Effects of three Organophosphorus Insecticides on Tropical Anuran Tadpoles: Lethality and Implications to Motor Activity

[EN] Organophosphorus (OP) pesticides are widely used worldwide as pest controls, but they can have negative effects on non-target species such as amphibians, which are the most threatened group of vertebrates. The aim of this study was to evaluate the lethal (median lethal concentration, LC50) and sublethal effects (burst swimming speed and activity of acetylcholinesterase, AChE) of three OP insecticides, chlorpyrifos, diazinon, and monocrotophos on tadpoles of three tropical anuran species from Colombia, Rhinella horribilis, Boana xerophylla, and Engystomops pustulosus. Each species was exposed independently to each of the three OP insecticides under laboratory conditions for 96 h. Chlorpyrifos was the most lethal insecticide in all species (LC50 range: 1.18–1.85 mg L–1), followed by diazinon (LC50: 6.21–7.92 mg L–1) and monocrotophos (LC50: 419–554 mg L–1). Chlorpyrifos fully inhibited the burst swimming speed in all three species, while increased sublethal concentrations of diazinon decreased it in E. pustulosus or did not have an effect in B. xerophylla and R. horribilis. Monocrotophos reduced the burst swimming speed in both B. xerophylla and R. horribilis and inhibited all swimming activity in E. pustulosus. A parallel pattern was found in the levels of AChE activity that was reduced by the insecticides in E. pustulosus and R. horribilis, although B. xerophylla tadpoles showed a hormetic dose-response. The lethal effect of the three OP insecticides on anuran tadpoles at the recommended doses of application in field, and their negative implications to the swimming and AChE activity at low concentrations show the negative impact of these agrochemicals on tropical amphibians. We suggest reevaluating the toxicological categories currently assigned by the Colombian government to these insecticides (monocrotophos > diazinon > chlorpyrifos), taking into consideration the aquatic fauna, as our results of lethality were contrary to this classification.

[ES] Los plaguicidas organofosforados (OP) se utilizan ampliamente en todo el mundo como control de plagas, pero pueden tener efectos negativos en especies no objetivo como los anfibios, que son el grupo de vertebrados más amenazado. El objetivo de este estudio fue evaluar los efectos letales (concentración letal media, CL50) y subletales (velocidad de nado y actividad de la acetilcolinesterasa, AChE) de tres insecticidas OP, clorpirifos, diazinón y monocrotofos, en renacuajos de tres especies de anuros tropicales de Colombia, Rhinella horribilis, Boana xerophylla y Engystomops pustulosus. Para ello, cada especie se expuso de forma independiente a cada uno de los tres insecticidas OP en condiciones de laboratorio durante 96 horas. El clorpirifos fue el insecticida más letal en todas las especies (rango de CL50: 1,18–1,85 mg L–1), seguido del diazinón (CL50: 6,21–7,92 mg L–1) y monocrotofos (CL50: 419–554 mg L–1). El clorpirifos inhibió completamente la velocidad de natación en las tres especies, mientras que el aumento de las concentraciones subletales de diazinón la disminuyó en E. pustulosus o no tuvo efecto en B. xerophylla y R. horribilis. El monocrotofos redujo la velocidad de natación tanto en B. xerophylla como en R. horribilis e inhibió toda la actividad de natación en E. pustulosus. Se encontró un patrón similar en los niveles de actividad de la AChE que fue reducida por los insecticidas en E. pustulosus y R. horribilis, aunque los renacuajos de B. xerophylla mostraron una dosis-respuesta hormética. El efecto letal de los tres insecticidas OP sobre los renacuajos de anuros a las dosis recomendadas de aplicación en campo, y sus implicaciones negativas para la actividad de natación y AChE a bajas concentraciones, muestran el impacto negativo de estos agroquímicos en los anfibios tropicales. Adicionalmente, también sugerimos hacer una reevaluación de las categorías toxicológicas actualmente asignadas por el gobierno colombiano a estos insecticidas (monocrotofos > diazinón > clorpirifos), tomando en consideración la fauna acuática, ya que nuestros resultados de letalidad fueron totalmente contrarios a esta clasificación.
