Berndtson W.E. | Igboeli G.
Data from 34 yearling Hereford or Angus bulls were used to investigate relationships of testicular size, quantitative rates of sperm production, Sertoli cell numbers, numbers of germ cells supported per Sertoli cell, and the efficiency of spermatogenesis to daily sperm output and seminal quality. Two ejaculates were collected by electroejaculation from each bull on each of 2 days/week throughout the study. The percentage of progressively motile sperm and the percentage of morphologically normal sperm were determined from aliquots of fresh semen. Additional aliquots of semen were frozen in glass ampules or plastic straws and subsequently evaluated for postthaw motility and percentage of sperm with intact acrosomes. Sertoli cell numbers, the numbers of germcells per Sertoli cell, and the efficiency of spermatogenesis were unrelated to the quality of fresh or frozen semen (P greater than 0.05). In first ejaculates, the numbers of sperm and motile sperm were related (P less than 0.05) to testicular parenchymal weight (r = 0.38, and 0.50), daily sperm production (r = 0.45 and 0.53), and spermatids per gram of testicular parenchyma (r = 0.35 and 0.34). Testicular parenchymal weight and daily sperm production also were related to daily sperm output and to the average daily motile sperm output of these bulls (P less than 0.05), but could account for less than 25% of the variability in these end points among bulls.

The aim of this study was to present two outbreaks of bovine abortion due to Leptospira infection in cattle herds located in the northern part of Sicily (Italy). The animals were positive for Leptospira interrogans serogroup Sejroe serovar Hardjo in a microscopic agglutination test (MAT). A total of 23 Charolaise cows (farm A) and 75 Limousine bulls and Cinisara and Modicana cows (farm B) were enrolled in this study. The blood samples were collected from all subjects at the following time points: before a cycle of intramuscular treatment with oxytetracycline dihydrate (T0), after 5–6 weeks from the treatment (T1), and every 10 weeks until seronegativisation (T2 in Farm A and T3 in Farm B). A serological test (MAT) was used for the diagnosis of leptospirosis. Two samples from farm A (2/23) and 29 samples from farm B (29/75) were positive to Leptospira interrogans, serogroup Sejroe, serovar Hardjo in the MAT. Leptospira spp. DNA was detected by real-time PCR in the urine sample of one positive cow on farm A, and in placenta and brain samples belonging to one aborted foetus on farm B. It is important to use serological and molecular diagnostic techniques complementarily to identify infected individuals.

Introduction: Glycolic changes which occur post-mortem have an impact on the physical and sensory features of beef, which in turn determine the successive processes and influence such beef quality traits as colour, tenderness, and cooling loss. The aim of this study was evaluation of the post-mortem changes in bovine meat during aging, quantitative analysis of glycogen and lactic acid, as well as examination of their impact on technological and sensory quality of selected muscles from Holstein-Friesian × Limousin breed carcasses.Material and Methods: The study included three muscles of different metabolic qualities and sarcomere length: m. semitendinosus, m. longissimus dorsi, and m. psoas major, collected from nine bull carcasses aged 24 ±2 months. Results: Significant correlations were found between the volume of cooling loss on individual days of aging and the pH value of muscle tissue, lactic acid and glycogen content, as well as beef lightness. However, no significant dependency between the volume of glycogen and the intensity of red and yellow colours was detected. Conclusion: The colorimetric analysis of glycogen and lactic acid can be an effective tool in predicting the quality of beef.

Introduction: This article presents the analysis of the correlation between the category and health status of calves and the results of their rearing and levels of selected blood parameters.Material and Methods: The study included 105 Polish Holstein-Friesian and beef (Limousine, Charolaise and Hereford) crossbred calves. Young bulls were purchased at the age of two to four weeks. The animals underwent quarantine, were dehorned, and 46 young bulls were castrated. The germ horns were removed by burning out. Castration was carried out with a bloodless method using a rubber band. The calves were kept in groups and fed a milk replacer administered via teats from automated milk-feeding stations. After the period of milk feeding, the calves were fed grass silage ad libitum and a concentrate at 2.5 kg/animal/day. The calves were weighed every two weeks. Blood for analyses was sampled at 43 d of age. Results: After the rearing period finished at the age of six months, young bulls and steers had similar body weights (176.17 and 176.55 kg) and approximate average daily weight gains from birth (0.756 and 0.767 g/day). The healthy calves at six months of age weighed 180.47 kg, whereas the animals which at least once suffered from some diseases during rearing were lighter by approx. 30 kg (P ≤ 0.01). A statistically significant (P ≤ 0.01) difference was found for the count of red blood cells and white blood cells. In comparison with healthy individuals, the diseased animals had less RBC (8.33 and 9.42 10¹²/L respectively) and more WBC (27.03 and 12.26 10⁹/L respectively). Conclusion: Castration of young bulls did not have any impact on the results of rearing and health status of the calves. The magnitude of the analysed parameters depended on the health status of the calves. Thus RBC and WBC parameters may be used to predict the health status of calves during rearing.

Introduction: The present study is a comprehensive overview of the natural occurrence of 17β-oestradiol and testosterone in serum of cattle in Poland. Material and Methods: The serum samples (n = 826) were collected from cattle within five years. The samples were examined for the presence of oestradiol and testosterone using ELISA or gas chromatography with mass spectrometry. Results: In 98 samples (24%) 17β-oestradiol was detected above decision limits of applied methods, including five samples over the recommended concentration of 0.1 μg L⁻¹. Of the serum samples taken from cows (≤18 months of age), 95 and 99 percentiles of the animals had 17β-oestradiol concentration below 0.027 and 0.086 μg L⁻¹ and of samples from cows over 18 months of age - below 0.059 and 0.125 μg L⁻¹ respectively. Calculated values for bulls (≤18 months of age) were 0.025 and 0.034 μg L⁻¹ and for the animals older than 18 months of age - 0.035 and 0.041 μg L⁻¹. The natural presence of testosterone was detected in 201 serum samples (48.7%). According to the obtained data, 95% and 99% of cows (≤18 months of age) serum samples had testosterone concentration below 0.05 and 0.23 μg L⁻¹ and the animals over 18 months of age - 0.30 and 0.49 μg L⁻¹, respectively. For bulls these values did not depend on the age of the animals and were in the ranges of 5 - 6.3 μg L⁻¹ (95%) and 11.4 - 12.1 μg L⁻¹ (99%). Conclusion: Our study showed that the threshold values for these hormones in plasma of cattle designated years ago are correct, but they need to be supplemented for animals older than 18 months.

Objective-To compare the recovery rates of Campylobacter fetus subsp venerealis (Cfv) from preputial scrapings of infected bulls with passive filtration on selective medium versus nonselective medium, with and without transport medium. Samples-217 preputial scrapings from 12 bulls (4 naturally and 8 artificially infected with Cfv). Procedures-Preputial scrapings were collected in 2 mL of PBS solution and bacteriologically cultured directly on Skirrow medium or passively filtered through 0.65-μm filters onto blood agar, with or without 24 hour preincubation in modified Weybridge transport enrichment medium (TEM). After 72 hours, plates were examined for Cfv and bacterial and fungal contamination or overgrowth. Results-Passive filtration of fresh preputial scrapings onto blood agar yielded significantly higher recovery rates of Cfv (86%) than direct plating on Skirrow medium (32%), whereas recovery from TEM was poor for both media (35% and 40%, respectively). Skirrow cultures without TEM were significantly more likely to have fungal contamination than were cultures performed with any other technique, and fungal contamination was virtually eliminated by passive filtration onto blood agar. Bacterial contamination by Pseudomonas spp was significantly more common with Skirrow medium versus passive filtration on blood agar, regardless of TEM use. Conclusions and Clinical Relevance-The use of transport medium and the choice of culture medium had significant effects on Cfv recovery and culture contamination rates from clinical samples. Both factors should be considered when animals are tested for this pathogen.
Show more [+] Less [-]Sample collection factors affect the sensitivity of the diagnostic test for Tritrichomonas foetus in bulls
2003
Parker, Sarah | Campbell, John | Ribble, Carl | Gajadhar, Alvin
The current diagnostic test for Tritrichomonas foetus involves the culture of collected preputial or vaginal samples. In an earlier study, which evaluated sampling tools for use with bulls, it was observed that the sensitivity of the diagnostic test was higher for 2nd samples collected from the right side of the prepuce than it was for samples collected 1st from the left side. The study described in this paper was conducted to evaluate which of these factors was responsible for the effect on diagnostic sensitivity. Twenty-nine bulls infected with T. foetus were repeatedly sampled in a 2-factor cross-over design. Samples taken from the right side of the prepuce were 4 times as likely to be positive as samples taken from the left side (P = 0.03). Other factors did not have a significant effect on the outcome of the diagnostic test. Unexpected factors may affect the sensitivity of the diagnostic test for T. foetus.

At initiation of a 140-day postweaning weight gain test, Angus bulls were assigned in equal numbers (n = 5) to 1 of 3 treatment groups to determine effects of implantation with zeranol, an estrogenic growth promotant, on selected reproductive characteristics. The bulls, whose age (mean +/- SD) was 233 +/- 20 days at initiation of the test (day 0), were implanted with 36 mg of zeranol on day 0, on days 0 and 60, or were not implanted. At day 140, scrotal circumference and testicular consistency were unaffected by zeranol implantation. Zeranol implantation did not affect the morphologic characteristics of semen samples collected by electroejaculation on day 139. There was no effect of zeranol treatment on paired weights of testes, epididymides, or vesicular glands from bulls at slaughter 47 to 68 days after day 140. Microscopic lesions associated with estrogenic exposure were not observed in accessory sex glands or epididymides of any bull. Histopathologic changes in the seminiferous epithelium were not induced by zeranol treatment. Implantation with zeranol did not affect body weight or hip weight at day 140 or carcass weight at slaughter. Plasma concentration of luteum hormone was increased (P = 0.04), whereas testosterone concentration tended to be less (P = 0.08) in both groups of zeranol-implanted bulls after administration of gonadotropin-releasing hormone on day 140.

1990
Williams, R.J. | Boudinot, F.D. | Smith, J.A. | Knight, A.P.
Six mature Holstein bulls were given an 8-day course of phenylbutazone (PBZ) orally (loading dose, 12 mg of PBZ/kg of body weight and 7 maintenance doses of 6 mg of PBZ/kg, q 24 h). Plasma concentration-vs-time data were analyzed, using nonlinear regression modeling. The harmonic mean +/- pseudo-SD of the biologic half-life of PBZ was 61.8 +/- 12.8 hours. The arithmetic mean +/- SEM of the total body clearance and apparent volume of distribution were 0.0021 +/- 0.0001 L/h/kg and 0.201 +/- 0.009 L/kg, respectively. The predicted mean minimal plasma concentration of PBZ with this dosage regimen was 75.06 +/- 4.05 microgram/ml. The predicted minimal plasma drug concentration was compared with the observed minimal plasma drug concentration in another group of bulls treated with PBZ for at least 60 days. Sixteen mature Holstein bulls were given approximately 6 mg of PBZ/kg, PO, daily for various musculoskeletal disorders. The mean observed minimal plasma concentration of PBZ in the 16 bulls was 76.10 +/- 2.04 microgram/ml, whereas the mean predicted minimal plasma concentration was 74.69 +/- 3.10 microgram/ml. Dosages of 4 to 6 mg of PBZ/kg, q 24 h, or 10 to 14 mg of PBZ/kg, q 48 h, provided therapeutic plasma concentrations of PBZ with minimal steady-state concentrations between 50 and 70 microgram/ml.
