Biochemical content and yield of some cultivars of green bean (<I>Phaseolus vulgaris</I> L.) under Moscow region conditions

Relevance. Increased demand in Russia for common bean (Phaseolus vulgaris L.) varieties of domestic breeding has stimulated the Federal Scientific Vegetable Center of breeding to intensify research on the creation of new asparagus bean varieties of vegetable direction.Materials and methods. Four vegetable bean varieties – Secunda, Lika, Ulyasha, Si Bemol of the laboratory of selection and seed production of vegetable Legume crops of the Federal Scientific Vegetable Center were used as the material for the study. The counts and observations were made according to the Methodology of the State Variety Trial of Agricultural Crops, 1975. The count of productivity was carried out in the technical stage of ripeness by replications. Harvesting was carried out when the biological stage of ripeness was reached. Biochemical studies of plants were carried out in the laboratoryanalytical department of the FSBSI FSVC. The biochemical composition of vegetable bean (green bean) varieties was studied according to the following indicators: determination of the total content of water-soluble antioxidants, ascorbic acid, dry matter, polyphenols, monosaccharides, total sugar content and starch.Results. Evaluation of samples by aggregate factors contributes to the selection of the most promising and high-quality varieties for their introduction into production. As a result of the research of vegetable bean on the main economically valuable features and biochemical composition for the period from 2018 to 2020, the varieties Lika and Ulyasha were selected. They were distinguished by high productivity of beans and seeds. Green bean variety Lika was noted for the highest total content of water-soluble antioxidants, ascorbic acid and the total of sugars content, the green bean variety "lobio" Ulyasha - high content of polyphenols and the total of antioxidants in the alcohol extract. Stable high yield, adaptive potential and high quality of beans, confirmed by the results of biochemical evaluation, allow us to recommend these varieties for consumption by the population as part of the diet and as an alternative to meat and dairy products.

Relevance. Potato are one of the main sources of starch. Potato starch is widely used in human economic activities. The starch content in tubers, including the number of starch granules and their size and shape, are varietal characteristics. For this reason, it is important to identify potato varieties with starch grains suitable for starch production already at the early stages of breeding process.Materials and Methods. The Aim was to study the morphological structure of starch granules in cells of potato tubers belonging to varieties from different maturity groups. Eighteen potato varieties (Solanum tuberosum L.) of various breeding origin and from different maturity groups were used as the research object. The granulometric composition was determined by the method of fluorescent microscopy. The research was conducted using a CLSM 800 confocal laser scanning microscope (Zeiss, Germany) with a laser wavelength of 488 nm.Results. The research allowed us to obtain data on the morphology of variously-shaped starch granules in cells: round, oval, elongated, and irregular. The research established a direct correlation between the number of large starch grains in a cell and the time required for a given potato genotype to mature. Mid-season and medium late potato varieties were observed to have a higher starch content than varieties from the groups with earlier maturity. On average, the difference in the starch content between these groups was 1.97%. The highest number of starch grains was found in 40-70 μm cells and larger. The research identified Solanum tuberosum L. genotypes with large and small starch granules. These genotypes might be recommended for further use in the practical breeding to create high-quality varieties.

Relevance. Potato starch is widely used in the food, textile, perfume and pharmaceutical industries. The starch content and the size of starch granules in potato tubers are varietal characteristics (Solanum tuberosum L.). The knowledge of morphological properties of starch plays a key role in the technologies for the production of consumer and industrial goods.Materials and methods. The studies were carried out in 2020-2021 at the experimental base of Federal State Budget Scientific Institution “Federal Scientific Center of Agricultural Bio-technology of the Far East named after A.K. Chaiki” (Primorsky Krai). The object of the research were 11 varieties of different maturity dates. Morphological structure of starch granules was evaluated by image analysis on Countess II FL automated cell counter.Results. The varieties studied had predominantly rounded and oval pellets regardless of maturity and genetic origin. Differences in starch granule size depending on tuber size and ripeness group were noted. Early maturing varieties had granule size less than 20 microns regardless of tuber fraction. The exception is the variety Queen Anne, in small tubers of this variety starch granules larger than 20 microns is 67.86%. It was noted that the proportion of medium and large grains increased in varieties with a longer growing season. For example, the variety Augustin medium-ripening had a proportion of medium and large grains in the large and small tuber of 47.72 and 41.48%, respectively. The maximum number of grains larger than 20 microns was observed in the mid-late varieties Kazachok (63.77%) and Smak (92.22%).Conclusion. The method using a cell counter and subsequent processing of microphotographs of starch granules is an accessible, economical, simple and effective approach to phenotyping potato varieties and hybrids of Solanum tuberosum L. by physical and chemical parameters of starch. This method can be used for accelerated analysis of a large number of samples on a limited amount of natural material, including in field and farm laboratories.

Potatoes is the most important food crop, one of the main crops grown throughout the world. A way to improve the profitability of the potato industry is processing. The processing production requires special varieties of potatoes. Tests were carried out by the department of vegetable breeding «Agrarian scientific center of republic of Dagestan» in 2017-2018 with the purpose of study the effect of soil-climatic conditions of Dagestan highlands on dry matter and starch content in potato tubers for production of processed potato products. Processing of potatoes is one of the methods to increase profitability of industry. One of the quality indexes of the use of potato varieties for their processing is high level of dry matter and starch. As a result of the research, the following varieties were identified: high-yielding are Impala, Irbit, Zhukovsky Ranniy, Manifesto, Matushka, Nevsky, Primabella, Rosara, Silvana, Spiridon and Udacha; high dry matter content are Alena, Vektor, Dzhokonda, Desiree, Matushka, Nart, Primabella and Rossi; high in starch: Desiree - 23%, Vector and Primabella - 22.2% each. The results of the present investigation revealed that climate of the mountainous areas of Dagestan republic promotes the increase of dry matter and starch content in potato tubers by 3-4%.
