Effects of ivermectin and fenbendazole in strategic treatment of gastrointestinal nematode infections in cattle.

Four groups of 18 beef calves each were used to evaluate effects of different treatments on parasite control and weight gains. The investigation extended from November 1986 (weaning) to October 1987. Group-1 calves were treated with ivermectin (200 micrograms/kg of body weight, SC) at approximately 6-week intervals for a total of 8 treatments; group-2 calves were given the same dosage of ivermectin by the same route of administration as group-1 calves in November, March, and July; group-3 calves were given fenbendazole paste (5 mg/kg, PO) at the same times as group-2 calves; and group-4 calves served as untreated controls with provision for ivermectin salvage treatment. All groups grazed on individual pairs of larval-contaminated, 1.6-ha pastures. Highest (P < 0.05) initial worm counts in fall tracer calves were found in group 3 (Ostertagia ostertagi and Trichostrongylus axei adults) and group 4 (O ostertagi and Haemonchus adults). Fecal egg counts of group-1 calves were low throughout the experiment and pasture larval counts remained negligible after July. Egg counts and larval counts of other groups remained higher into summer. Worm counts, including O ostertagi inhibited early fourth-stage larvae (EL4), were highest (P < 0.05) in groups-3 and -4 spring tracer calves; numbers of O ostertagi EL4 were similarly high in groups 2, 3, and 4; and T axei counts were highest (P < 0.05) in groups-3 and -4 yearlings slaughtered in spring. Liveweights of group-1 calves were greater (P < 0.05) than in other groups from March 2 to October, and by July 2, group-2 calves had a liveweight advantage over group-4 calves. Group-3 calves had the lowest rate of gain from March to July and mean liveweight of the group was less (P < 0.05) than in all other groups from April to October. Only minimal worm numbers were recovered from groups-1 or -2 calves in October. Large numbers of O ostertagi and T axei were recovered from group-4 calves and O ostertagi from group-3 calves. A few calves in groups 3 and 4, but 365 kg in group 1, 328 kg in group 2, 316 kg in group 4, and 281 kg in group 3.

Serum samples obtained from 40,927 swine at various locations in North Carolina between Aug 1, 1987 and July 31, 1988, were tested for antibodies to Trichinella spiralis, using an ELISA based on a larval T spiralis excretory-secretory antigen. In the ELISA, samples were considered to have positive results if the optical density (OD) reading was equal to or 5 times greater than the mean OD value of 4 negative-control sera from trichina-free swine. Of the 40,927 serum samples tested, 154 (0.38%) were positive by ELISA; the rate for breeding swine was 0.35% (105/30,162), and the rate for cull swine was 0.45% (49/10,765). Of the 49 seropositive samples from cull swine, 11 were from out of the state, 22 had no identification, and 16 were known to originate from North Carolina. Seropositivity had a bimodally seasonal distribution, with peaks in March and September. There was no difference between the mean age of seropositive and seronegative swine, but males were at greater risk for seropositivity than were females. Pigs from lots with < 100 sera tested were at increased risk for seropositivity, as were pigs from the central coastal region of North Carolina.

The macrolytic lactone F28249-alpha was titrated in experimentally infected sheep and found to be highly effective against most of the common gastrointestinal nematodes as a single oral dose, given at a rate of 0.025, 0.05, or 0.1 mg/kg. Specifically, maximal activity was evident at even the lowest dosage against adult Haemonchus contortus, Ostertagia circumcinta, Trichostrongylus axei, and T colubriformis and L4 O circumcinta. Activity against Oesophagostomum columbianum was also high at all dosages, with a calculated ED95 of 0.029 mg/kg. Cooperia curticei was eliminated at 0.1 mg/kg, but control was erratic at the lower dosages. The greatest weakness of this compound was its activity against C oncophora. The activity against this parasite was weak (<less than or equal to 85%) at all dosages, and the dosage-response curve was flat, suggesting dosages substantially higher than those given would be necessary for high-order control of this species.

The efficacy, safety, and compatibility of fenbendazole (FBZ) and clorsulon (CLN) were tested after oral administration of label recommended and of higher (5 x) dosage rates to calves naturally infected with gastrointestinal nematodes and Fasciola hepatica. Results for 42 calves allotted to 4 treatment groups indicated a similar efficacy against mature F hepatica by FBZ (5 mg/kg of body weight) and CLN (7 mg/kg) in a combined oral suspension, compared with CLN (7 mg/kg) alone (100 vs 99% reduction). A lesser efficacy was observed against immature flukes (88.6 and 84.9% reduction, respectively). Calves given 25 mg of FBZ/kg and 35 mg of CLN/kg had nearly complete reduction of both mature (99.6%) and immature flukes (99.1%). Fasciola egg counts were reduced by > 99.5% in all treated groups. Against Ostertagia ostertagi, the percentage of efficacy of the combined FBZ (5 mg/kg) and CLN (7 mg/kg) treatment was 94.3% against adults and 81.3% against inhibited larvae. Efficacy against all other nematodes was 100%, except against Cooperia spp adults (98.3%) and immature Oesaphagostomum radiatum (88.0%). At 5 x dosage rates for FBZ and CLN, percentage of removal of adults and inhibited larvae of O ostertagi was 99.3 and 99.0%, respectively, and 99 to 100% for other nematodes. Results indicate that FBZ and CLN are compatible when mixed together and administered as an oral suspension to cattle and that the efficacy is similar to that of the drugs individually. On the basis of further results, we suggest that summer treatment may be superior in preventive value for gastrointestinal nematodes and F hepatica, compared with spring treatment, because of seasonal infection dynamics of the major cattle parasites in Louisiana.

The possible development of type-1 hypersensitivity reactions in the abomasal mucosa caused by soluble L3 products of Ostertagia ostertagi was studied in 4-month-old calves sensitized by repeated exposure to L3 over a 50-day period followed by anthelmintic treatment. Four groups each of 4 calves were used. Group 1 served as nonsensitized controls and group 2 as sensitized controls, group 3 was challenge exposed at 2-week intervals beginning at week 10 with a soluble L3 product (OAG), and group 4 was challenge exposed at 2-week intervals with an oral dose of L3, followed by anthelmintic treatment 3 days later. All calves infected with L3 became sensitized, as indicated by a positive reaction to an intradermal skin test. However, a passive cutaneous anaphylaxis was only partly effective in indicating the presence of homocytotropic antibody in the infected calves. Sensitized calves had significantly (P < 0.05) higher eosinophil counts and plasma pepsinogen values for the entire 14 weeks than uninfected controls. Globule leukocyte and mast cell counts from the abomasal mucosa were also significantly (P < 0.05) higher. Studies for possible immunomodulation revealed that lymphocyte counts decreased between every 2-week challenge-exposure period for groups-3 and -4 calves. A transient depression of blood lymphocyte (BL) responses to phytohemagglutinin (PHA), a T-cell mitogen, was observed over the first 8 weeks in the infected calves. Increases in BL responses to OAG were also observed. Differences were not observed in BL responses to pokeweed mitogen, a T- and B-cell mitogen. Blood lymphocyte responses to PHA in group-3 calves were low following the initial challenge exposure with OAG. The sensitized calf lymphocytes did not have suppressive activity on the response of control calf lymphocytes to PHA. Differences were not observed in lymphocyte responses to PHA in a suppressive assay done on abomasal lymph node lymphocytes. Increases in abomasal lymph node mass and lymphocyte responses to PHA, pokeweed mitogen, and OAG were observed in all sensitized calves. Histologic examination of abomasal lymph node sections from challenge-exposed calves revealed increased mitotic activity in germinal centers. Plasma pepsinogen values in groups 3 and 4 increased between each challenge exposure, which further suggested that type-1 hypersensitivity reactions had occurred in the abomasal mucosa, resulting in increased permeability and leakage of macromolecules.

A study was made to determine the effect of Haemonchus contortus parasitic infection in lambs on the clearance of several IV administered drugs. Clearance of sulfobromophthalein or sulfathiazole from the plasma of lambs was unaffected by infection with H contortus. Clearance of antipyrine was enhanced by the infection, and thiabendazole treatment did not alter this effect. Clearance of chloramphenicol (CAP), administered as the succinate ester (CAPS), was not changed by the infection, but it was increased after treatment with thiabendazole. Changes in the mean body residence time and initial plasma concentration of CAPS and CAP after treatment with thiabendazole indicate that hydrolysis of CAPS to CAP was reduced. High concentrations of CAPS apparently enhanced its own elimination directly rather than via the expected sequence involving hydrolysis, glucuronidation, and excretion of CAP-glucuronide. Enhanced clearance of antipyrine following infection of lambs with H contortus can be explained in at least 2 ways. First, it is possible that thelambs did not have mature amounts of hepatic drug metabolizing enzyme activity as reported by other investigators, which may be explained by breed differences or animal husbandry practices. Second, infection of lambs by H contortus may have triggered an inductive response in hepatic cytochrome P-450-mediated activities, which might result via a generalized enhancement in hepatic protein synthesis associated with the physiologic response to replace plasma proteins and other blood components lost through gastrointestinal hemorrhage caused by the active feeding of adult worms. Other phase-II reactions such as acetylation, glucuronidation, and glutathione-S-transferase apparently were not affected.

Previous work indicated that adult Ancylostoma caninum can be removed from experimentally infected dogs, using a formulation of milbemycin oxime at dosage of 0.5 mg/kg of body weight. To determine the efficacy of this treatment in dogs naturally infected with adult hookworms, 24 mixed-breed dogs with patent hookworm infections were purchased from an out-of-state vendor, and 6 male and 6 female dogs were assigned to either a control group or a group that would be treated. Dogs were treated 10 days after their arrival and were euthanatized 1 week after treatment. Beginning 3 days before treatment, fecal samples were collected daily from all dogs, and the number of Ancylostoma eggs per gram of dry weight of feces was determined from each sample. By 1 week after treatment, the mean number of eggs being passed by the treated dogs had dropped from 12,700 to 10 eggs/g of dried feces; there was no apparent change in fecal egg counts for dogs of the control group. At necropsy, the mean number of adult A caninum in dogs of the treated and control groups was 1.3 and 56, respectively; in these naturally infected dogs, efficacy of treatment was calculated to be 97.8%. The mean number of adult Trichuris vulpis recovered in dogs of the control and treated groups at necropsy was 24 and 0, respectively, which yielded treatment efficacy of 100%. Although Uncinaria stenocephala and Toxocara canis appeared also to be removed by use of this dosage, too few dogs were in the study to calculate meaningful efficacies. The milbemycin oxime formulation appeared to have no effect on the cestodes (Taenia pisiformis and Dipylidium caninum) and spirurids (Physaloptera rara) that were present in some dogs.

Semiselective mesenteric arteriography was performed at regular intervals (inoculation weeks [IW] 0, 18, and 24) in 9 of 10 pony foals raised to be free of parasites. Fifty infective larvae (L3) of Strongylus vulgaris were administered weekly for 4 weeks, then every 2 weeks through the 20th week. Three ponies were given ivermectin (oral paste, 0.2 mg/kg of body weight) treatment at IW 8, 16, and 24. Four ponies were inoculated, but did not receive ivermectin, and a third group of 2 ponies acted as uninoculated controls. Control ponies did not have gross or arteriographic lesions, whereas the inoculated untreated ponies had gross and progressive arteriographic lesions typical of verminous arteritis. Arteriographic lesions in the ivermectin-treated inoculated ponies were not as severe those in the untreated inoculated group, and there was either a partial resolution or a lack of progression of arteriographic lesions in all treated ponies. One untreated inoculated pony did not have progressive arterial lesions as did the 3 others in the group, and may develop resistance to the parasite.

The efficacy of paste and granule formulations of pyrantel pamoate against concurrent infections of Toxocara cati and Ancylostoma tubaeforme in cats was examined in a controlled trial. Three groups of 8 cats received either no medication (controls) or pyrantel pamoate in paste or granule formulations at a dosage of 20 mg/kg of body weight. After administration of the paste formulation, fecal egg counts of A tubaeforme and T cati were decreased by 98.6 and 96.4%, respectively, and 100% of hookworms and 93.5% of ascarids were removed from the intestine. After administration of the granule formulation, fecal egg counts of A tubaeforme and T cati were decreased by 99.4 and 78.2%, respectively, and 100% of adult hookworms and 88.9% of ascarids were removed. All reductions of egg counts and worm numbers were significant (P < 0.01). The clinical safety of pyrantel pamoate was evaluated in 4- to 6-week-old kittens. Three groups of 10 kittens received either no medication (controls) or pyrantel pamoate in paste or granule formulations at the rate of 100 mg/kg for 3 consecutive days. Adverse effects were not observed in young kittens following administration of the high dose of pyrantel pamoate.
