PCR-based detection of Schistosoma bovis in cattle in Maiduguri Metropolis and Jere Local Government Areas in Borno State, Nigeria

Objective: To determine the prevalence of Schistosoma infection in cattle in Maiduguri Metropolis (MMC) and Jere Local Government Areas (LGAs) of Borno State, Nigeria.Materials and Methods: Blood samples (n=200) were collected from cattle consisting of one hundred (100) each from five (5) ward levels each of MMC and Jere LGAs. DNA samples were extracted from the serum samples, analysed and quantified using a Nano-drop machine. The extracted DNA were subjected to polymerase chain reaction (PCR).Results: The overall prevalence of Schistosoma infection was 2% (n=200). Jere LGA had 3% (n=100) while MMC had 1% (n=100). There was no statistical significant association in prevalence rate in the two LGAs studied (P=0.621) (P>0.05). At the ward levels, Custom Area in Jere LGA had 15%, Jiddari ward in MMC had 5%, and the remaining ward levels had no cases. Of the 103 female and 97 male cattle screened, the prevalence in female was 1(0.97%) and 3(3.09%) in the male. Of the 177 serum samples from above 1year (adult) examined, 4 (2.26%) were positive and none in the young. There was no statistical significant association in prevalence rate among ward levels, sex groups and age groups in the study areas (P=0.621) (P>0.05), (P=0.356) (P>0.05) and (P=1.000) (P>0.05) respectively. Of the eight (8) breeds screened, Kuri had 2.7%, Sokoto Gudali (1.82%), Abore (2%), Red Bororo (2.63%), White Fulani, Porland, Mbala and Wafara recorded no cases. The difference in prevalence rates among the breeds based on the trend of occurrence of Schistosoma infection were not significantly associated statistically (P=1.000) (P>0.05). Conclusion: There is a prevalence of Schistosoma infection in cattle in the two LGAs of Borno state. It is recommended that a system be established to maintain preventive and control programs. During growth, central venous, right ventricular, pulmonary arterial, Pulmonary capillary wedge, and systemic arterial pressures, heart rate, and cardiac Output were repeatedly measured in 41 Friesian calves, considered as having conventional muscular conformation, and in 19 Belgian White and Blue double-muscled calves. A total of 123 and 70 recordings were collected in conventional and double-muscled calves, respectively. These circulatory indices were calculated: stroke volume, cardiac and stroke indices, pulmonary and systemic pulse pressures, pulmonary and systemic vascular resistance indices, and right and left ventricular work indices. Results indicated that systemic arterial and pulse pressures, as well as cardiac output, stroke volume, cardiac and stroke indices, and right and left ventricular work indices were significantly (P less than or equal to 0.05 to 0.001) lower but, in contrast, pulmonary and systemic vascular resistance indices were significantly (P less than or equal to 0.001) higher in double-muscled than in conventional calves. Right-sided vascular pressures and heart rate were similar in the 2 groups. These results indicated that global cardiac performance may be considerably poorer in double-muscled calves. Diminished cardiac performance of double-muscled calves appears to be related neither to relative bradycardia nor to reduced ventricular preload. The potential role of increased ventricular afterload or of reduced myocardial contractility in double-muscled cattle should be determined by direct measurements.

Influence of age, breed, and stage of pregnancy on hepatic ultrasonographic findings of cows was determined. In addition, the relation between body weight, height at the withers, milk production, and the measurements determined via ultrasonography was investigated. The liver of 186 cows was examined ultrasonographically. The cows comprised Swiss Braunvieh, Simmental, and Holstein breeds, and age ranged from 2.5 to 11.5 years. The ultrasonographic findings of the liver, gallbladder, caudal vena cava, and portal vein were described, and the position, size, thickness, and distal angle of the liver were determined. In addition, the position and diameter of the caudal vena cava and portal vein were determined. There was no significant difference between any of the variables determined and breed or age. Therefore, measurements for the 3 breeds and for the various ages were summarized into 1 group. There were significant correlations between body weight, milk production, and size and thickness of the liver. In 3 pregnant cows, the liver was examined ultrasonographically 8 times during the course of pregnancy. Positive correlation was detected between stage of pregnancy and diameter of the caudal vena cava. There was a negative correlation between stage of pregnancy and diameter of the portal vein. In 23 cows, the ultrasonographically determined measurements of the liver were compared with those determined at slaughter. Weight of the liver correlated well to thickness of the liver determined via ultrasonography.

We investigated the biochemical composition of blood from Holstein cows, native breed (criollas), and cows descended from fighting bulls (Vacas de lidia) raised at an altitude of 3,000 m (moderately high altitude, MHA), and compared the results with those from Holsteins and cows of similar genetic ancestry as the criollas (scrub cows), both raised at sea level (SL), to determine blood biochemical values characteristic of adaptation to high altitude. Only potassium and calcium concentrations were similar among groups. Glucose concentration was lower in MHA cows, with the exception of Vacas de lidia. Serum bicarbonate concentration was lower in MHA cows; this finding can be explained by hyperventilation in the hypoxic environment. Serum magnesium concentration was lower in SL and MHA Holsteins than in other groups. Serum phosphate concentration was lower in scrub cows, MHA Holsteins, and criollas than in other groups. Cholesterol concentrations were lower in SL Holsteins, whereas triglycerides were higher in scrub cows and MHA Vacas de lidia. Concentration of high-density lipoprotein was significantly greater in Vacas de lidia and less in MHA criollas than in the other groups. Uric acid and total protein were higher in MHA groups. Using radioimmunoassay for human proteins, thyroxine-binding globulin was undetectable. Total and free thyroxine and free triiodothyronine were higher in scrub cows, followed by Vacas de lidia; lower values were detected in SL and MHA Holsteins and MHA criollas.

Gene frequencies of RBC antigens were determined in Holsteins and Colombian (criollas) cattle living at 3,000 m, and in cattle descended from fighting bulls (Vacas de lidia) living at 2,500 m. These frequencies were compared with those of Holsteins, cattle native to Florida (scrub cattle), longhorns, and native cattle from Brazil (caracu cattle) living at sea level. The criollas, Vacas de lidia, scrub cows, longhorns, and caracu are descendants of original Iberian stock introduced to the Americas. We found that despite common ancestry (scrub cattle, longhorns, criollas, and caracu), genetic differences may have been derived through years of demographic isolation. The most remarkable blood-group differences were found in the high prevalence of the B system phenogroup (heritable group of antigenic factors) BQA'G'34 in the Vacas de lidia, and of the S system phenogroup U1H' in these cattle and in caracu. Furthermore, the gene frequencies differed in the Holsteins maintained at moderately high altitude (descended from Holsteins kept at sea level), and may have been reflective of the need to adapt to moderately high altitude and chronic hypoxemic conditions. Blood group polymorphism was found in all groups of cattle, although it was reduced in the Vacas de lidia, possibly because their breeding has been carefully controlled and they appear to be highly inbred.

Six morphometric traits of chest girth (CG), withers height (WH), body length (BL), chest depth (CD), hip height (HH) and rump width (RW) were performed in this study. Objective: This study aimed to evaluate rumen fermentation parameters influenced by both the graz¬ing system and breed. Materials and Methods: A 2 × 2 factorial design was employed, involving 40 cows with matched age, parity, and physiological status. The cows were evenly divided between Bonsmara and Nguni breeds, as well as communal and commercial grazing systems. Rumen fluid samples were collected and analyzed for parameters including ammonia-nitrogen (NH3-N), pH, temperature, and volatile fatty acids (VFAs). Results: Nguni cows exhibited significantly higher ruminal NH3-N levels (p < 0.05) compared to Bonsmara, ranging from 69.05 to 96.78 mg/l. Commercial grazing demonstrated significantly higher NH3-N concentrations (p < 0.05) than communal grazing. Ruminal pH, temperature, total VFAs, and specific VFAs (Iso-butyrate, valeric, and iso-valeric) did not show significant differences (p > 0.05). However, total VFAs were slightly lower in communal grazing (78.87 mmol/l) than in commercial grazing (89.80 mmol/l). Acetate, propionate, butyrate, and the acetate to propionate ratio did not display significant differences (p > 0.05) between breeds but varied between grazing systems. Communal systems had higher acetate and acetate to propionate ratio (p < 0.05), while commercial systems showed higher propionate and butyrate levels (p < 0.05). Conclusion: Grazing conditions significantly influenced rumen fermentation parameters, irrespec¬tive of breed. Further research is necessary to explore the relationship between forage conditions, diversity, and rumen fermentation within different grazing systems. [J Adv Vet Anim Res 2024; 11(2.000): 349-355]
