Analysis of β-agonists in different biological matrices by liquid chromatography–tandem mass spectrometry

Wide use is made of β-agonists in therapy due to their smooth muscle–relaxant properties. They also have a side effect of increasing muscle mass. Besides improving oxygen utilisation as bronchodilators, β-agonists increase protein synthesis and promote fat burning. The growth- and performance-enhancing effects are often exploited in illegal use. The guiding objective of this study was to develop a procedure for the determination of β-agonists by a single method in different types of matrices.

A total of two hundreds of slaughtered animals; 50 each of cattle buffaloes, sheep and buffalo calves were collected from slaughterhouse Giza. Each animal was represented by muscular part, prenephric fat, liver, and kidneys. The Four Plate Technique (FPT) is intended to detect antimicrobial residues in collected samples. The liver samples showed high detection incidences (30, 16, 16 and 8%) in cattle, buffaloe, sheep and buffaloe calves respectively as compared to kidney samples (16, 12, 10 and 6% respectively). The lowest incidence was detected in fat samples (0%) followed by muscle samples (4%). High incidence was detected in medium I followed by medium II and III, finally medium IV as well as medium V which failed to be showed any incidence of detection. The tetracycline residues in tissues of slaughtered animals depend on husbandry as well as on withdrawal time after use. The liver (67 + 15, 50 + 14, 54 + 10 and 3 +0.8) and kidney (63 + 16, 46 + 12, 56 + 12 and 8 + 2) samples in cattle, buffaloe, sheep and buffaloe calves respectively showed high residual levels as compared with muscles and fat. The detection of sulfonamide residues in tissues of slaughtered animals may be attributed to misuse of these compounds for long term in animal feed as well as result in accumulation in animal tissues.

An initial census of layer farms in Khartoum State, Sudan, was carried out in late 2007 and early 2008 and found that there were 252 layer farms with a total population of 2 221 800 birds. This paper reports the findings of the census. Based on this information, a structured questionnaire survey of 92 farms was then conducted in the state in April 2008 to collect data on antibiotic usage, demographic data and public health awareness. Ninety-eight per cent of participating farms comprised open-sided houses. It was found that 49% of the farms surveyed were on antibiotic treatment when the survey was conducted, whilst 59% of the farms had used antibiotics within the last 3 months. The study found that farmers and producers had a lack of knowledge about antimicrobial residues, their withdrawal periods and the risk posed by the consumption of these residues. The study also concluded that traditional farming systems in Sudan relied heavily on antimicrobial medication to control disease and almost half of the farms surveyed were treating their flocks with antimicrobials. In addition to this, there was a lack of disease control programmes which probably resulted in a massive use of antibiotics to control endemic diseases. This was further compounded by the absence of governmental supervision and control on the use of drugs.

Fifteen samples of fish fillets were collected from different Egyptian markets to detect the residues of some heavy metals in them. The average weights of fish fillets samples were 250g. The samples were separately packed in polyethylene bag and sent to the laboratory for preparation and analysis. All equipment surfaces and utensils which used in this study were thoroughly cleaned with detergent, rinsed with water, dried and then rinsed with 2-propanol solution. The samples were stored at –2C prior to analysis. The mean levels of lead and cadmium in fish fillets collected from the Egyptian markets were 1.51 0.34 and 0.60 0.0.25 mg/kg; respectively. The concentration of lead in fish muscle were less than the permissible heavy metal limit in fish meat (2.0 mg kg) as stated by the World Health Organization, also these results were in line with the corresponding EU regulations. However the level of cadmium concentration was over than the heavy metal limits in fish meat (0.5 mg/kg) for the World Health Organization.

The risk to consumers of antimicrobial residues in table eggs produced in Khartoum State, Sudan, was studied. All producing layer farms (n = 175) in the state were sampled in April, June and August 2008. A total of 933 eggs from 335 layer houses were screened for antimicrobial residues by using the growth inhibition of Geobacillus stearothermophilus var. calidolactis in-house test. A high proportion of layer farms (72% in April, 61% in June and 66% in August) and layer houses (63% April, 59% in June and 61% in August) were found to have antimicrobial residues, with no significant difference in prevalence (p = 0.57) between study periods. The study showed that the consumer was at constant risk of exposure to antimicrobial residues in table eggs. The paper discusses reasons for the high prevalence of antimicrobial residues in Sudanese eggs and its implications, and makes recommendations to address this important public health problem.

A total of 100 fat samples were collected from slaughtered cattle at Beni-Suef slaughter house. Animals were randomly selected from the slaughter line. The age of the slaughtered cattle was normally less than three years (18-30 months). Samples were separately packed in plastic bags, identified and stored at -18°C until analysis. The analysis was conducted at Max Rubbner Institute, Kulmbach, Germany for PCDDs, PCDFs and dl-PCBs. The average concentration sum for PCDD calculated as upper bound pg WHO-TEQ/g was 1.71 pg WHO-TEQ/g fat. While theaverage concentration sum for PCDFs was 4.93 pg WHO-TEQ/g fat. The average concentration sum of dl PCBs including non-ortho and mono-ortho PCBs was 5.091 pg WHO-TEQ/g fat. The total TEQ value for PCDD/Fs and dl-PCBs was 11.74 pg WHO-TEQ/g fat. The residual levels of PCDD/Fs and dl-PCBs in the examined beef fat samples were higher than the proposed maximum permitted levels set by the European Commission (2006).

Ninety chicks were experimentally, orally received different doses of estradiol-17β and diethylstilbestrol with ration. Samples were collected from tissues (wings, breast muscles, thigh muscles, skin and fat) and giblets (liver and gizzard) for detection of hormonal residues after 4 and 21 days from the last dose, which proved the presence of such residues in all samples. Effect of temperatures (boiling, roasting and freezing) on hormonal residues of positive samples was evaluated. It was proved that There is no significant variations in reduction of hormonal residues in each of breast and thigh muscles of chickens at (p < 0.05) after boiling, roasting and freezing at - 20˚ C as well as a significant differences was detected in skin and fat samples at (p < 0.05) after boiling and roasting. Public health importance of hormonal residues was discussed.

A total of one hundred samples of marketed fish farm; 25each of tilapia (Oreochromis niloticus), Mugil cephalus, Carp (Silver) and fry; were collected from different localities at Al- Behera and Kafr El-Shakh Provinces. The samples were examined for detection of testosterone hormone by Thin Layer chromatography (TLC) as well as it was quantitatively assayed by Radio-Immuno-Assay (RIA). The obtained result revealed that the testosterone residues were detected in flesh of farm fish of Tilapia (Oreochromis nilotocus) and Carp (Silver), each constituting 24% and 4 %; respectively but it could be failed to detect in each of Mugil cephalus and fry. The hormonal residues of testosterone was ranged from 3.25 to 34.9 ng/g with mean value 4.22±1.1 ng/g in positive samples of Tilapia, while only one sample of Carp showed 22.0 ng/g, Mugil cephalus and Fry showed no detectable level of hormonal residues. The public health significance of detectable levels was discussed as well as the recommendations to avoid health hazards from such fish were mentioned.

The goal of this work was the validation of a method for detection of residues of multiple classes of pesticide and determination of their residues in chicken breast fillets.
