Effect of serial in vivo passages on the adaptation of H1N1 avian influenza virus to pigs

The lack of proofreading activity of the viral polymerase and the segmented nature of the influenza A virus (IAV) genome are responsible for the genetic diversity of IAVs and for their ability to adapt to a new host. We tried to adapt avian IAV (avIAV) to the pig by serial passages in vivo and assessed the occurrence of point mutations and their influence on viral fitness in the pig’s body. A total of 25 in vivo avIAV passages of the A/duck/Bavaria/77 strain were performed by inoculation of 50 piglets, and after predetermined numbers of passages 20 uninoculated piglets were exposed to the virus through contact with inoculated animals. Clinical signs of swine influenza were assessed daily. Nasal swabs and lung tissue were used to detect IAV RNA by real-time RT-PCR and isolates from selected passages were sequenced. Apart from a rise in rectal temperature and a sporadic cough, no typical clinical signs were observed in infected pigs. The original strain required 20 passages to improve its replication ability noticeably. A total of 29 amino-acid substitutions were identified. Eighteen of them were detected in the first sequenced isolate, of which 16 were also in all other analysed strains. Additional mutations were detected with more passages. One substitution, threonine (T) 135 to serine (S) in neuraminidase (NA), was only detected in an IAV isolate from a contact-exposed piglet. Passaging 25 times allowed us to obtain a partially swine-adapted IAV. The improvement in isolate replication ability was most likely related to S654 to glycine (G) substitution in the basic protein (PB) 1 as well as to aspartic acid (D) 701 to asparagine (N) and arginine (R) 477 to G in PB2, glutamic acid (E) 204 to D and G239E in haemagglutinin and T135S in NA.

A useful approach for evaluating antitussive drugs in humans is to determine the sensitivity of the cough reflex to a standard challenge. The purpose of this study was to determine if methods used to induce coughing in humans would be effective when used on awake, untrained, healthy dogs for future application in therapeutic trials involving dogs with spontaneous disease. Methods tested were: mechanically stimulating the trachea by digital compression as well as by vibration from an electric shaver, neck massager, and palm sander (11 dogs), and administering nebulized irritant (3000 micromolar capsaicin), acidic (1 M citric acid), and hypotonic (deionized water) solutions using face masks (4 dogs). The threshold for success was defined as induction of at least 2 moderate or strong coughs in at least 75% of the dogs. None of the methods tested was successful. Digital compression induced soft (n = 2) or moderate (n = 1) coughing in 3 of 11 dogs tested. Nebulization of citric acid induced 1 soft cough in 1 of 4 dogs. It was concluded that coughing cannot be successfully induced in awake, healthy dogs using methods that are successful in humans. Other strategies must be developed so that cough sensitivity can be objectively and non-invasively measured in dogs for clinical research purposes.

A prospective cohort study was undertaken in a farrow-to-farrow swine herd to describe patterns of pneumonia, and to identify host risk factors associated with the extent of pneumonic lesions in 2 weight groups of slaughter swine. The risk of coughing and pneumonic lesions increased with increasing age of pigs within the herd (P < 0.0001). The age-specific prevalence of pneumonic lesions was low (2.7%) in pigs < 16 weeks old at slaughter, but increased rapidly when pigs were between 16 and 22 weeks old (8.6 to 67.9%). After 22 weeks, the prevalence remained relatively constant at about 80%. Associations between possible risk factors and pneumonia were investigated by use of multiple-regression models. Age at weaning (< 24 days) and birth weight (< 1 kg) exerted small, but significant (P < 0.002), effects on the extent of pneumonic lesions in pigs slaughtered at 30 to 50 kg live weight. For pigs slaughtered at 90 to 110 kg, pneumonic lesions were more extensive (P = 0.007) in pigs sired by Yorkshire boars than pigs sired by non-Yorkshire sires (Duroc, Hampshire, Chester White, or American Spotted). Other host factor variables including weaning weight and clinical diseases (atrophic rhinitis, diarrhea, and arthritis) were not associated with pneumonia extent in either weight group. Higher pneumonia percentages were also associated with reduced growth rates in the grower/finisher phase. Pigs sired by Yorkshire boars grew significantly (P < 0.0001) more slowly from entry into shed 2 (mean, 38 kg) until about the time of exit (mean, 92 kg) than pigs sired by other breeds (747 g/d and 795 g/d, respectively).

A rare case is presented of a 58-year-old woman who developed ahepatic-bronchopleural fistula following a hydatid hepatic cyst complication. The hepatic-bronchopleural fistula was diagnosed when patient complained of severe repetitive attacks of productive cough of brown yellow to dark colour sputum biliptysis (bile in cough) with vomiting associated with night sweating and fever, for the past one month. She also complained of right side chest and abdominal pain for the past 3 years. This paper describes a unique case of hepatic-bronchopleural fistula caused by hydatid disease, emphasising the clinical feature and its surgical management. Hydatid disease is a zoonotic infection dueto the damage caused by Echinococcus granulosus. E. granulosus, also called the hydatid worm, hyper tape-worm or dog tapeworm, is a cyclophyllid cestode that parasitises the small intestine of canids as an adult, but which has important intermediate hosts such as livestock and humans, where it causes cystic echinococcosis, also known as hydatid disease. It is hoped thatthis case report will add to the meagre case management reports of hepaticbronchopleural fistula in literature.

Objective: To characterize clinical and behavioral changes in calves following inoculation with Mycoplasma bovis and evaluate relationships between those changes and pulmonary disease. Animals: 22 healthy Holstein steers. Procedures: 20 calves were inoculated intranasally with < 10(8) CFU or > 10(9) CFU of M bovis. Calves were assigned a clinical illness score (CIS) on a scale of 1 through 4 twice daily on the basis of severity of cough, labored breathing, and lethargy. For each calf, distance traveled and time spent near the waterer, feed bunk, or shelter were determined via a remote location monitoring device. Calves were euthanized and necropsied 22 days after inoculation. Results: 13 calves became clinically ill after challenge inoculation; 3 calves were euthanized within 20 days. Among all calves, consolidation was evident in 0% to 79.9% of the lungs; extent of lung consolidation did not differ between the challenge dose groups. Distance traveled and percentages of time spent in proximity to the feed bunk and shelter were associated with CIS; calves with more severe disease traveled less distance and spent less time at the feed bunk and more time in the shelter. Distance traveled by calves was negatively associated with extent of lung consolidation (< or ≥ 10% of lungs affected); this effect was modified by trial day. Conclusions and Clinical Relevance: Following inoculation with M bovis, calf behavior patterns were associated with both CIS and severity of pulmonary disease. Use of behavior monitoring systems may aid in recognition of respiratory tract disease in calves.
