Objectives: The objective of this study was to isolate and identify Staphylococcus aureus and Escherichia coli from raw milk samples of cattle and buffalo, and to evaluate the antibiotic sensitivity pattern. Materials and methods: A total of 34 milk samples were collected twice from 17 different healthy cattle (n=14) and buffaloes (n=3) at one-month interval, and analyzed in laboratory by staining, cultural and biochemical characteristics followed by polymerase chain reaction targeting nuc gene of S. aureus and 16 S rRNA of E. coli. Antibiotic sensitivity pattern of the isolated bacteria was assessed using the disc diffusion method. Results: Confirmation of the isolates as S. aureus and E. coli were carried out by PCR using nuc gene, 16S rRNA gene specific primers specific for S. aureus and E. coli respectively. A total of 12 samples (35.29%; 11 from cattle, 1 from buffalo) were found to be positive for S. aureus; 5 and 7 during first and second month, respectively. The E. coli were found in three samples (2 from cattle, 1 from buffaloe); one in first month and two in the second month. The antibiotic sensitivity test using 4 commonly used antibiotics indicated that the most of the isolates were resistant to Gatifloxacin and one isolate showed intermediate resistance to Ofloxacin while sensitive to Ciprofloxacin and Levofloxacin.Conclusion: Two different species of bacteria i.e., S. aureus and E. coli are contaminating with milk samples. The pathogenic bacteria can be controlled effectively by using Ciprofloxacin and Levofloxacin in the case of mastitis in cattle and buffaloes in Bangladesh. A crucial first step in preventing the introduction and spread of harmful microorganisms in poultry farms is biosecurity. The objectives of this investigation were to assess biosecurity measures and the prevalence of different E. coli species in commercial chicken farms in Sharkia province, Egypt. Sensitivity of E. coli strains was identified, and their susceptibility to antibiotics and disinfection was evaluated as well. Four farms provided 21 samples each, for a total of 84 samples. Three samples of each were collected for water, feed, litter, cloaca, wall, hand, and foot boots. All the studied farms obtained a "poor biosecurity" grade, meaning that their overall biosecurity score, which is comprised of 40% external and 36.54% internal biosecurity, was less than 50%. Escherichia coli species were found in 79.76% of the samples. It was isolated from water, feed, litter, cloaca, walls, hands, and foot boots with percentages of 91.6; 83.3; 91.6; 100; 58.3; 58.3 and 75%, respectively. Biosecurity level had significant negative correlations with the isolated E. coil species. Escherichia coli O119 was the most common serotype in litter and the cloaca, while E. coli O1 was the most prevalent serotype in walls and hands. Additionally, serotypes for E. coli O26, O159, O128 and O78 were included. According to the patterns of antibiotic sensitivity, amikacin, nitrofurantoin, gentamycin, and levofloxacin showed intermediate sensitivity, whereas E. coli O119 bacteria were highly sensitive to ciprofloxacin. In the absence and presence of organic matter, Virocid, Cid 2000 and finally Virkon S were the most efficient against E. coli O119. According to this study, there were differences in the investigated farms' biosecurity levels in relation to the occurrence of Escherichia coli. To reduce the introduction and persistence of E. coli in poultry farms, several biosecurity practices and management, including stringent cleaning and disinfection measures should be applied.

Avian colibacillosis is one of the most serious diseases that affect poultry and causes substantial morbidity and mortality rates as well as high economic losses. E. coli, are capable of acquiring resistance genes via gene transfer. The development of extended spectrum lactamases, or broad-spectrum lactamases, in E. coli is the most serious resistance mechanism. The goal of the current study was to detect the resistance associated genes of multi drug resistant E. coli isolated from broiler chicken by using PCR technique. In the current study PCR applied on 10 multidrug resistant E. coli isolates for detecting β-lactamases resistance genes (blaTEM, blaSHV), integron resistance gene (Int1, Int3), PCR was also used to detect disinfectant resistance genes as Quaternary ammonium compounds resistance genes (QacCD, QacA/B and QacED1). PCR results for antibiotic resistance associated gene showed that (10/10) of tested isolates had blaTEM and int1(10/10), blaSHV (6/10) and int3 (2/10) also PCR results for disinfectant resistance associated genes showed that (8/10) of E. coli isolates had QacED1, QacCD (2/10), and QacA/B (2/10). The (10) broilers flocks investigated in the study were infected with multi drug resistant, strains of E. coli, that haboured β-lactamases, integron resistance associated gene and quaternary ammonium compounds resistance genes.

This paper describes a fatal case of pneumonia in a 14-day-old dog caused by extraintestinal pathogenic Escherichia coli (ExPEC). The necropsy showed that almost all of left lobes of the lungs had dark-red consolidation. A histopathology examination revealed moderate acute fibrino-hemorrhagic necrotizing pneumonia with intralesional bacterial colonies. Non-suppurative epicarditis, congestion in the liver, and necrosis in the white pulp of the spleen also were found. E. coli with cytotoxic necrotizing factor 1 and α-hemolysin was isolated from the lung. This case was confirmed to have fatal pneumonia caused by ExPEC that led to a systemic infection.

While serum amyloid A (SAA) has been investigated as a potential marker for septic arthritis in horses, no study has reported on whether SAA can be used to detect eradication of joint infection. Therefore, the objective of this study was to investigate whether the eradication of joint infection in experimentally induced septic arthritis in horses can be detected using serum and synovial fluid SAA. A total of 17 horses were randomly assigned to 3 groups. A middle carpal joint of each horse was injected with saline (control group, n = 3), lipopolysaccharide (LPS) (nonseptic synovitis group, n = 6), or Escherichia coli (septic arthritis group, n = 8) on day 0. Starting on day 1, horses underwent treatment for septic arthritis. Sequential samples of serum and synovial fluid were collected, and quantification of SAA was carried out. Concentrations of serum and synovial fluid SAA were compared among groups and time points. A concurrent study was conducted and determined that infection was eradicated on day 4 in this experimental model of septic arthritis. Concentrations of serum and synovial fluid SAA rapidly increased after inoculation of E. coli and were highest on day 3 and day 4, respectively. Thereafter, both serum and synovial fluid SAA decreased with eradication of joint infection, although they remained significantly increased from baseline until day 9 and day 10, respectively. Serum and synovial fluid SAA did not increase in the control or nonseptic synovitis group. These findings suggest that serial measurements rather than a single measurement of SAA are required to determine eradication of infection from septic arthritis in horses.

Pyometra is an inflammation of the uterus which is characterized by purulent to sanguinopurulent intraluminal contents. Pyometra has been mostly observed in queens older than 6 years but it has no age predilection. The average age of cats with pyometra is between 5 - 7 years. Traditionally queens reach puberty at a bodyweight of 2.3-2.5 kg. Here in this case we have a 5-month-old cat which has not experienced oestrus but yet developed a pyometra. In this case ovariohysterectomy was performed on this queen. The treatment procedure continued with fluid therapy and antibiotics.

Antimicrobials (AM) are used for growth promotion and therapy in pig production. Its misuse has led to the development of resistant organisms. We evaluated Escherichia coli virulence genes, and compared phenotypic-genotypic antimicrobial resistance (AMR) patterns of faecal E. coli from pigs receiving routine farm treatment without antimicrobial agents against pigs treated routinely with AM over 70 days. Recovered E. coli were tested for AMR using disk diffusion and polymerase chain reaction. Virulence genes were detected in 24.8% of isolates from antimicrobial group and 43.5% from non-antimicrobial group (p = 0.002). The proportion of virulence genes heat-stable enterotoxins a & b (STa, STb), enteroaggregative heat stable enterotoxin 1 [EAST1] and Shiga toxin type 2e [Stx2e]) were 18.1%, 0.0%, 78.7% and 3.0% for antimicrobial group and 14.8%, 8.5%, 85.1% and 12.7% for non-antimicrobial groups, respectively. Resistance to oxytetracycline was most common (p = 0.03) in samples collected between days 10 and 21. Resistance shifted to amoxicillin on days 56-70, and trimethoprim resistance was observed throughout. Seventeen phenotypic AMR combinations were observed and eight were multidrug resistant. At least one tetracycline resistance gene was found in 63.9% of the isolates. tet (A) (23.3%) was most common in the antimicrobial group, whereas tet (B) (43.5%) was prevalent in the non-antimicrobial group. Usage or non-usage of antimicrobial agents in growing pigs does not preclude virulence genes development and other complex factors may be involved as previously described. Heavily used AM correspond to the degree of resistance and tetracycline resistance genes were detected during the growth phase.

Septic arthritis is an important disease in horses, necessitating aggressive and prolonged therapy. In order to guide therapy, reliable methods of detecting the eradication of infection are needed. Therefore, the objective of this study was to investigate detection of eradication of infection in an experimental model of equine septic arthritis using standard diagnostic techniques. For this purpose, 17 adult horses were assigned to 3 experimental groups. The middle carpal joint of each horse was injected with Escherichia coli (Septic group, n = 8), lipopolysaccharide (LPS) (LPS group, n = 6), or sterile saline (Control group, n = 3) at day 0. Contralateral joints were not injected. Standard therapy was applied to all joints except non-injected joints in the Control group at day 1. Sequential samples of synovial fluid (SF) were collected for bacterial culture using 3 culture media [Columbia blood agar (CBA), brain heart infusion broth (BHI), and Signal blood culture medium] and for cytological evaluation [percentage neutrophils (PN), total nucleated cell count (TNCC), and total protein (TP)]. Escherichia coli-specific polymerase chain reaction (PCR) was carried out to detect E. coli DNA in synovial fluid. Culture and PCR were positive for E. coli in all joints injected with E. coli at day 1 and 1 joint was positive on BHI at day 4. Based on the results of bacterial culture, PCR, and TNCC, the elimination of infection in our experimental model occurred by day 4 post-infection in 6 out of 7 cases. Total protein (TP) and PN remained elevated at clinical threshold used for diagnosis of septic arthritis until day 14. In our experimental model of E. coli-induced arthritis, we conclude that TP and PN may not be good indicators for detecting the eradication of bacterial infection caused by E. coli from infected and subsequently treated joints.

Poultry has been identified as a reservoir of foodborne enteric pathogens and antimicrobial resistant bacteria. The objective of this study was to describe and compare antimicrobial resistant isolates from an Ontario broiler chicken farm-level baseline project (2003 to 2004) to the Canadian Integrated Program for Antimicrobial Resistance Surveillance (CIPARS) Ontario abattoir and retail surveillance data from 2003, and to the most recent (2015) CIPARS Ontario chicken surveillance data in order to assess the impact of an industry-wide policy change in antimicrobial use. Ceftiofur resistance (TIO-R) prevalence in Salmonella decreased by 7% on farm between 2003 and 2004 and 2015. During the same timeframe, TIO-R E. coli prevalence decreased significantly by 16%, 11%, and 8% in farm, abattoir, and retail samples, respectively. Gentamicin resistant (GEN-R) E. coli, however, increased by 10% in farm and 15% in retail-derived isolates, and trimethoprim-sulfamethoxazole resistant (TMSm-R) E. coli increased significantly by 20%, 18%, and 5% in farm, abattoir, and retail isolates, respectively. Similarly, ciprofloxacin-resistant (CIP-R) Campylobacter spp. significantly increased in retail isolates by 11% and increased in farm (33%) and abattoir isolates (7%). The decrease in TIO-R Salmonella/E. coli in recent years is consistent with the timing of an industry-led intervention eliminating the preventive use of ceftiofur, a third generation cephalosporin and class of antimicrobials deemed critically important to human medicine. The rise in GEN-R and TMSm-R prevalence is indicative of recent shifts in antimicrobial use. Our study highlights the importance of integrated surveillance in detecting emerging trends and determining the efficacy of interventions to improve food safety.

Colistin is one of the last-resort antibiotics for the treatment of multidrug-resistant infections in humans, but transmissible colistin-resistance genes have emerged in bacteria from animals. The rapid and sensitive detection among animals of colonization with bacteria carrying these genes is critical in helping to control further spread. Here we describe a method for broth enrichment of colistin-resistant Escherichia coli from animal fecal and cecal samples followed by real-time polymerase chain reaction (PCR) for the simultaneous detection of two of the main colistin-resistance genes, mcr-1 and mcr-2. The PCR uses a single set of nondegenerative primers, and mcr variants can be differentiated by melt-curve analysis. Overnight culture enrichment was effective for amplifying colistin-resistant E. coli, even when initially present in numbers as low as 10 bacteria per gram of sample. The mcr-1 and mcr-2 genes were not found in any of the Ontario swine and poultry samples investigated.
