Identification of resistant tomato genotypesusing the gamete technologies

Relevance. High temperatures are among the environmental factors that negatively affect the development of most crops including tomatoes. Thus, important condition for the realization of potential plant productivity is the heat resistance.Methods of the gamete selection in combination with classical approaches can be used to select resistant genotypes and create initial breeding material with resistance to the temperature factor. Materials and methods.The experiments were carried out with intraspecific hybrids F4 of tomato: Elvira x Milenium, Elvira x Tomis, Elvira x Prestij, Mihaela x Milenium, Mihaela x Tomis, Mihaela x Prestij, Jubiliar х Milenium, Jubiliar x Tomis, Jubiliar x Prestij, Milenium x Elvira, Milenium x Mihaela. Plants were grown in the field. Flowers were collected, anthers were separated, pollen was isolated. Pollen was heated for 2 or 4 hours at 43°C in the experimental variants. In the control, pollen was maintained at a temperature of 26°C. Then pollen was sown and germinated on a cultural medium. The viability was determined by the length of pollen tubes.Results.A different reaction of pollen grains to temperature influence was established. The main sources of variability and their contribution to the variability of the pollen and seedling traits were determined. The temperature and genotype mainly determine the variability of the gametophyte and sporophyte. More than half of the hybrids combined high indicators of resistance of both pollen and seedlings, formed a larger number of flowers and better set fruits. Thus, tomato genotypes with good indicators of heat-resistance for applying in breeding have been identified based on a complex of methods for assessing the resistance of hybrids using gametophyte and sporophyte characteristics, as well as results of genetic-statistical analysis.

Relevance. Modern cultivation facilities require serious costs for their construction and subsequent operation. For the construction of 1 hectare of modern greenhouses with the cultivation of vegetable plants in light culture, about 200-250 million rubles are needed. The payback period of greenhouses is 5-6 years or more, which depends on the output from a unit area of greenhouses in value terms. And the latter, in turn, depends on the crop grown, the timing of its cultivation and receipt of products, as well as yield. Of course, the most important influence on the timing of crop cultivation and cost the resulting harvest is provided by the presence of an artificial lighting system greenhouses and its power. So expensive cultivation facilities should be used as long as possible during the year. Plan most the effective use of greenhouses throughout the year is called cultural turnover. Typically, a culture rotation includes one or more cultures. Time, one culture is called turnover. Throughout the year in each greenhouse plants are grown in one or several revolutions. And to remove or to shorten the breaks between turns on the elimination of old plants, preparation of greenhouses for a new turn and their disinfection use interplanting.Methods. The experiment was conducted in one of the greenhouses of the Moscow region in 2018 - 2019 in greenhouses equipped with artificial light of 2 hectares.Results. For various reasons, a number of greenhouse plants refuse Interplanting. Here, from our point of view, there are both objective and subjective reasons. The features of the use of interplanting when growing in light culture will be discussed in this article.

The practice of application of terms characterizing the phenomenon of variability and heterogeneity of seeds, fruits and diasporas is given. The ambiguity and inconsistency of some of them is shown. The methods of systematization and typification of seeds of different quality, based on the features and nature of the manifestation of variability of seed characteristics and causes of their causes, are considered. The main directions of practical use of knowledge about seed polymorphism, including to increase seed productivity and optimize the variability of seed parameters in the growing process, are shown. The principles of evaluation and selection of aligned fractions by morphological features correlated with high sowing and productive qualities in the process of seed refinement in the post-harvest period are presented. The morphological and anatomical causes of defects and injuries in the process of drying and processing of seeds as specific indicators of different quality are considered. The use of dispersion analysis to identify the contribution of hereditary, environmental and matrix factors in the variability of morphological characteristics of seeds, including the length of the embryo, is discussed. The features of the signs of abnormal variability of seeds, which have an obvious and hidden nature of manifestation, are shown. Methods of selection improvement of morfometric parameters, physiological, biochemical and productive properties of seeds as methods of cardinal improvement of quality of seeds are discussed.

Relevance. The creation of heterotic hybrids of watermelon is one of the modern breeding trends for solving the problem of providing the population of the Russian Federation with melon production. The process of creating heterosis hybrids can be accelerated by the assessment of the total combination ability (ОCS) of parental lines.Materials and methods. The studies were carried out in the zone of the Volgograd Trans-Volga region at the Bykovskaya melon selection experimental station, in dry conditions in 2018 and 2019. The object of research was 9 paternal lines and 3 maternal lines of watermelon. The assessment of the general combining ability was carried out according to two indicators: yield and dry matter content.Results. In the course of the research, the obtained F1 watermelon hybrids were assessed for a set of economic characteristics in comparison with the standards: Zenit, Sinchevsky and Barkhan. Based on the results of the work, F1 hybrids were identified according to the length of the growing season, yield, and dry matter content. By selecting valuable genetic sources with a high GCS, it is possible to successfully conduct breeding to improve the economically valuable indicators of F1 watermelon hybrids. According to the results of studies on yield, both in 2018 and in 2019, heterotic hybrids were distinguished, obtained from crossing with the paternal lines Favorit, Stimul, Ikar, the average effect over two years ranged from 3.5-10.6 t/ha. Among the studied maternal lines, the line Chms-4 has a positive effect, the average effect was 1.2 t/ha. In terms of dry matter content, a positive effect was found in the paternal lines Stimul – 1.8%, Medunok – 1.6%, Favorit – 0.8%, and in the maternal line Chms – 41.8%Conclusions. Based on the results of the analysis of the combining ability in terms of yield and dry matter content, we identified two promising paternal lines for creating heterotic watermelon hybrids: Favorite, Stimul. Also, one maternal line Chms-4 was distinguished. When using these lines in heterotic breeding of watermelon, in most crossing combinations, heterosis will be manifested in one or two characters.

Relevance. A large reserve to increase the production of tomatoes for different periods of cultivation and purposes of use are highly productive new varieties and heterotic hybrids. In this regard, one of the important conditions now is the maximum compliance of newly created varieties and hybrids with modern technologies that allow obtaining competitive products. In addition to the main indicators (high yield, early maturity, favorable maturation), varietal features such as transportability of fruits,high taste appeal (especially salad-type hybrids), appearance, resistance to major diseases and manufacturability are also important. In recent years, tomato and other vegetable crops have grown mainly hybrids of the first generation F1 and line (the result of targeted selection), less often populations. For more than a decade, only hybrid varieties have been grown in industrial greenhouse vegetable production.Methods. The article presents the results of 7-year research on the yield, its structure and quality of fruits of indeterminate tomato hybrids. Research on the yield and quality of fruits of indeterminate tomato hybrids was carried out in 2011-2017, in JSC "Zavyalovsky Greenhouse plant" of the Udmurt Republic.Results. Hybrid F1 tomato Ismail, F1 Macarena, F1 Mace, F1 Geronimo and F1 Starbuck were distinguished by high productivity. This indicator was respectively: 34.5 kg / m2; 33.1 kg / m2; 32.3 kg / m2; 32.0 kg / m2 and 31.2 kg / m2. Tomatoes F1 Yakimanka, F1 Foronti, F1 Starbuck, F1 Taganka and F1 Mace stood out for large-fruited. More than 30 mg / 100 g was contained in F1 Izmail F1 Ochakov, F1 Mariachi, F1 Kokhava vitamin C fruits. The nitrate content in tomato fruits was not high for years of research, it was within the MPC.

Relevance. The use of molecular methods allows reliable and fast determination of the resistance of genotypes (varieties) to pathogens, thereby reducing possible product losses and, at the same time, maintaining its environmental safety. It is very important in conditions of increasing demand for high-quality agricultural production. Aim: Using molecular diagnosis of ʹCandidatus Phytoplasma solaniʹ to compare the degree of infection in some Moldavian tomato varieties at different stages of plant development.Material and methods. The molecular analysis (nested-PCR) of plants of the four Moldavian tomato varieties (Elvira, Cerasus, Mary Gratefully, Desteptarea) created at the Institute of Genetics, Physiology and Plant Protection, and the wild formSolanum habrochaites, was carried out for the presence of the phytopathogen ʹCa. P. solaniʹ. Researches were made during two growing seasons. Results. The distribution of infection between the studied varieties was different in the process of plants development. The spread of infection in the tomato field was recorded under the climatic conditions of two growing seasons: the season of 2018, which was hot but with normal rains in the middle of summer, and the season of 2019, in conditions of a very hot and dry summer. During both seasons, Cerasus variety manifested the highest resistance to ʹCa. P. solaniʹ infection. A little more than half of plants of this variety were affected by stolbur only at the end of the growing season, after harvesting most of the crop. Varieties Elvira and Desteptarea had similar levels of infection of plants with phytoplasma during two years of research. These varieties manifested a higher sensitivity to phytoplasma infection compared with Cerasus. Mary Gratefully was the genotype with the highest dependence of the sensitivity toʹCa. P. solaniʹ infection from the climatic conditions of the growing season. Plants of the wild form Solanum habrochaites demonstrated complete immunity to phytoplasma infection during the growing season. Conclusion.The Cerasus variety, as well as the wild form Solanum habrochaites, can be recommended for including in breeding programs for the creating tomato varieties or hybrids resistant to phytoplasma. Thus, molecular diagnosis may be a useful tool for the breeding resistant genotypes.

Relevance. The most complex and common physiological disease on the cabbage is a tip burn, associated with impaired Ca2 + transport inside the plant. Of all the ways to control this physiological disease, the most effective is the cultivation of tolerant hybrids. Material and methods.The aim of the research was to evaluate the mid-late and late ripening F1 hybrids of white cabbage for a tip burn and to determine the degree of hybrids responsiveness to calcium deficiency depending on weather conditions. The studies were carried out at breeding and experimental plot of the vegetable growing department at All-Russian Rice Research Institute in 2017-2019 on drip irrigation. Material - hybrid combinations of ARRRI breeding.Results. The manifestation of a physiological disturbance called “burn of the top of the inner leaves of the cabbage head” or “tip burn” on individual hybrids was observed during all years of cultivation, but to a different degree; The greatest damage to the heads was recorded in the years (2017 and 2019) with high productivity, which is due to the intensive assimilation ability against the background of warm sunny weather. In 2019, the manifestation of burns was noted in 25% of hybrids, which allowed them to be divided into 4 groups, depending on the degree of damage and the spread of the disease.It was established that hybrids with overmature standing for 2.5-3 weeks and also hybrids that were promptly removed at the time of mass technical ripeness had a high degree of damage. The lines that are most often found in hybrid combinations with signs of burns (269-824, Agr1321, 269Jas13, Byum112, Yas25, etc.) were identified, but it is rather difficult to assume the nature of inheritance, and, therefore, to develop the principle of pair selection when developing relatively stable hybrids also not yet possible.

Relevance. The breeding process in identifying new species and flexible capabilities allows the use of genetic sources for the most important economically valuable traits. Market requirements to expand the assortment of cucumber are determined both directly and new approaches to the study of the gene pool and its use. In turn, the domestic market is not always predictable and logical. Thus, the issue of a comprehensive study of the sorting of a gunshot and smooth type of fruit for growing in film unheated greenhouses in order to isolate genetically valuable material for breeding work remains relevant.Materials and methods. The studies were carried out in 2015-2018 on the basis of a pumpkin crop laboratory of the Research Institute of Vegetable Production (NIISOK), of the Gavrish breeding station in Moscow, Krymsk (Krasnodar Territory), the Gavrish Jordanian Experimental Station. Research material – 34 different types of origin from the NIISOK collection. The work used both traditional methods of assessment and modern methods of molecular marking. The purpose of the work is the selection of breeding valuable source material for creating parthenocarpic lines of a cucumber with a smooth type of fruit, a complex of economically significant features, and resistance to disease.Results. The results of variety testing of 20 hybrids of foreign selection on the territory of the Krasnodar Territory (Krymsk) and Jordan are presented. A comparative assessment was made of the inter-varietal and ecological variability of productivity and yield of the studied samples of cucumber culture. Based on the results of studying the modern assortment of hybrids in different regions (Krymsk, Jordan), the optimal parameters of the parthenocarpic hybrid model of a cucumber with a smooth type of fruit adapted to specific growing conditions are determined.

Relevance.Tomato hybrids of the "beef" type are a commodity group characterized by fruits whose mass exceeds 220-240 g. The fruits of this group are distinguished by an attractive aligned shape and color, large size and are in constant demand among consumers. In the assortment of supermarkets and markets, the share of "beef" tomatoes is 10-20% of the total volume. Methods.The results of studying the characteristics of tomato hybrids allowed us to identify the most valuable characteristics for selection in the selection of "beef" tomatoes. Traditionally, we select donor lines for the signs "fruit weight from 200 g and above", "high yield", "disease resistance". For hybrids of professional use, we also evaluate such characteristics as" fruit density", "high uniformity", "ability to form fruits with high uniformity and marketability throughout the growing season". For hybrids and varieties for the hobby market, the most important characteristics are "High taste qualities", "aroma", "high dry matter content". Results. As a result of selections based on these characteristics, such hybrids as Rumyani shar F1, Korallovy rif F1 (which are in demand not only in our country, but also in other countries), Katarina F1 were created.

Relevance. The study of the resistance of garden cultures to unfavorable abiotic factors of the winter period is a priority direction of scientific studies, because the low winter hardiness can annul advantages the varieties on other signs. Concerning in the middle zone of Russia, studies on the resistance of cherry to frost during the thaw period remain relevant.Methods. The studies were carried out in the laboratory of physiology of fruit plant resistance at VNI-ISPKin2017-2018. The object of research was the Turgenevka cherry variety on new clonal root-stock of the Institute breeding were studied. Control - Turgenevka on the Rubin stock. For artificial freezing in early December the material was prepared for III and IV components of frost hardiness.Results. As a result of artificial freezing, the Turgenevka cherry variety was characterized by frost resistance of vegetative buds and tissues of annual shoots during a three-day winter thaw of + 2°C with a decrease in temperature of -25°C (III component of frost resistance). Inthiscase, thehighest frost resistance of the generative buds of the Turgenevka variety on rootstock 74340, 82987 is shown. After a three-day thaw of +2°C and repeated hardening with a subsequent decrease in temperature to -30°C in March (IV frost resistance component), the Turgenevka variety on the rootstock 74332, 74363, 82987 was characterized by frost resistance of vegetative buds, bark and wood of annualshoots. Moreover, in the studied scion-rootstock combinations, low frost resistance of the generative kidneys was revealed. Only in the studied variety on the rootstock 82987 was the largest percentage of living flower primordia revealed. The conducted studies revealed the Turgenevka variety in a rootstock of 82987 with the greatest potential for frost resistance during the winter thaws.
