Avian bornaviruses – from discovery to control | Aviäre Bornaviren – von der Entdeckung zur Bekämpfung

English. Avian bornaviruses were first discovered in 2008 and constitute a genetically diverse group of viruses within the genus Orthobornavirus (family Bornaviridae). To date, at least 15 avian bornaviruses are known in psittacine, passerine and aquatic birds. While relatively little is known about the pathogenic potential of passerine and waterbird bornaviruses, parrot bornaviruses (PaBVs) are of marked clinical relevance as the primary cause of proventricular dilatation disease (PDD). PDD was first described in the late 1970s as an often fatal disease characterized by chronic gastrointestinal or neurological disorders, but also sudden fatalities and subclinical courses may occur. It is distributed in captive psittacine populations worldwide and threatens private bird collections, zoological gardens as well as rehabilitation projects of endangered species. This habilitation treatise summarizes the results of a study initiated in 2011. Using optimized as well as self-established diagnostic tools, my group discovered several novel avian bornaviruses in parrots and finches, thereby contributing markedly to the growing knowledge on the genetic diversity of the Bornaviridae family. Extensive field studies and phylogenetic analyses provided insights into their epidemiology and distribution in various captive and free-ranging avian populations. Experimental infection models in cockatiels (Nymphicus hollandicus) and domestic canaries (Serinus canaria) revealed valuable information on possible transmission routes, course of infection and the pathogenesis of the disease. Furthermore, they facilitated the evaluation of vaccines. Using viral vectors vaccines encoding for avian bornavirus antigens, my group developed the first vaccination strategy providing protection against avian bornavirus infection and the development of PDD. Particularly the “modified vaccinia virus Ankara” (MVA) proved to be a promising tool for future control of the disease.

German. Aviäre Bornaviren wurden erstmals im Jahr 2008 beschrieben und sie bilden eine genetisch variable Gruppe innerhalb der Gattung Orthobornavirus in der Familie der Bornaviridae. Bisher wurden mindestens 15 verschiedene aviäre Bornaviren bei Psittaziden, Sperlingsvögel und Wasservögeln beschrieben. Während über das Krankheitspotential der Bornaviren der beiden letztgenannten nur sehr wenig bekannt ist, besitzen die Bornaviren der Psittaziden große veterinärmedizinische Relevanz als Erreger der „neuropathischen Drüsenmagendilatation“ (proventricular dilatation disease, PDD). Die PDD wurde erstmals Ende der 1970er Jahre als eine oftmals tödliche, zumeist durch chronische gastrointestinale oder neurologische Verläufe gekennzeichnete Erkrankung beschrieben. Sie ist bei in Menschenhand gehaltenen Psittaziden weltweit verbreitet und betrifft nicht nur Privathaltungen und zoologische Gärten, sondern gefährdet auch Zucht- und Auswilderungsprojekte gefährdeter Papageienarten. Diese Habilitationsschrift beschreibt und diskutiert die Ergebnisse seit Studienbeginn im Jahr 2011. Mit Hilfe optimierter und neu etablierter diagnostischer Methoden gelang es meiner Arbeitsgruppe durch die Erstbeschreibung mehrerer Bornaviren von Papageien und Finken das Wissen über die genetische und biologische Variabilität dieser Virusgruppe erheblich zu erweitern. Umfangreiche Feldstudien in verschiedenen gehaltenen und wildlebenden Vogelpopulationen, unterstützt durch phylogenetische Analysen, lieferten zudem wertvolle Einblicke in die Verbreitung und Epidemiologie der aviären Bornaviren. Mit Hilfe experimenteller Infektionen von Nymphensittichen (Nymphicus hollandicus) und Kanarienvögeln (Serinus canarius) konnten wir wichtige Informationen zu Infektionsverläufen, Krankheitsentstehung und potentiellen Infektionsrouten sammeln. Darüber hinaus stellten die Infektionsmodelle auch die Voraussetzung für die Impfstofftestung dar. Basierend auf viralen Vektorvakzinen, insbesondere dem ‘Modifizierten Vakziniavirus Ankara’ (MVA), die Antigene aviärer Bornaviren exprimieren, gelang uns die Entwicklung der ersten Impfstrategie, die sowohl die aviäre Bornavirusinfektion als auch die Entstehung der PDD verhindern kann.
