BACKGROUND: Dietary nucleotide has been shown to improve growth indices, nutritional function and higher vertebrates in fish. OBJECTIVES: We investigated the effect of different levels of dietary nucleotide on growth parameters, whole-body composition and fluctuations of cortisol and glucose due to acute stress in Snow trout (Schizothorax zarudnyi). METHODS: One-hundred-twenty Snow trout fingerlings with average weight 7.58±0.05g, were fed with four levels of dietary nucleotides including 0 (control), 0.15, 0.25, and 0.5 percent for 8 weeks. Ten fingerlings were stocked into each 300l tank with triplicate in four treatments and fed three times daily at 3-5% body weight rate. At the end of feeding trial, weight gain, Weight gain rate, feed efficiency, Food efficiency ratio and survival were computed. Also, the numbers of fish that were exposed to acute stress (temperature and pH). RESULTS: The results showed that none of the nucleotide-supplemented diets had any significant effect on growth indices (p>0.05). None of the dietary treatments resulted in any significant changes to dry matter, ash and protein. The only significant difference was observed in the fat (p>0.05). The fish fed 0.25% nucleotide showed higher whole lipid compared to other dietary treatments. Stress test results showed no significant difference at the beginning of the experiment, whereas after, acute stress cortisol levels were significantly higher in trials. The lowest response was observed in trial with 0.50% nucleotide and the highest was in control groups. Glucose levels were not significantly different, neither before nor after acute stress. CONCLUSIONS: The nucleotide composition used in the present study failed to provide significant effects on growth of Snow trout juveniles but could improve response to acute stress.

This study aimed to determine the profile of immunoglobulins and cortisol concentrations in serum around the periparturient period in sows suffering from postpartum dysgalactia syndrome (PDS) and in healthy sows.

The study measured the hormonal and protein markers of acute stress, those of oxidative stress and total antioxidant capacity (TAC) in swine oral fluid, determined which of these parameters would be the most appropriate for future livestock welfare assessment and established the time when the samples should be taken.

A glutamina e o aminoácido livre mais abundante no organismo de mamíferos, muito importante para a proliferação celular, metabolismo da amônia e para produção de energia na musculatura esquelética, principalmente durante o exercício aeróbico. O objetivo do presente trabalho foi investigar se os níveis séricos de glutamina, amônia e cortisol, em cavalos de enduro, podem ser modelos de estudo da dinâmica deste importante aminoácido durante o exercício, pois estes animais são submetidos a esforço aeróbico prolongado e intenso. Foram utilizados 33 cavalos de enduro que competiram nas categorias de 80 km (n = 13), 120 km (n = 14) e 160 km (n = 5) em quatro provas. Não houve diferença estatística nos níveis de glutamina entre os momentos de coleta (P > 0,05). Houve aumento significativo dos níveis plasmáticos de amônia apos o termino das provas (P < 0,05). Esse aumento foi mais marcado nos cavalos de 80 km, seguido pelos de 120 e por ultimo os de 160 km. Houve correlação positiva entre os níveis de amônia e glutamina nas categorias de 160, 120 e 80 km (r = 0,209, r = 0,151 e r = 0,176, respectivamente; P < 0,05). Oscilações séricas dos níveis de glutamina são difíceis de se detectar em cavalos de enduro, já que as fibras musculares oxidativas são importantes reservatórios desse aminoácido. Ainda, e difícil saber sobre consumo de glutamina dadas as reservas orgânicas e a facilidade de metabolização. A concentração sérica de amônia pode servir como indicador indireto do consumo de glutamina. Assim, concluiu-se que as dosagens de amônia e cortisol podem servir como medidas indiretas da utilização da glutamina durante o exercício aeróbico e ambas como marcadores de esforço.

Thirty apparently healthy lambs with average weight and age of 5.8±0.5 kg and 15±1.6 days respectively were chosen in Sids Agricultural Research Station belonging to Animal Production Research Institute in Beni-Suef governorate to investigate the effect of castration and docking on maintenance behaviour and blood cortisol level during and after such operations. Lambs were classified into castration (n=10), docking (n=10) and control (n=10) groups. The results revealed that castration and docking could significantly affect the behaviours of standing idle, recumbency, sleep, suckling and ingestion in lambs. Moreover, blood cortisol level was significantly (p<0.05) increased after operation in castrated and docked lambs as compared to the time before, at operation and 3 hours after operation.

The study measured the hormonal and protein markers of acute stress, those of oxidative stress and total antioxidant capacity (TAC) in swine oral fluid, determined which of these parameters would be the most appropriate for future livestock welfare assessment and established the time when the samples should be taken. Stress was induced in 7 out of 14 castrated six-week-old Danbred×Duroc pigs by immobilisation on a nasal snare at 8 a.m., 1 p.m., and 6 p.m. and samples were taken both directly after the stressor was applied and 30 min later. The remaining pigs were the control group, which were not immobilised; their samples were taken at the same times. The concentrations of hormones and malondialdehyde (MDA) were measured using liquid chromatography with tandem mass spectrometry, while those of alpha-amylase and TAC were measured using spectrophotometry. The levels of cortisol and cortisone increased with statistical significance immediately after the acute stress response and 30 min later. A cut-off value set at 0.25 ng/mL cortisol concentration was capable of distinguishing between the stressed and control groups with 100% accuracy in evening samples and 95% accuracy overall. Prednisolone was not present, and the levels of testosterone and corticosterone were low and not distinctive. Alpha-amylase became significantly more concentrated during stress induction and 30 min later. The TAC and MDA levels rose after the stress but without statistical significance. The most suitable markers of acute stress were cortisol, cortisone and alpha-amylase. Oral fluid is a reliable material for monitoring the level of pigs’ stress and should be collected in the evening.

OBJECTIVE: To assess the analgesic efficacy of an IV constant rate infusion (CRI) of a morphine-lidocaine-ketamine (MLK) combination in calves undergoing umbilical herniorrhaphy. ANIMALS: 20 weaned Holstein calves with umbilical hernias. PROCEDURES: Calves were randomly assigned to receive a CRI of an MLK solution (0.11 mL/kg/h; morphine, 4.8 μg/kg/h; lidocaine, 2.1 mg/kg/h; and ketamine, 0.42 mg/kg/h) for 24 hours (MLK group) or 2 doses of flunixin meglumine (1.1 mg/kg, IV, q 24 h) and a CRI of saline (0.9% NaCl) solution (0.11 mL/kg/h) for 24 hours (control group). The assigned CRI was begun after anesthesia induction. A pain-scoring system and incisional algometry were used to assess pain, and blood samples were obtained to measure serum cortisol concentration at predetermined times for 120 hours after CRI initiation. RESULTS: Mean pain scores did not differ significantly between the MLK and control groups at any time. Mean algometry score for the MLK group was significantly greater (calves were less responsive to pressure) than that for the control group at 4 hours after CRI initiation. Mean cortisol concentration decreased over time for both groups and was significantly greater for the MLK group than the control group at 1, 4, and 18 hours after CRI initiation. CONCLUSIONS AND CLINICAL RELEVANCE: A CRI of MLK provided adequate postoperative analgesia to calves that underwent umbilical herniorrhaphy. However, the technical support required for CRI administration limits its use to hospital settings. Kinetic analyses of MLK infusions in cattle are necessary to establish optimal dosing protocols and withdrawal intervals.

OBJECTIVE To determine the effects of stacked wedge pads and chains applied to the forefeet of Tennessee Walking Horses on behavioral and biochemical indicators of pain, stress, and inflamation. ANIMALS 20 Tennessee Walking Horses. PROCEDURES Horses were randomly assigned to 2 treatment groups: keg shoes (control; n = 10) or stacked wedge pads and exercise with chains (10). Ten days before treatment application, an accelerometer was attached at the left metatarsus of each horse to record daily activity. Horses were exercised for 20 minutes daily, beginning on day -7. On day 0, exercise ceased, the forefeet were trimmed, and the assigned treatment was applied. From days 1 through 5, horses were exercised as before. Blood samples for measurement of plasma cortisol, substance P, and fibrinogen concentrations were collected on days -5, 1, and 5 before and after exercise and every 30 minutes thereafter for 6 hours. RESULTS No significant differences in plasma concentrations of cortisol, substance P, and fibrinogen were detected between groups. Although lying behaviors changed after shoes were applied, these behaviors did not differ significantly between groups. Shoeing appeared to have altered behavior to a greater extent than did the type of treatment applied. CONCLUSIONS AND CLINICAL RELEVANCE Application of stacked wedge pads and chains to the forefeet of horses for a 5-day period as performed in this study evoked no acute or subacute stress or nociceptive response as measured. Although these findings should not be extrapolated to the long-term use of such devices in Tennessee Walking Horses performing the running walk, the data should be considered when making evidence-based decisions relating to animal welfare and the use of stacked wedge pads and chains.

OBJECTIVE To assess the discriminatory value for corticosteroid-induced alkaline phosphatase (CiALP) activity and other variables that can be measured routinely on a CBC and biochemical analysis for the diagnosis of hypoadrenocorticism in dogs. SAMPLE Medical records of 57 dogs with confirmed hypoadrenocorticism and 57 control dogs in which hypoadrenocorticism was suspected but ruled out. PROCEDURES A retrospective case-control study was conducted. Dogs were included if a CBC and complete biochemical analysis had been performed. Dogs with iatrogenic hypoadrenocorticism and dogs treated previously with glucocorticoids were excluded. Cortisol concentration for dogs with hypoadrenocorticism was ≤ 2 μg/dL both before and after ACTH administration. Cortisol concentration for control dogs was > 4 μg/dL before or after ACTH administration. RESULTS Area under the receiver operating characteristic (ROC) curve for CiALP activity was low (0.646; 95% confidence interval, 0.494 to 0.798). Area under the ROC curve for a model that combined the CiALP activity, Na-to-K ratio, eosinophil count, activity of creatine kinase, and concentrations of SUN and albumin was high (0.994; 95% confidence interval, 0.982 to 1.000). Results for this model could be used to correctly classify all dogs, except for 1 dog with hypoadrenocorticism and no electrolyte abnormalities. CONCLUSIONS AND CLINICAL RELEVANCE CiALP activity alone cannot be used as a reliable diagnostic test for hypoadrenocorticism in dogs. Combined results for CiALP activity, Na-to-K ratio, eosinophil count, creatine kinase activity, and concentrations of SUN and albumin provided an excellent means to discriminate between hypoadrenocorticism and diseases that mimic hypoadrenocorticism.

The objective of this study was to develop a time-resolved immunofluorometric assay (TR-IFMA) for quantification of salivary alpha-amylase in sheep. For that purpose, after the design of the assay, an analytical and a clinical validation were carried out. The analytical validation of the assay showed intra- and inter-assay coefficients of variation (CVs) of 6.1% and 10.57%, respectively and an analytical limit of detection of 0.09 ng/mL. The assay also demonstrated a high level of accuracy, as determined by linearity under dilution. For clinical validation, a model of acute stress testing was conducted to determine whether expected significant changes in alpha-amylase were picked up in the newly developed assay. In that model, 11 sheep were immobilized and confronted with a sheepdog to induce stress. Saliva samples were obtained before stress induction and 15, 30, and 60 min afterwards. Salivary cortisol was measured as a reference of stress level. The results of TR-IFMA showed a significant increase (P < 0.01) in the concentration of alpha-amylase in saliva after stress induction. The assay developed in this study could be used to measure salivary alpha-amylase in the saliva of sheep and this enzyme could be a possible noninvasive biomarker of stress in sheep.
