Evaluation of the Effects of Ascorbic Acid on Doxorubicin-Induced Hepatotoxicity in Mice

BACKGROUND: Doxorubicin is one of the most widely used anticancer chemotherapeutic agents in small animal practice. The use of doxorubicin can cause cardiotoxicity, hepatotoxicity, neurotoxicity, and nephrotoxicity. OBJECTIVES: This study was carried out to evaluate the effects of ascorbic acid on doxorubicin hepatotoxicity in mice. METHODS: Twenty-four Balb/c mice were randomly divided into four groups. Group one received normal saline, group two received 100 mg/kg ascorbic acid, group three received 8 mg/kg doxorubicin and group four received ascorbic acid and doxorubicin intraperitoneally, with the same doses of groups 2 and 3. Twenty-one days after injection, the mice were euthanized. The activities of ALP, ALT, AST enzymes and total bilirubin levels in the serum samples were measured. Liver samples were evaluated histopathologically. RESULTS: The activities of ALP, ALT, AST, and total bilirubin levels and histopathologic scores of hepatotoxicity were significantly lower in the group that received ascorbic acid + doxorubicin in comparison to those of the doxorubicin group. CONCLUSIONS: Ascorbic acid may be useful in the prevention of doxorubicin hepatotoxicity in mice. Further studies are recommended for evaluation of the use of ascorbic acid in small animals.

Nowadays, with the development of new technologies, improved and progressed methods have been taken to diagnose, treat and prevent cancers. Pathologic study and some molecular methods have been helpful in diagnosing and predicting cancer but these methods are not enough in many cases. Omics technology investigates many parts of cells such as genes, proteins, transcripts, and metabolites simultaneously. This procedure provides a more real and general feature of cellular processes, especially in cancer cells. In human, Omics technology is widely used to diagnose and treat various cancers and predict prognosis of tumors and survival of patients. In parallel to the studies of cancers in human, similar investigations were conducted in the canine cancers. Regarding the importance of Omics method in oncology, we described various Omics techniques including genomics, transcriptomics, and proteoimcs. In addition, corresponding studies carried out in different canine cancers were summarized in the next step.

Cancer is a growing threat to human and animal health, The use of synthetic chemotherapy in the treatment of cancer is short because of lack of appropriate safety due to their drastic side effects. So, this study evaluated the ameliorative effect of Chrysanthemum on genotoxicity as well as renal and hepatic toxicity of Capecitabine (xeloda), as a model of anticancer drug. Thirty six male albino rat were divided into 6 groups. Serum samples were taken at 21 and 45 days and bone marrow samples at 45 days. Comet assay, biochemical tests for liver and kidney functions were done also serum oxidative biomarker were measured. The results of the present study revealed that capecitabine (xeloda) caused harmful genotoxic effect, biochemical changes in oxidative biomarkers, liver and kidney function, however the use of capecitabine (xeloda) in combination with Chrysanthemum ameliorated these changes especially with high dose. In conclusion, capecitabine adverse effects could be successfully alleviated through the co-administration of the natural antioxidant Chrysanthemum.

The benefits of taking these drugs generally outweigh the risks, but Potential adverse effects should be taken into consideration. This review will present updated information on the negative reactions associated with PPIs and identify areas for further research. The research for this paper was gathered from a variety of sources, including Google Scholar, Science Direct, PubMed and other journals published between 1998 and 2022.
