The Effect of Replacement of Poultry by-Product with Fish Meal in the Diet of Nile Tilapia (Oreochromis niloticus) on their Intestine Histology

BACKGROUND: Tilapia, Oreochromis niloticus, is an omnivorous and important breeding species in the world. Over the recent years, tilapia breeding has begun in Iran. In aquaculture, it is very important to regulate a balanced diet in terms of nutrients needed for growth, which would be also economically appropriate.OBJECTIVES: The main objective of this study was to investigate the effect of substituting chicken waste powder in the diet of fish at levels of 0, 25, 50, 75, and 100 % on the tissue structure of different parts of the intestine of Nile tilapia.METHODS: 75 fish (three frequency per treatment) were fed with chicken waste powder instead of fish meal in their diet for eight weeks at 0, 25, 50, 75, and 100 % levels. At the end of the treatment, separate samples were taken from the initial, middle, and end parts of the intestine. After passing the usual histological steps, the slides were assessed by hematoxylin-eosin staining and TUNEL immunohistochemistry technique.RESULTS: In histometric studies, the highest villi height, thickness, muscle layer, and number of goblet cells were reported in the control group and the lowest in the 100 % group. In immunohistochemical studies, the number of apoptotic cells in groups 75 and 100 % was higher than that in the other groups.CONCLUSIONS: According to the results of the present study, it could be suggested that the replacement of chicken waste powder instead of a fish meal up to 50 % for indigo tilapia is tolerable. However, higher levels of chicken waste powder will reduce absorption, and acceptance of food and have adverse effects on growth.
Show more [+] Less [-]The preventive role of Spirulina platensis (Arthrospira platensis) in immune and oxidative insults in a stress-induced rat model

There is a balance between oxidative stress, antioxidant capacity and immune response. Their roles in physiological and behavioural mechanisms are important for the maintenance of the organism’s internal equilibrium. This study aimed to evaluate the antioxidant effects of the exogenous alga Spirulina platensis (Arthrospira platensis) in a stress-induced rat model, and to describe its possible mechanism of action. Thirty-six adult male Sprague Dawley rats were separated into four groups: control (C), stress (S), S. platensis (Sp), and S. platensis + stress (SpS). The rats in groups Sp and SpS were fed with 1,500 mg/kg b.w./day Spirulina platensis for 28 days. All rats were exposed to prolonged light phase conditions (18 h light : 6 h dark) for 14 days. The SpS and S groups were exposed to stress by being kept isolated and in a crowded environment. Blood samples were obtained by puncturing the heart on the 28th day. The effect of stress on serum corticosterone, oxidative stress markers (TOS, TAC, PON1, OSI) and immunological parameters (IL-2, IL-4, IFN-ɣ) were tested. Also, the brain, heart, intestines (duodenum, ileum, and colon), kidney, liver, spleen, and stomach of the rats were weighed. Serum corticosterone levels were higher in the S group than in the C group, and significantly lower in the SpS group than in the S group. Mean total antioxidant capacity were lower in the S group than in the C group, and Spirulina reversed this change. Although not significantly different, IL-2 was lower in the S group than in the C group. However, in the SpS group, IL-2 increased due to Spirulina platensis mitigating effects of stress. Male rats fed a diet with Spirulina platensis could experience significantly milder physiological changes during stress, although stress patterns may be different. Exogenous antioxidant supplements merit further investigation in animals and humans where the endogenous defence mechanism against stress may not be sufficient.

Eggs are important to the diet of Canadians. This product is one of the supply-managed commodities in Canada, but unlike other commodities, where food safety risks are extensively explored and reported, information on the prevalence of enteric organisms (e.g., Salmonella, Campylobacter) and antimicrobial resistance (AMR) in layers in Canada are limited. This study was conducted to determine the prevalence of select bacteria and the associated AMR patterns in layer flocks using 2 sample matrices. Farms were located within FoodNet Canada and the Canadian Integrated Program for Antimicrobial Resistance Surveillance sentinel sites (SS). Fecal samples (Ontario: ONSS1a, ONSS1b) and environmental sponge swabs (British Columbia: BC(SS2a)) were collected. Salmonella prevalence was 29% and 8% in ONSS1a and ONSS1b, respectively, and 7% in BC(SS2a). S. Kentucky and S. Livingstone were the most frequently isolated serovars and no S. Enteritidis was detected. Campylobacter was not detected in the BC sponge swabs but was isolated from 89% and 53% of Ontario fecal samples (ON(SS1a) and ON(SS1b), respectively). Seven C. jejuni from Ontario were ciprofloxacin-resistant. Escherichia coli prevalence was high in both sample types (98%). Overall, tetracycline resistance among E. coli ranged from 26% to 69%. Resistance to ceftiofur (n = 2 isolates) and gentamicin (n = 2) was relatively low. There were diverse resistance patterns (excludes susceptible isolates) observed among E. coli in Ontario (10 patterns) and British Columbia (14 patterns). This study revealed that fecal samples are more informative for farm-level monitoring of pathogen and AMR prevalence. Without further validation, sponge swabs are limited in their utility for Campylobacter detection and thus, for public health surveillance.
