Effect of Short-Chain Fatty Acids on the Yield of 2,3-Butanediol by <i>Saccharomyces cerevisiae</i> W141: The Synergistic Effect of Acetic Acid and Dissolved Oxygen

As a platform chemical, 2,3-butanediol (2,3-BDO) has been widely used in various industrial fields. To improve the yield of 2,3-BDO produced by <i>Saccharomyces cerevisiae</i> W141, this paper explored the effects of exogenous short-chain fatty acids (SCFAs) as well as the synergistic effects of acetic acid and dissolved oxygen content on the yield of 2,3-BDO from the perspective of physiological metabolism. The results indicated that different SCFAs had different effects on the production of 2,3-BDO, and higher or lower concentrations of SCFAs were not conducive to the generation of 2,3-BDO. However, exogenically adding 1.0 g/L acetic acid significantly increased the yield of 2,3-BDO and the expression level of <i>bdh1</i>, a key gene in the synthesis of 2,3-BDO (<i>p</i> < 0.05). In addition, a dissolved oxygen concentration of 4.52 mg/L was proven to be the optimal condition for 2,3-BDO production. When the dissolved oxygen content and acetic acid concentration were 4.52 mg/L and 1.0 g/L, respectively, the maximum yield of 2,3-BDO was 3.25 ± 0.03 g/L, which was 66.59% higher than that produced by <i>S. cerevisiae</i> W141 alone. These results provide methodological guidance for the industrial production of 2,3-BDO by <i>S. cerevisiae</i>.
