Comparative expression analysis of inflammatory and immune-related genes in cattle during acute infection with foot-and-mouth disease virus in Egypt

Foot-and-mouth disease is a highly infectious viral disease affecting all cloven-footed domestic animals. The three foot-and-mouth disease virus (FMDV) serotypes A, O and SAT2 are at present the greatest threat to susceptible animals in Egypt. The aim of the present study was, for the host factors associated with different FMDV infections in cattle during the acute phase, to compare these factors’ influence on the expression of the IL-10, TLR-2, TNF-α, CXCL10, CD48, NFATC4 and IFNG inflammatory and immune-related genes.

The aim of the present study was, for the host factors associated with different FMDV infections in cattle during the acute phase, to compare these factors’ influence on the expression of the IL-10, TLR-2, TNF-α, CXCL10, CD48, NFATC4 and IFNG inflammatory and immune-related genes. Vesicular fluid and epithelium samples were obtained from at least three infected cattle on the same affected farm during three different FMDV outbreaks and were used for serotyping of the virus and for expression analysis of host genes. A two-step RT-PCR was used for diagnosis of the virus with primers specific for each serotype. In quantitative PCR analysis, the expression patterns of TLR-2 and IFNG were prominent, while NFATC4 expression was absent in all FMDV-infected cattle. The highest expression of CD48 was associated with increased expression of other inflammatory and immune-related genes (IL-10, TLR-2, TNF-α and IFNG), which may be an indication of rapid virus clearance. The use of vesicular fluid and epithelium for investigation of viral and immune-related gene expression levels in acute FMDV infection is possible. Host-dependent variation in the expression of the studied genes was observed in different FMDV serotype outbreaks.

Objective: Molecular based study was conducted to determine the associated serotypes in the reemergence of Foot and Mouth Disease (FMD) outbreak in Assiut governorate, Egypt during 2014 and 2015. Materials and methods: One hundred and twenty cattle with clinical signs suggesting their infection by FMDV were examined clinically and twenty three of them were used for confirmation by laboratory diagnosis. Different clinical samples including vesicular fluid and tongue epitheliums were collected and after RNA extraction using commercial kit, RT-PCR was done using different primer sets. Results: Serotype O was detected in 8 samples, 2 of them were also positive for SAT2 serotype. The determination of specific serotype was failed in case of the rest 13 samples although they were positive when tested by the universal primer specific for FMDV. Conclusion: Serotypes O and SAT2 were the more prevalent serotypes in the current outbreak in Assiut governorate, Egypt. [J Adv Vet Anim Res 2017; 4(1.000): 32-38]

This research work was conducted to investigate the effects of age, sex and breed on the induction of immune response against experimentally prepared inactivated trivalent (type O, A, and Asia-1) FMD vaccine. Twenty six cattle were divided into four test groups (Group A, B, C, and D; 5 cattle in each group) and one control group (n=6) based on breed (local and cross), age (≤12 months and >12 months), and sex (male and female). Test cattle were vaccinated with the experimentally prepared trivalent FMD vaccine. Pre- and post vaccinated sera from the vaccinated cattle were collected upto 63 days, and the sera were tested using liquid phase blocking enzyme linked immunosorbent assay (LPBE) that was specific for FMD serotypes O, A, and Asia-1. Antibody titers of all the pre-vaccinated serum samples were found to be under protection level. The females were found to be more protected (90%; n=9/10) as compared to males (70%; n=7/10). The titers obtained were statistically analyzed using t–test to observe the effects of age, breed and sex. It was observed that the mean values of antibody titer in cattle aging >12 months against O, A, and Asia-1 serotypes were significant (P<0.05) at 21, 49 and 63 days as compared to the values obtained from the cattle aging ≤12 months. In conclusion, the local female cattle aging >12 months showed better immune response towards trivalent FMD vaccine.

Objective: The study aimed to explore the seroprevalence of some bacterial (Brucella spp., Chlamydia abortus) and viral [Rift Valley fever virus (RVFV), foot and mouth disease virus (FMDV)] zoonoses in domestic ruminants in Medina. Materials and Methods: A total of 1,000 blood samples from 665 sheep, 228 goats, and 107 camels were collected from the central slaughterhouse, private farms, and veterinary clinics affiliated to the Ministry of Agriculture. The samples were screened using the Enzyme-Linked Immunosorbent Assay (ELISA). The results were statistically analyzed using Statistical Package for the Social Sciences version 20. Results: Brucella was found in 7.7%, 8.8%, and 6.5% of sheep, goats, and camels sera, respec¬tively. In humans, Brucella abortus and Brucella melitensis had higher frequencies in males (61.3%, 59.8%) than in females (38.7%, 40.2%). Chlamydia abortus was reported only in sheep at a rate of 0.75%. RVFV was prevalent in sheep (10.7%) and goats (17.9%). FMDV was reported in 27.8% of sheep and 7.9% of goats. There was a highly statistical significance between Brucella, RVFV, and FMDV seropositivity and locally bred animals (p < 0.01). Multiple seropositivities have been reported among sheep and goats. Brucella was commonly observed in mixed infection with other bacterial and viral agents under study. Conclusion: The surveyed viral and bacterial agents were prevalent in ruminants in the Medina region. Since Medina is an important destination for pilgrims from all over the world, therefore, an integrated approach involving strict control measures and routine vaccination programs should be adopted to reduce the possibility of global epidemics. Objective: This research work was conducted for the molecular characterization of the circulating foot-and-mouth disease (FMD) virus in Bangladesh and revealed out their serotype. Materials and methods: The VP1 gene of six field isolates of FMD virus (FMDV) serotypes (two serotypes O, two serotypes A, and two serotypes Asia 1) was subjected for sequencing and phy¬logenetic analysis. Neighbor-joining trees were constructed by using the Molecular Evolutionary Genetics Analysis 6, having the field nucleotide sequences of FMDV and related sequences avail¬able in the GenBank. Results: The nucleotide sequences of the VP1 genes of serotypes O, A, and Asia-1 of the isolates revealed that overall isolates were 91%100% similar to the isolates reported from Bangladesh and other neighboring countries. Among the isolates reported from Bangladesh, serotype O had 98%100% identity, serotype A had 91%100% identity, and serotype Asia-1 had 94%100% iden¬tity. A phylogenetic analysis revealed that the FMDV serotype O PanAsia-02 sub-lineage was con¬firmed in Bangladesh under the Middle EastSouth Asian (ME-SA) topotype. On the other hand, we identified genotype VII (18) of Asia topotype (serotype A) and lineage C (serotype Asia-1). Conclusion: The FMDV serotype O PanAsia-02 sub-lineage was confirmed in Bangladesh under the ME-SA topotype for the first time. Objective: The objective of the study was to investigate Foot and Mouth Disease virus (FMDV) outbreak in cattle in the Sarankhola Upazila under Bagerhat district of Bangladesh with isolation, identification, and molecular characterization of FMDV during April 2018. Materials and Methods: This Upazila is located at southern border of Bangladesh and surrounded by mangrove forest Sundarban. The outbreak investigation team collected epidemiological data from outbreak location. In addition, the team collected a total of 30 (15 calves, 15 adult) tongue epithelial tissue samples from a clinically FMD-affected cattle. The confirmation of FMDV and its three serotypes (A, O, and Asia-1) was performed by Reverse Transcriptase Polymerase Chain Reaction (RT-PCR). An amplified product of the VP1 region of FMDV genome was sequenced by Sanger sequencing method after cultivation and reconfirmation of FMDV into the BHK21 cell line. Genetic variability was studied by constructing a phylogenetic tree. Results: The investigation survey was carried out in overall 8,393 (8,393/15,580; 53.89%) cases including 3,050 (3,050/8,393; 36.34%) cases in calf and 5,343 (5,343/8,393; 59.77%) cases in adult cattle. The overall case fatality rate (CFR) was recorded as 2.27% (354/15,580) with significantly highest CFR in the calf (71.46%; 253/354) compared to an adult. The collected all 30 samples found with FMDV positive and mixed infection of all samples with serotype Asia-1 and serotype O were observed. In BHK 21 cell line, the eight FMDV positive samples showed a typical cytopathic effect during the third passage. Finally, DNA sequence data of two isolates found closely related with the isolates of bordering country India and Myanmar. Conclusion: The investigation identified the risk factors involved in an outbreak of FMDV, namely, sharing the common paddy land after harvesting, no FMD vaccination, the interaction between cattle and wildlife, and cross bordering movement. The study aimed for the detection and serotyping of Foot and Mouth Disease virus (FMDV) circulating in Kapasia Upazila, Gazipur district of Bangladesh during 2013. Twelve samples comprising of tongue epithelium (n=8) and inter digital tissue (n=4) were collected from suspected cattle, and inocula were prepared. The inocula were inoculated into confluent BHK-21 cell line for virus propagation. After 3 subsequent passages; progressive cytopathic effects (CPE) specific for FMDV i.e., rounding and flattening of cells, breaking down of the intercellular bridge and finally cell death (almost 100%) were observed; these were indicative of successful virus propagation in the cells. Viral RNA was extracted, and Reverse Transcription Polymerase Chain Reaction (RT-PCR) was performed using three sets of primers corresponding to the serotype and lsquo;O', and lsquo;Asia-1' and and lsquo;A', respectively. Out of the 12 samples, 10 (83.33%) were found to be positive for FMDV, and all of those were of serotype and lsquo;O'. It is concluded that FMDV serotype and lsquo;O' is circulating among the cattle of Gazipur district, Bangladesh. [J Adv Vet Anim Res 2015; 2(3.000): 291-295]

Objective: In this study, a serological survey was conducted in unvaccinated sheep and goat populations at Pyawbwe and Meikhtila townships of Mandalay region in Myanmar to determine the seroprevalence and associated risk factors of foot and mouth disease (FMD). Materials and methods: A total of 110 sheep and 107 goat sera samples were randomly collected from Pyawbwe. Similarly, 108 sheep and 109 goat sera were collected from Meikhtila. All samples were tested for the presence of non-structural protein (NSP) specific antibodies to FMD virus (FMDV) by Ceditest FMDV-NSP Enzyme-lined Immunosorbent Assay (ELISA), and were confirmed by Liquid Phase Blocking ELISA (LPB ELISA) . Results: Overall seroprevalence was 42.4%(n=184/434) by Ceditest-NSP ELISA, and 46.8%(n=203/434) by LPB ELISA against FMDV serotype O. The presence of antibodies against FMDV serotype O was higher (P<0.01) as compared to those of serotype A and Asia-1. The seroprevalence in Meikhtila (49.77%) was higher (P<0.01) than that of Pyawbwe (35.2%). The seropositivity in sheep and goats that were in-contact (77.19%) with infected cattle and pigs was higher (P<0.01) as compared to those in-contact with non-infected animals (37.14%). Similarly, the seropositivity in sheep and goats from high animal trade areas (49.4%) was higher (P<0.05) than that of those from low animal trade areas (37.97%). Conclusion: Rearing of sheep and goats in-contact with FMDV-infected cattle and pigs, and high animal trading areas are the major associated risk factors for FMDV infection for sheep and goats in the study areas in Myanmar.
