Comparison of long chain and medium chain polyunsaturated n-3 fatty acids for their ability to correct insulin resistance induced by saturated fatty acids in cultured bovine adipocytes

BACKGROUND: The modern dairy cows, due to intensive selection for higher milk production during past decades, have become more resistant to insulin. Accompanied with DMI depression in per parturient dairy cow, insulin resistance may lead to a massive release of non-esterified fatty acids (NEFA) to bloodstream which might act as a start point for development of other diseases. OBJECTIVES: The purpose of this study was to investigate potentiality of long chain polyunsaturated fatty acid (LCPUFA) of marine or polyunsaturated fatty acids (PUFA) of plant origin to correct the insulin resistance induced by saturated fatty acids in cultured bovine adipocytes. METHODS: Fatty acids from different oils were isolated using KOH and were mixed together with different proportions to obtain the following profiles: 1) saturated fatty acids (SFA); 2); saturated fatty acids+n-6/n-3 PUFA (medium chain) at ratio of 1:1 (SFA-MC1:1)and3) saturated fatty acids + n-6/n-3 PUFA (long chain) at ratio of 1:1 (SFA-LC 1:1). Adipocytes were cultured for 48 in the presence of treatments and after the incubation period an epinephrine challenge was applied to stimulate lipolysis. In the adjacent culture plates for each treatment insulin was added along with epinephrine challenge in different concentrations to test sensitivity of adipocytes to insulin. After conducting insulin sensitivity test, a sample was taken from culture media and analyzed for glycerol concentration as an index of lipolysis. RESULTS: Results indicated that saturated fatty acids effectively induced insulin resistance in adipocytes. Long chain polyunsaturated fatty acids from fish oil had a significant preventive effect on insulin resistance induced by saturated fatty acids so that increasing insulin concentration linearly increased the response of adipocytes to insulin in treatment LC-SFA 1:1. Shorter chain n-3 polyunsaturated fatty acids of plant origin did not show such effects on insulin sensitivity in adipocytes. ConclusionS: n-3 fatty acids from fish oil improved bovine adipocyte sensitivity to insulin but shorter chain n-3 fatty acids did not show this ability.

Two hundred and fifty samples (Marketable milk, Ras cheese, Domiati cheese and ice cream), were randomly obtained from various dairy shops, supermarkets and ice cream shops in Assiut City, Egypt, during the period of November 2022 to March 2023. These samples were examined for the detection and counting of Bacillus cereus. The positive B. cereus isolates were analyzed by PCR to detect 16s rRNA gene for B. cereus, cytotoxin K (cyt K) and emetic gene (ces). Out of the 250 examined samples, 14 (5.6%) were contaminated by B. cereus. The analyzed ice cream had the highest prevalence (11.53%), followed by Ras cheese samples. The prevalence of ces gene recorded in the examined isolates was 57.14%, while cyt K gene was found in all isolates. Moreover, proteolytic and lipolytic activity investigations revealed that nearly all strains released protease enzyme (92.85%), most of which (64.28%) could produce lipase enzyme.

From 50 to 60% of companion animals in the United States are overweight or obese and this obesity rate is rising. As obesity is associated with a number of health problems, an agent that can help weight loss in pets and assist in clinically managing obesity through veterinary prescription foods and medication would be beneficial. Many studies have shown that celastrol, a phytochemical compound found in Celastrus orbiculatus extract (COE), has anti-obesity and anti-inflammatory effects, although these effects have not yet been determined in canine or canine-derived cells. The objective of this study was to investigate the effects of celastrol on the adipogenic differentiation and lipolysis of canine adipocytes. Primary preadipocytes were isolated from the gluteal region of a beagle dog and the primary adipocytes were differentiated into mature adipocytes by adipocyte differentiation media containing isobutylmethylxanthine, dexamethasone, and insulin. In a water-soluble tetrazolium (WST) assay, the cell viability of mature adipocytes was decreased after treatment with COE (0, 0.93, 2.32, and 4.64 nM celastrol) in a concentration-dependent manner, although preadipocytes were not affected. Oil Red O (ORO) staining revealed that COE inhibited the differentiation into mature adipocytes and lipid accumulation in adipocytes. In addition, treatment with COE significantly reduced triglyceride content and increased lipolytic activities by 1.5-fold in canine adipocytes. Overall, it was concluded that COE may enhance anti-obesity activity in canine adipocytes by inhibiting lipid accumulation and increasing lipolytic activity.

The influence of various light regimes on productive indices of broiler chickens cultivation of СОВВ cross taking into consideration the course of some exchange processes was studied in the conditions of the Republic of Belarus. There were formed 4 groups of one-day-old chickens with the approximately identical body weight. For the chickens of a control group there was used the standard for Belarusian poultry farms light regime, i.e. 23 hours of light and 1 hour of darkness (23L:1D), in three experimental groups there were used the following light regimes: first - (3L:1D) х 6, second - (2L:1D) х 8, third - (1L:1D) х 12. The predetermined conditions were set by means of filament lamps with 15 W capacity. Throughout the whole experiment there was realized the identical feeding level for all groups: during the first growing period (1-30 days) chickens were fed with mixed fodder PK 5B containing 22% of crude protein and 296,6 kcal of exchange energy, during second period (31-42 days) - PC 6B containing 20% of crude protein and 332,2 kcal. In the applied mixed fodders there was used enzyme Rovabio. Research results showed that metabolic processes occurring in chicken body in the majority opf cases depended on application of various light regimes. Interchange of light and darkness periods (3 hours of light and 1 hour of darkness) х 6 at light exposure in 15 lk rendered the positive effect on the activity of endocrine systems of chickens of СОВВ crosses. In mentioned above group of chickens the hormones directed their metabolic processes towards increasing the productive indices, namely: daily average gain of chickens of the first group was authentically higher than the same index in the second and the third groups on 13,6% and 17,9%, respectively
