Reproductive tract disease associated with inoculation of pregnant white-tailed deer with bovine viral diarrhea virus

Objective-To inoculate white-tailed deer (Odocoileus virginianus) during the sixth or seventh week of gestation with bovine viral diarrhea virus (BVDV) and observe for signs of reproductive tract disease during a 182-day period. Animals-10 pregnant white-tailed deer (8 seronegative and 2 seropositive [control deer] for BVDV). Procedures-Deer were inoculated with 1 of 2 deer-derived BVDV strains (RO3-20663 or RO3-24272). Serum anti-BVDV antibody titers were determined prior to and 21 or 35 days after inoculation. Virus isolation (VI) procedures were performed on tissues from fetuses and does that died and on blood samples collected from live fawns. Ear notch specimens obtained from live fawns were assessed by use of BVDV antigen-capture ELISA (ACE). Results-Both RO3-20663-inoculated seropositive deer gave birth to apparently normal fawns. Among the RO3-24272-inoculated seronegative deer, 1 died, and 1 aborted and 1 resorbed their fetuses; among the RO3-20663-inoculated seronegative deer, 3 died, 1 aborted its fetus, and 1 gave birth to 2 fawns that were likely persistently infected. On the basis of VI and ACE results, those 2 fawns were positive for BVDV; both had no detectable neutralizing anti-BVDV antibodies in serum. Conclusions and Clinical Relevance-Reproductive tract disease that developed in pregnant white-tailed deer following BVDV inoculation was similar to that which develops in BVDV-exposed cattle. Methods developed for BVDV detection in cattle (VI, immunohistochemical evaluations, and ACE) can be applied in assessments of white-tailed deer. Fawns from does that had serum anti-BVDV antibodies prior to inoculation were protected against BVDV infection in utero.

Data on 5,711 Duroc-sired, 2,227 Landrace-sired, and 2,494 Yorkshire-sired male pigs born over a 9-year period were used to evaluate the genetic influence on scrotal herniation. Differences in frequency of this defect among boar breeds (Duroc, Landrace, and Yorkshire) were significant (P < 0.01). Differences among sires within the Duroc and Landrace boar groups were significant (P < 0.001 and P < 0.05, respectively), but differences within the Yorkshire group were not significant. Frequency of scrotal hernia among male full siblings of affected males was consistently higher than the overall frequency of the defect among progeny in each of their respective breed of boar groups. Percentage of affected pigs among male full siblings of affected males for Duroc, Landrace, and Yorkshire groups, respectively, was 3.0, 3.0, and 2.7 times greater than the overall percentage affected in their respective breed groups. Heritability of susceptibility to scrotal hernia development was estimated to be 0.29 +/- 0.17, 0.34 +/- 0.23, and 0.34 +/- 0.19 in Duroc, Landrace, and Yorkshire-sired pig groups, respectively.

Serologic analyses and virus isolation studies were carried out to determine the role of bovid herpesvirus-4 (BHV-4) in infections in cattle, principally those of the reproductive tract. Serologic analyses were performed, using an indirect fluorescent antibody test on thoracic fluid specimens from aborted fetuses and on sera from 3 sources of adult cattle. Virus isolation was attempted from field cases of abortion, early embryo death, and postpartum vulvovaginitis/metritis, using uterine discharge and buffy coat preparations obtained from cows and tissues obtained from aborted fetuses. Of 420 fetal thoracic fluid specimens examined, 5 were positive for BHV-4 antibodies. Seventeen percent of adult cattle from 2 sources ie, clinically normal herds and abattoir cattle, were seropositive for BHV-4 antibodies. Cattle from a third source, 4 herds with high incidence of reproductive tract disorders, had a seroprevalence rate between 36 and 88%. Two isolates of BHV-4 were also obtained from this group. the overall incidence of BHV-4 antibodies in clinically normal cattle was higher than previously recognized, with relatively higher prevalence in herds having reproductive problems (chi-squared = 156.5, P less than 0.005). At least 10% of the BHV-4 antibody-positive sera did not have neutralizing antibody against bovine viral diarrhea virus and/or bovid herpesvirus-1, both important causes of bovine reproductive tract disorders.

There were studied the most effective methods of diagnostics of genital forms of clamidiosis at cows in pregnancy, calving and the postnatal period. The description is resulted and the estimation is given methods of laboratory diagnostics, serological methods, a method of light microscopy, immunofluorescence, enzyme immunoassay, chlamydia allocation method on laboratory models, complement linkage reaction, hemagglutination reaction, a method of polymerase chain reaction. As a result of complex use of reaction of complement linkage reaction, on the basis of the analysis of clinical and histologic research, studying of a condition of immune system and nonspecific resistance of an organism it is possible to carry out successfully diagnostics of genital forms of clamidiosis at cows. | Целью исследований явилось изучение наиболее эффективных методов диагностики генитальной формы хламидиоза у коров в период беременности, ca и послеродового периода. Приведено описание и дана оценка методам лабораторной диагностики, серологическим методам, методу световой микроскопии, иммунофлюоресценции, иммуноферментному анализу, методу выделения хламидии на лабораторных моделях, реакции связывания комплемента, реакции гемагглютинации, методу полимеразной цепной реакции. В результате комплексного использования реакции связывания комплемента (РДСК), на основании анализа клинических и гистологических исследований, изучения состояния иммунной системы и неспецифической резистентности организма можно успешно проводить диагностику генитальной формы хламидиоза у коров.
