Studies on pathogenicity of Aeromonas species to native breed (Fayoumi) chickens

The pathogenic effect of representative local isolates of Aeromonas organisms was studied on 1٠0 native breed chickens. At 2 weeks of age, one hundred chicks were grouped into four equal groups (1-4); 25 birds each; after collection of fecal from all groups, examined and proved to be free from Aeromonas species. All groups were subcutaneously (s.c) inoculated with 0.5 ml containing 9×105 cfu /bird. Group 1 of chickens was infected by A. hydrophila; while group 2 was infected with A. caviae. The third group was infected with A. sobria; while, chicks of Group 4 were kept as non infected control. Results were showed mild clinical signs in some birds in theform of alternative diarrhea. Post mortem lesions showed general congestion of all carcasses. In severely emaciated cases the lesions were confined to the intestine, which filled with watery fluid and distended with gas. Results of Aeromonas reisolation revealed that all collected weekly fecal swabs were positive. While, percentage of A. hydrophila and A. caviae was 100% out of all testedorgans, while isolation of A. sobria was 100%, 100% and 80% from liver, intestine and heart; respectively. Histopathological examination of infected chicken organs showing focal coaggulative necrosis in liver with mononuclear cells infiltration that varied in severity between groups as less severe in A. hydrophila (Gr.1) than A. caviae (Gr. 2), while A. sobria (Gr. 3) was markedly affected showing severe degenerated and dissociated hepatocytes. Intestinal changes were severe in group 3 than 2 while group 1 showing the mildest comparatively the changes was consist of necrosed mucosa, gland with leucocytic infiltration in lamina propria. Under thecondition of our study we can conclude that the used Aeromonas isolates from field diseased chickens were of mild pathogenicity to s.c. inoculated 12 days old Fayoumi chicks with long course affection.

In this study, we isolated 12 of Brucella (B.) spp. from cattle, which have been positive in Rose Bangal test and tube agglutination test in Gyeongbuk province in 2009. According to AMOS PCR analysis, isolated 12 strains were identified as B. abortus. Murine derived macrophage, RAW 264.7 cells, were infected with isolated 12 strains or reference strain (B. abortus 544), and bacterial internalization were characterized. According to these results, we divided the isolated strains into the following three groups: class Ⅰ, lower internalization than that of B. abortus 544; class Ⅱ, similar internalization to that of that of B. abortus 544; class Ⅲ, higher internalization than that of B. abortus 544 within RAW 264.7 cells. Furthermore, intracellular growth, bacterial adherent assay, LAMP-1 colocalization, virulence in mice and surface protein pattern were characterized. From these results, representative strains of class Ⅲ showed lower LAMP-1 colocalization, higher adherent efficiency, higher virulence in mice than those of B. abortus 544, and showed different pattern of surface proteins. These results suggest that B. abortus field strains, isolated from cattle in Korea, possess various virulence properties and higher internalization ability of field strain may have an important role for its virulence expression.

Objective—To evaluate whether an equine-derived canine H3N8 influenza A virus was capable of infecting and transmitting disease to ponies. Animals—20 influenza virus-seronegative 12- to 24-month-old ponies. Procedures—5 ponies were inoculated via aerosol exposure with 10(7) TCID50 of A/Canine/Wyoming/86033/07 virus (Ca/WY)/pony. A second group of 5 ponies (positive control group) was inoculated via aerosol exposure with a contemporary A/Eq/Colorado/10/07 virus (Eq/CO), and 4 sham-inoculated ponies served as a negative control group. To evaluate the potential for virus transmission, ponies (3/inoculation group) were introduced 2 days after aerosol exposure and housed with Ca/WY- and Eq/CO-inoculated ponies to serve as sentinel animals. Clinical signs, nasal virus shedding, and serologic responses to inoculation were monitored in all ponies for up to 21 days after viral inoculation. Growth and infection characteristics of viruses were examined by use of Madin-Darby canine kidney cells and primary equine and canine respiratory epithelial cells. Results—Ponies inoculated with Ca/WY had mild changes in clinical appearance, compared with results for Eq/CO-inoculated ponies. Additionally, Ca/WY inoculation induced significantly lower numbers for copies of the matrix gene in nasal secretions and lower systemic antibody responses in ponies than did Eq/CO inoculation. The Ca/WY isolate was not transmitted to sentinel ponies. Conclusions and Clinical Relevance—Inoculation of ponies with the canine H3N8 isolate resulted in mild clinical disease, minimal nasal virus shedding, and weak systemic antibody responses, compared with responses after inoculation with the equine H3N8 influenza isolate. These results suggested that Ca/WY has not maintained infectivity for ponies.
