen Membranverankerung dieses Proteins in Zusammenhang gebracht. Erns ist nicht über eine Transmembranregion am Viruspartikel verankert, sondern über eine amphipatische Helix. Nach den bisher vorliegenden Daten ist diese Struktur neben der Verankerung auch für die intrazelluläre Lokalisation des Proteins und seine Freisetzung aus dem Polyprotein durch die zelluläre Signalpeptidase verantwortlich, für die ansonsten nur Substrate bekannt sind, bei denen die Spaltstelle direkt auf eine Transmembranregion folgt. Somit vereint der Erns-Membrananker mehrere wichtige Funktionen in einem relativ kleinen Sequenzabschnitt, nämlich die Membranverankerung, die ungewöhnliche Signalpeptidasespaltstelle sowie die Steuerung des Sekretion/Retentionsmechanismus. Um diese vielfältigen Eigenschaften des Proteins besser verstehen zu können, ist die Ermittlung der Struktur des Membranankers im Kontakt mit der Lipidmembran von entscheidender Bedeutung. Aus diesem Grund wurde in der vorliegenden Arbeit zum einen die Bindung des Erns-Membranankers an Lipide in einem in vitro System untersucht und zum anderen die Struktur und die Orientierung des Membranankers in der Membran näher analysiert. Dabei konnte entdeckt werden, dass der Erns-Membrananker spezifisch an negativ geladene Lipide bindet, die positive Gesamtladung des Proteins dafür aber nicht verantwortlich ist. Auch die meisten Fragmente des Erns-Membranankers wiesen eine spezifische Bindung an negativ geladene Lipide auf. Zudem wiesen Mutationen, die im biologischen System eine drastische Auswirkung auf die Membranbindung und die Sekretionsrate hatten, keine Änderung in der Lipidbindungsfähigkeit auf. Somit müssen diese Mutationen im biologischen System andere Effekte abseits der reinen Membranbindung auslösen, um für ihren Phänotyp zu sorgen. Durch CD-Analysen konnte die erwartete starke helikale Faltung des Erns-Membranankers bestätigt werden. Im Gegensatz zur erwarteten planaren Orientierung der helikal gefalteten Bereiche ergaben orientierte CD-Analysen, dass sich der Erns-Membrananker nicht planar, sondern leicht schräg zur Membranoberfläche orientiert. Durch NMR-Analysen des C-terminalen Bereiches des Erns-Membranankers in einem Bizellensystem konnte anschließend die helikal gefaltete Region näher eingegrenzt werden. Zudem gelang es, über die Messung der Protonenaustauschrate der NH-Gruppe den Wasserkontakt der Aminosäuren zu bestimmen. Ungewöhnlicherweise zeigte die helikal gefaltete, amphipatische Region in weiten Teilen keinen Wasserkontakt, so dass sich ein größerer Abschnitt der Helix in der Lipidphase der Membran befinden muss. Eine solche Membranverankerung wurde bisher noch in keinem weiteren Protein entdeckt. Die in der vorliegenden Arbeit vorgestellten Daten ergaben einen ersten Anhaltspunkt über die biochemische Funktion und Wirkweise des Erns-Membranankers und bilden die Grundlage für weitere Analysen an diesem faszinierenden Protein.

English. The glycoprotein Erns represents one of the four structural proteins of pestiviruses. It is not only an essential component of the virion but also a virulence factor of these viruses. It has an intrinsic RNase activity which is required for establishment of persistent infection in fetuses. It has been suggested that this RNase activity is important to suppress the innate immune response of the host. We and others have put forward the hypothesis that the latter function of Erns correlates with the known partial secretion of the protein. Secretion of the Erns protein is caused by its atypical membrane anchor which is folded as an amphipathic helix. Our previously obtained data show that the Erns membrane anchor not only manages the membrane binding of the protein but also influences the retention/secretion of the protein and achieves the cleavage of the Erns protein from the polyprotein by the cellular signal petidase, which normally cleaves only downstream of a transmembrane helix. In summary, the Erns membrane anchor combines several important functions in a short stretch of sequence, namely membrane anchor, unusual signal petidase cleavage site and control of the retention/secretion ratio. The molecular mechanisms behind these functions can only be understood when the structure and the membrane binding ability of this region is elucidated. For that reason the membrane binding ability of the Erns membrane anchor was analyzed in an in vitro system. In addition, the secondary structure of the Erns membrane anchor was determined in a membrane mimicking environment. The Erns membrane anchor showed a strong and specific binding to negatively charged lipids which didn’t depend on the positive charge of the protein anchor. In addition the Erns membrane anchor was separated into different parts and mostly all fragments showed the same binding ability. Furthermore, mutations which led to a dramatic increase in the secretion rate in cell culture experiments didn’t show any significant difference in the lipid binding compared to the wild typ. Therefore the biological effects of these mutations couldn’t depend on changes in membrane binding efficiency but should be correlated with other features of the Erns membrane anchor. To have a look on the structure of the membrane anchor we used CD Spectroscopy to determinate the secondary structure. As expected, the membrane anchor showed a strong helical folding. But the orientation of the helical region was not as expected because orientated CD Spectroscopy indicated that the helical region slightly inclines into the membrane. For further understanding of the membrane anchor we used NMR Spectroscopy to obtain a deeper insight into the structure of the c-terminal end of the membrane anchor. To this end we determined the positions of the amino acids in a bicelle system by measuring the proton exchanging rate of the NH group. This technique approach allowed to distinguish between an amino acid which is located in the membrane and surrounded by lipids from an amino acid which is located in the solvents and surrounded by water. A surprising result of these analyses was that a large part of the amphipathic helix is located in the membrane and not as expected at the membrane-water interface. Such a type of membrane anchor is described here for the first time. This present work offers new insight into the biochemical and structural function of the Erns membrane anchor which will provide a better understanding of this fascinating protein and its biological functions.
