BACKGROUND: The trachea is the main air passages which is important for taxonomic reasons. The structure of trachea varies considerably in different avian species. OBJECTIVES: This study has been carried out in order to determine the histological and histochemical structure of the trachea in ostriches. METHODS: Sixteen tracheas of 8 female and 8 male nine-month-old healthy blue-necked African ostriches in slaughterhouse of Isfahan were selected. Each trachea divided into cranial, middle and caudal portions and fixed in 10% neutral buffered formalin. Tissues sections were stained with H&E and special stains included Masson’s trichrome, Verhoeff’s, Foot’s, Van Gieson’s, Periodic acid-Schiff, and Alcian blue. RESULTS: The trachea of blue-neck ostriches was composed of tunica mucosa-submucosa, cartilaginous, muscular and serosa. The epithelium was ciliated pseudostratified columnar contained simple alveolar goblet mucous glands. These glands reacted negatively to Periodic acid-Schiff but positively to Alcian blue. The propria-submucosa was composed of dense connective tissue. The muscularis mucosa was absent. Tunica cartilaginous and muscular was made up of sternotrachealis muscle and cartilaginous rings. The rings were only composed of hyaline cartilage. There is no osseous tissue in the tracheal rings. Tunica serosa was composed of loose tissue contained parasympathetic ganglia, adipose tissues, vessels, and all the connective fibers. Three final cartilage rings were shaped tympanum of the syrinx. The histological structure of the trachea showed no significant differences between the male and female ostriches. Except for the decreased number of mucous glands in caudal portion of the trachea, the histological structures of the trachea showed no considerably differences among various portions. CONCLUSIONS: Based on this study, it can be concluded that although the histological and histochemical structure of the trachea in ostrich was similar to those of some other species, but that there were also some differences.

BACKGROUND: Cryptorchidism is a congenital anomaly in which one (unilateral cryptorchidism) or both (bilateral cryptorchidism) testes fail to descend into the scrotum.                  OBJECTIVE: The aim of this study was to describe the anatomical and histological structure of the inguinal testis in the adult donkey. METHODS: In this study, after examination of the 59 donkeys, three of them with unilateral cryptorchidism in inguinal region were identified. These animals were euthanized, and their testicles were removed and evaluated biometrically. Then, the samples were fixated in 10% formalin solution and after sectioning, were stained with Hematoxylin-eosin and PAS, and examined under a light microscope. RESULTS: The results showed that the inguinal testes were stiff, epididymis was not determined and their size and weight were less than scrotal testes. The difference between the weight of cryptorchid and healthy testicles was statistically significant (p<0.05). Seminiferous tubules had lost their natural shape and inner cavity tubes did not have germ cells, and only a limited number of Sertoli cells could be seen. Remaining seminiferous tubules were only visible in the mediastinum. The cortical and subcapsular regions were without tubes and were occupied by loose connective tissues. CONCLUSIONS: Results indicated that the inguinal testes in adult donkeys lost their natural structure and more connective tissues and blood vessels are substituted.

Objective: The objective of this research was to study the histological structures of proventriculus by light and scanning electron microscope and to investigate the localization of glucagon immunoreactive cells within the turkey proventriculus.Materials and methods: Ten adult healthy turkeys were used in this study. The specimens were fixed in 10% buffered neutral formalin. The tissue samples were studied through routine histological and immunohistochemical techniques. Other samples were used for scanning microscope. Results: This study confirmed that the turkey proventriculus was formed from four tunics; tunica mucosa, tunica submucosa, tunica muscularis and tunica serosa. The lamina epithelialis was simple columnar and gave positive reaction in Periodic Acid Schiff (PAS) stain. The lamina propria contained simple tubular glands lined with columnar cells with lymphocyte infiltration. The submucosa contained proventricular glands formed with many round lobules. Each lobule was consisted of secretory tubules radiating from the central lumen of the lobule. The lining epithelium of the central lumen was columnar epithelium. The epithelium of the secretory tubules was cuboidal. Scanning electron microscopic observations showed the mucous membrane was consisted of many macroscopic papillae that formed from many folds which separated by furrows. Immunohistochemical observations revealed that glucagon immunoreactive cells were mainly determined inside the proventricular glands than within the surface epithelium mainly in the basal portion of the lobules and in the epithelium of central lumen of gland lobules. Conclusion: The structure of proventriculus structure in turkey has some variations as compared to other species of birds, and this may be attributed to the diet and its nutritious behavior. OBJECTIVE To determine reference intervals for total nucleated cell count, total protein concentration, pH, RBC count, and percentages of neutrophils, lymphocytes, and large mononuclear cells in synovial fluid samples (SFSs) obtained from the carpal and tarsal joints of healthy swine. ANIMALS 54 healthy commercial finisher pigs that had no evidence of lameness or gross joint swelling. PROCEDURES Each pig was anesthetized, and SFSs were collected from 1 carpal and 1 tarsal joint for fluid analysis, cytologic evaluation, bacterial culture, and PCR analyses for common swine joint pathogens. Each pig was euthanized after SFS collection, and synovial tissue samples were collected for histologic assessment. If necessary, postmortem SFSs were collected. RESULTS Overall, 37 of 50 tarsal and 46 of 53 carpal SFSs met inclusion criteria of sufficient volume, no gross blood contamination, and negative results of bacterial culture and PCR analyses, and were from joints with histologically normal synovial tissues. For the carpal and tarsal joints, upper reference limits were as follows: total nucleated cell count, 3,281 cells/μL and 2,368 cells/μL, respectively; total protein concentration, 3.6 g/dL and 3.6 g/dL, respectively; pH, 7.2 and 7.0, respectively; RBC count, 0.8 × 10(6) cells/μL and 0.1 × 10(6) cells/μL, respectively; and percentage of neutrophils, 46.5% and 33.7%, respectively; percentage of lymphocytes, 40.6% and 56.3%, respectively; and percentage of large mononuclear cells, 92.0% and 95.3%, respectively. CONCLUSIONS AND CLINICAL RELEVANCE Results have provided reference intervals for selected variables in SFSs obtained from the carpal and the tarsal joints of healthy swine, which should be useful in diagnostic investigations of swine lameness and arthritis.

Clinically healthy pigs used in research are assumed to have normal cardiac structure and function. Subclinical cardiac abnormalities may adversely affect the responses being measured in these experiments. The gross and histologic lesions observed in hearts collected from a Canadian abattoir between 2012 and 2015 indicated an unexpectedly high prevalence of cardiac abnormalities: 75% (297/396) of the hearts examined had such lesions. The ratios of total heart weight to body weight and of right ventricle weight to body weight were significantly greater for the hearts with lesions than for the hearts with no lesions, which suggests that cardiac remodeling, particularly hypertrophy, had occurred. The large percentage of hearts with cardiac remodeling from asymptomatic market pigs demonstrates an increased probability that subclinical cardiac abnormalities may exist in research pigs, especially those accessed through commercial channels. Researchers should be aware of this likelihood if subclinical cardiac abnormalities could adversely affect their experimental findings.

As little information is available on the reproductive system of guinea fowl (Numida meleagris), a study was conducted on 49 male guinea fowl to document the histological structure and developmental changes in the luminal diameter of the ducts within the excurrent duct system and associated changes in concentrations of testosterone. Age-related changes were analyzed using the Kruskal-Wallis test and medians separated by the Mann-Whitney U-test. Tubuli recti were clearly visible in the guinea fowl and the rete testes were both intracapsular and extracapsular. Regardless of age, the luminal diameter of the proximal ductuli efferentes was the largest, while that of the connecting duct was the smallest. The luminal diameter of all ducts within the epididymal region increased (P < 0.001) monthly until 20 wk of age, and then increased marginally every month thereafter. Peripheral testosterone concentrations also peaked at 20 wk of age and declined thereafter. In adult birds, the ductus deferens enlarged posteriorly, from an average of about 279 μm cranially to 678 μm caudally. Peripheral testosterone concentrations strongly and positively correlated with the luminal diameter of ducts within the excurrent duct system. The pattern of increase in the luminal diameter of all ducts followed the pattern of testosterone secretion in these birds, which indicates that testosterone concentrations may be closely related to the development of the excurrent duct system in male guinea fowl.

OBJECTIVE To evaluate acute changes of the liver by use of shear wave elastography (SWE) and CT perfusion after radiofrequency ablation (RFA). ANIMALS 7 healthy Beagles. PROCEDURES RFA was performed on the liver (day 0). Stiffness of the ablation lesion, transitional zone, and normal parenchyma were evaluated by use of SWE, and blood flow, blood volume, and arterial liver perfusion of those regions were evaluated by use of CT perfusion on days 0 and 4. All RFA lesions were histologically examined on day 4. RESULTS Examination of the SWE color-coded map distinctly revealed stiffness of the liver tissue, which increased from the normal parenchyma to the transitional zone and then to the ablation zone. For CT perfusion, blood flow, blood volume, and arterial liver perfusion decreased from the transitional zone to the normal parenchyma and then to the ablation zone. Tissue stiffness and CT perfusion variables did not differ significantly between days 0 and 4. Histologic examination revealed central diffuse necrosis and peripheral hyperemia with infiltration of lymphoid cells and macrophages. CONCLUSIONS AND CLINICAL RELEVANCE Coagulation necrosis induced a loss of blood perfusion and caused tissue hardening (stiffness) in the ablation zone. Hyperemic and inflammatory changes of the transitional zone resulted in increased blood perfusion. Acute changes in stiffness and perfusion of liver tissue after RFA could be determined by use of SWE and CT perfusion. These results can be used to predict the clinical efficacy of RFA and to support further studies, including those involving hepatic neoplasia.

OBJECTIVE To describe histologic findings in the small airways during postmortem examination of actively racing horses and to quantify the degree of airway inflammation by use of a semiquantitative scoring system. SAMPLE Lung tissues obtained from 95 horses (Thoroughbreds, Standardbreds, and Quarter Horses) that had been actively racing or training. PROCEDURES 10 standardized lung sections were obtained during postmortem examination of 37 horses that had been actively racing or training, and 2 histologic sections of the caudodorsal lung lobes previously collected from each of 58 horses that died or were euthanized during racing or training were also obtained. Sections were evaluated by use of a validated histologic small airway scoring system. RESULTS Scores for inflammatory cells, smooth muscle, and hemosiderin typically were high. Signalment and cause of death were not significant predictors of lung scores. Lung sample location was a significant predictor, with the highest scores in the caudal and dorsal sections. Inflammatory cell infiltration in peribronchiolar tissues, smooth muscle hyperplasia, and hemosiderin (prevalence of 86%, 98%, and 80%, respectively) were common findings in lungs of these horses, with the caudodorsal regions more severely affected. Correlation was moderate between smooth muscle hyperplasia and inflammatory cell infltration, with minimal correlation between hemosiderin and inflammatory cell infiltration. CONCLUSIONS AND CLINICAL RELEVANCE Inflammatory airway disease has been identified by use of bronchoalveolar lavage in young athletic horses throughout the world. In the study reported here, pathological changes were detected in the wall of small airways of horses that were actively training or racing.

OBJECTIVE To assess differences in skin shrinkage between grossly visible tumor and grossly normal marginal skin of dogs for cutaneous mast cell tumors (MCTs) excised with curative intent and to determine an equation to estimate postexcisional gross tumor margins from preexcisional measurements and vice versa. SAMPLE 19 cytologically confirmed and surgically excised cutaneous MCTs obtained from dogs. PROCEDURES Tumors were measured in craniocaudal and dorsoventral directions before excision, immediately after excision, and after fixation in formalin. Both grossly visible tumor and surrounding grossly normal skin that comprised the surgical margin were measured at each time point. Percentage of shrinkage was compared among time points and between the tumor and surrounding grossly normal skin. Patient and histopathologic variables were correlated to skin shrinkage. RESULTS Overall shrinkage was 17.70%. The amount of shrinkage within the grossly visible tumor (4.45%) was less than that within the surrounding grossly normal skin (24.42%). Most of the shrinkage occurred immediately after excision. There was no effect of age, sex, completeness of excision, or degree of edema. Accuracy of an equation to estimate postexcisional margins from preexcisional measurements was only 18.4%. CONCLUSIONS AND CLINICAL RELEVANCE Grossly evident MCTs of dogs shrunk less than did the grossly normal surrounding skin. Although an equation to estimate postexcisional margins from preexcisional measurements could be derived, it likely would need to contain additional variables not included in the study reported here. Until such an equation exists, care must be used when extrapolating surgical margins from histologic margins and vice versa.

OBJECTIVE To create a bioactive synovium scaffold by infusing decellularized synovial-derived extracellular matrix (synECM) with synovial-derived mesenchymal stem cells (synMSCs). SAMPLE Synovium from the femoropatellar and medial femorotibial joints of equine cadavers. PROCEDURES The synMSCs were cultured in monolayer and not treated or cotransduced to enhance expression of green fluorescent protein (GFP) and human bone morphogenetic protein (BMP)-2. The synECM was decellularized with 0.1% peracetic acid and then seeded with synMSCs (0.5 × 10(6) cells/0.5 mL) by use of a 30% serum gradient. Samples were evaluated on days 0, 3, 7, and 14. Cell migration, differentiation, and distribution into the synECMs were determined by cell surface marker CD90, viability, histologic morphology, and fluorescence microscopy results and expression of GFP, BMP-2, hyaluronan (HA), and proteoglycan (PG). RESULTS At day 14, synMSCs were viable and had multiplied 2.5-fold in the synECMs. The synECMs seeded with synMSCs had a significant decrease in CD90 expression and significant increases in HA and PG expression. The synECMs seeded with synMSCs cotransduced with GFP, or BMP-2 had a significant increase in BMP-2 expression. CONCLUSIONS AND CLINICAL RELEVANCE The synECM seeded with synMSCs or synMSCs cotransduced with GFP, or BMP-2 yielded a bioactive synovial scaffold. Expression of BMP-2 by synMSCs cotransduced to enhance expression of BMP-2 or GFP and an accompanying increase in both HA and PG expression indicated production of anabolic agents and synoviocyte differentiation in the scaffold. Because BMP-2 can promote repair of damaged cartilage, such a bioactive scaffold could be useful for treatment of injured cartilage.
