Diagnostic test for bovine mastitis by the determination of ATP based on firefly bioluminescence

This study was carried out to diagnostic test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The infection rate of bovine mastitis investigated with 521 cows in 47 dairy farms were found to be 3.6 % of clinical form and 44.1 % of subclinical form according to the degree of infection. The light yield produced in firefly bioluminescence system was proportional to the concentration of ATP giving stright line within the range of 100PM-luM. When the number of somatic cell in milk was determined by the ATP assay and compared with three conventional methods such Fossomatic. California mastatic test (CMT), and rolling ball viscometer (RBV), it was shown that r= 0.92 for Fossomatic, 0.63 for CMT and 0.7 for RBV. The microorganisms causing mastitis were isolated Staphylococcus sp. (53.3 %), Streptococcus sp. (17.9 %), Micrococcus sp. (13.5 %), Gram negative bacilli (6.3 %), Gram positive bacilli (5.5 %) and Yeast-like fungi (5.4 %). The endogeneous ATP levels of bacteria in a raw milk determined by the firefly bioluminescence system and compared with the results of the conventional methods. The correlation was 0.88 for raw milk

Normal swine brains at 1 to 70 days after birth were used to investigate the presence and morphology of the subependymal layer (SL) in the ventricle walls. The brain samples were taken from 27 pigs of 4 swine breeds. SLs were observed on the walls of the lateral ventricle (LV) but none were observed on the walls of the 3th and 4th ventricles. SLs of the LV walls were composed of mainly 3-to 10-cell layers in thickness. The thinnest region of SLs was composed of only 1-to 2-cell thick on the dorsal and ventral walls, and the thickest region was composed of 250-to 300-cell thick on extension region of the SLs into the angle between the corpus callosum and caudate nucleus. SLs were extensively thick in young age but gradually decreased in size and cell number with age after 20-day age. SL layers were composed of mainly oligodendrocytes, astrocytes and immature cells of them. Morphological differences of SL in different breeds of pigs were not observed.

Endocrine cells were observed in the gastrointestinal tract of the squirrels by light microscopy using Grimelius and Masson-Hamperl silver impregnation, and in the stomach and duodenum by electron microscopy. Light microscopically, argyrophil cell decreased gradually in number in the pyloric region and ileum, stomach, duodenum and jejunum and rectum in the order listed. Argentaffin cells were moderately distributed in the stomach, pyloric region and duodenum, while absent in the jejunum, ileum and rectum
