Das Verbundprojekt GENMON: Einrichtung eines genetischen Langzeit- Monitorings in Buchenbeständen (Fagus sylvatica L.) | The joint research project GENMON:Implementation of long-term genetic monitoring in beech stands (Fagus sylvatica L.)

German. Genetische Vielfalt ist eine wesentliche Grundlage von Waldbaumpopulationen, um sich an Veränderungen der Umweltbedingungen aus eigener Kraft anzupassen. Deshalb sind für die Baumart Buche deutschlandweit insgesamt 14 Monitoringflächen eingerichtet worden, auf denen sowohl der Zustand als auch die räumlichen und zeitlichen Veränderungen genetischer Vielfalt und der dazugehörigen Mechanismen erfasst werden. Damit ist die Basis für ein Beobachtungssystem geschaffen worden, das künftig die Wirkung von Einflussfaktoren auf die Anpassungsfähigkeit von Baumpopulationen abschätzen und bewerten soll. Die genetische Charakterisierung der Bestände erfolgt mit Hilfe hochvariabler DNA-Marker (Mikrosatelliten). Diese ermöglichen die Berechnung von genetischen Vielfalts- und Differenzierungsparametern in Altbeständen und deren Nachkommenschaften und lassen über die Rekonstruktion der Bestandeshistorie auf Basis räumlich-genetischer Strukturen Rückschlüsse auf anthropogene Einflüsse zu. Weitere Analysen und Datenauswertungen ermöglichen künftig die Beurteilung der Funktionsfähigkeit genetischer Mechanismen, die die Erhaltung, Erzeugung (Neukombination) und Weitergabe (Ausbreitung in Raum und Zeit über Pollen und Samen) genetischer Vielfalt an die nächste Generation gewährleisten Im Rahmen des Projekts sollen auch anpassungsrelevante Genmarker entwickelt werden. Da die Spätfrosttoleranz ein entscheidendes Selektionskriterium im Klimawandel darstellt, werden in Ergänzung zu den neutralen Mikrosatelliten mittels genomweiter Analysen SNP-Marker für das Merkmal „Blattaustrieb“ evaluiert. Die Entwicklung solcher Marker und ihre Erprobung hinsichtlich möglicher Korrelationen mit phänologischen Merkmalen sollen künftig wesentlich zur Beurteilung der klimatischen Anpassungsfähigkeit von Waldbeständen beitragen.

English. Genetic diversity is an essential basis for forest tree populations to adapt to changes in environmental conditions. For this reason, a total of 14 plots have been set up for beech throughout Germany to record both the status and the spatial and temporal changes in genetic diversity including associated mechanisms. This provides the basis for a monitoring system which will in future be used to estimate and evaluate the effect of influencing factors on the adaptability of tree populations.The genetic characterisation of the stands is carried out using highly variable DNA markers (microsatellites). These enable the calculation of genetic diversity and differentiation parameters in old stands and their progeny and allow conclusions to be drawn about anthropogenic influences by reconstructing the stand history on the basis of spatial-genetic structures. Further analyses and data evaluation will in future allow the assessment of the intactness (operability) of the genetic mechanisms that ensure the conservation, production (recombination) and transmission (dispersal in space and time via pollen and seeds) of genetic diversity to the next generation. In addition to the neutral microsatellites, gene markers with adaptive relevance will also be developed in the framework of this project. Since late frost tolerance is a decisive selection criterion in climate change, SNP markers for "leaf flushing" will be evaluated by means of genome-wide analyses. The development of such markers and their testing with regard to possible correlations with phenological traits should significantly contribute to the assessment of climatic adaptability of forest stands in future.
