The usefulness of a QTL marker in breeding for apple powdery mildew resistance | Przydatność markera QTL w hodowli jabłoni odpornej na mączniaka (Podosphaera leucotricha (Ellis et Ev.) Salm.)

English. Scab and powdery mildew, caused by Venturia inaequalis (Cke.) Wint. and Podosphaera leucotricha (Ellis et Ev.) Salm. respectively, are the most important apple diseases. The apple clone U 211 is resistant to scab and highly resistant to powdery mildew in field conditions. The QTL U 7, one of the quantitative trait loci in the clone U 211, is responsible for 48–72% of the phenotypic variation, and its effect is stable over years. It was identified in the progeny Idared × U 211. The aim of this study was to assess effectiveness of U 7 marker for marker-assisted selection (MAS). Powdery mildew resistance was evaluated visually in a seedling population Granny Smith  U 211 for two years in the nursery, and one year in the orchard using a 0–5 scale. Selection based on the SSR marker NZ28f04 allowed to eliminate 33–72% of plants from class 3 (nursery and orchard), and 100% of plants from class 4 (orchard). The efficiency of selection depended on the year. In the progeny Granny Smith × U 211 neither of the individuals grown in the nursery was classified in class 4 or 5 in the 2- year period of investigations. Twenty-two individuals from the progeny were planted in the orchard. In the following year some of them were classified in class 4. Neither of these plants possessed the NZ28f04 allele, associated with the higher level of resistance to powdery mildew.

Polish. Parch i mączniak jabłoni, powodowane przez Venturia inaequalis (Cke.) Wint. i Podosphaera leucotricha (Ellis et Ev.) Salm. należą do najważniejszych chorób jabłoni. Klon U 211 jest odporny na parcha, a w polowych warunkach także na mączniaka jabłoni. QTL U 7, jeden z loci cechy ilościowej (QTL — Quantitative Trait Loci) w klonie U 211 charakteryzuje się trwałym efektem działania i kontroluje duży zakres zmienności cechy (48–72%). Został on zidentyfikowany w potomstwie Idared × U 211. Celem przeprowadzonych badań była ocena przydatności jednego z markerów U 7 do selekcji wspomaganej markerami (MAS — marker-assisted selection). Stopień porażenia mączniakiem był oceniany szacunkowo w skali 0–5 w populacji Granny Smith × U 211 przez 2 lata w szkółce i przez rok w sadzie. Selekcja prowadzona przy użyciu markera mikrosatelitarnego NZ28f04 pozwoliłaby na eliminację 33–72% roślin z klasy 3 (szkółka i sad) oraz 100% roślin z klasy 4 (sad). Skuteczność selekcji była różna w zależności od sezonu wegetacyjnego. W potomstwie Granny Smith × U 211 w ciągu 2 lat obserwacji w szkółce żadnego z osobników nie zakwalifikowano do klas 4 i 5. Część potomstwa (22 rośliny) została posadzona w sadzie i tam niektóre rośliny zostały zaliczone do klasy 4. W żadnej z nich nie stwierdzono obecności allela NZ28f04 związanego z większą odpornością na mączniaka.
